<P>Mice lacking the IL-1R-associated kinase 4 (IRAK4) are completely resistant to LPS-induced endotoxic disorder or the TLR9 agonist CpG DNA plus D-galactosamine-induced acute liver injury (ALI), whereas wild-type strains succumb. However, translational drugs against sepsis or ALI remain elusive. Lonicerae flos extract is undergoing the clinical trial phase I in LPS-injected healthy human volunteers for sepsis treatment. In the current study, chlorogenic acid (CGA), a major anti-inflammatory constituent of lonicerae flos extract, rescued endotoxic mortality of LPS-intoxicated C57BL/6 mice, as well as ameliorated ALI of LPS/D-galactosamine-challenged C57BL/6 mice. As a mechanism, CGA inhibited various TLR agonist-, IL-1 alpha-, or high-mobility group box-1-stimulated autophosphorylation (activation) of IRAK4 in peritoneal macrophages from C57BL/6 or C3H/HeJ mice via directly affecting the kinase activity of IRAK4, a proximal signal transducer in the MyD88-mediated innate immunity that enhances transcriptional activity of NF-kappa B or AP-1. CGA consequently attenuated protein or mRNA levels of NF-kappa B/AP-1 target genes encoding TNF-alpha, IL-1 alpha, IL-6, and high-mobility group box-1 in vivo under endotoxemia or ALI. Finally, this study suggests IRAK4 as a molecular target of CGA in the treatment of innate immunity-related shock and organ dysfunction following insult of various TLR pathogens from bacteria and viruses.</P>

Aims: The number of the patients with acute hepatitis A is increasing due to improved hygienic environment and low seroprevalence of anti-HAV IgG in Korea. There were two peak periods of acute hepatitis A about 10 years ago and last year. This study aims to analyze the changes of the clinical features of acute hepatitis A patients between two peak periods in Korea. Methods: Patients with acute hepatitis A who had been hospitalized at Dankook University Hospital from 1998 to 2019 were included. We reviewed patient’s medical records especially, two peak periods (2008-2009 vs. 2019). Results: From 1998 to 2019, total 798 patients had admitted with acute hepatitis A. The mean age of the patients was 33.0 ± 10.1 years, and male patients (61.4%) were more. The mean peak AST and ALT were 2407.1 ± 2892.2 IU/L and 2547.2 ± 1896.3 IU/L. Total 8 (1%) patients were not recovered. Among 8 patients, 6 patients took liver transplantation and 2 patients expired. There were two peak periods for 22 years. First peak period was 2008-2009 year (219 patients) and second peak period was 2019 year (106 patients). The mean age increased from 31.7 ± 8.8 to 40.1 ± 9.4 years The proportion of male patients decreased from 69.4% to 58.5%. The symptom duration days decreased from 7.6 ± 8.0 days to 6.0 ± 3.9 days and total admission days also decreased from 7.4 ±3.4 days to 5.9 ± 3.2 days. The peak INR increased from 1.3 ± 0.5 to 1.4 ± 0.6. The death or transplantation rate increased from 0.9% to 2.8%. Conclusions: The mean age of patients with acute hepatitis A increased about 9 years over two peak periods. The death or transplantation rate increased very much although the admission days decreased.
Sensor Alignment Calibration for Precision Attitude Determination of Spacecrafts
Il-Hyoung Lee,Chang-Kyung Ryoo,Hyochoong Bang,Min-Jea Tahk,Sang-Ryool Lee 한국항공우주학회 2004 International Journal of Aeronautical and Space Sc Vol.5 No.1
A new alignment calibration method of attitude sensors for the precision attitude determination of a spacecraft based on the extended Kalman filter is proposed. The proposed method is divided into two steps connected in series: the gyro and the star tracker calibration. For gyro calibration, alignment errors and scale factor errors are estimated during the calibration maneuver under the assumption of a perfect star tracker. Estimation of the alignment errors of the star trackers and compensation of the gyro calibration errors are then performed using the measurements including payload information. Performance of the proposed method are demonstrated by numerical simulations.

In this study, we prepared amphotericin B (AmpB)-encapsulated polymeric micelle of poly(DL-lactide-co-glycolide) (PLGA) grafted-dextran (DexLG) copolymer for the cytotoxicity test. The average particle size of AmpB-encapsulated DexLG polymeric micelles was around $30{\sim}70\;nm$ and their morphology showed spherical shapes. Since aggregation states of AmpB are related to intrinsic cytotoxicity, prevention of AmpB aggregation in aqueous solution will provide low cytotoxicity and increased antimicrobial activity for the infectious disease. At UV/VIS spectrum measurement, polymeric micelle prepared from methanol/water mixture (method B) showed a monomeric state of AmpB while polymeric micelle prepared from DMSO (method A) showed an aggregated state. During the hemolysis activity test, polymeric micelle from method B showed reduced hemolysis activity compared to AmpB itself and polymeric micelle from method A. These results indicated that AmpB-incorporated polymeric micelle prepared from methanol/water mixture has low cytotoxicity and favorable antimicrobial activity.

<P>Infection with dengue virus (DENV) is a serious health issue that causes severe dengue fever and occasionally lethal complications, such as dengue hemorrhagic fever. Therefore, rapid and sensitive DENV detection is important to reduce morbidity and mortality. Here, we screened dengue NS1-specific affibodies and developed highly stable anti-NS1 affibody-functionalized AuNPs to improve the detection sensitivity of the NS1 antigen in ELISA. First, we screened NS1 antigen-specific affibody molecules (ZNS112, ZNS116, and ZNS146) from an affibody phage library. The affibodies were then expressed and purified from <I>Escherichia coli.</I> Among them, the ZNS112 affibody showed the highest equilibrium binding constant (<I>K</I>d) of 1 μM and was sufficiently stable at high temperatures. This affibody was functionalized on AuNPs measuring 20 nm in diameter, which were used as carriers to achieve amplification of the signal in ELISA. The developed anti-NS1 affibody-functionalized AuNPs ((ZNS112)2-AuNP) showed good properties, such as easy synthesis, a high number of affibody conjugations on the AuNPs, and excellent stability under harsh conditions with high salt concentrations and temperature. In addition, this nanoparticle-based enhanced ELISA resulted in a 14.2-fold signal amplification performance for dengue NS1 detection in comparison with the affibody-based ELISA. This novel and sensitive method using (ZNS112)2-AuNPs may have applications in the detection of DENV in infected patients at an early stage and for the detection of other pathogens in clinical diagnostics.</P>

<P>Content delivery service has become a major traffic load on today's Internet, and this has triggered the interest of ISPs in operating their own content delivery networks (CDNs) to optimize Internet traffic considering both content delivery caching and user-network proximity. ISPs, however, are typically regionally bound or network-domain-wise isolated; hence, their CDN gain is somewhat limited. In order to enhance the gain of ISP CDN services to the level of incumbent global CDNs, a CDN interconnection (CDNI) model is introduced by IETF, where local ISP CDN services can be extended among heterogeneous CDNs across network domains. However, despite the multiple benefits of a CDNI system, it is difficult to apply a CDNI service to the current CDN market due to the platform independence. Hence, we introduced a CDNI gateway model that is standard-capable and platform-independent. With the CDNI gateway model, we design and implement a complete CDNI system and conduct a CDNI service trial with three major ISPs in South Korea. To the best of our knowledge, this is the first CDNI service trial complying with the IETF standard achieved by a multi-ISP collaboration. According to the analysis of experimental results from the service trial, we observe that CDNI can reduce content traffic by about 40 percent at the Internet exchange link compared to a legacy CDN system.</P>
