        Silencing of JHEH and EcR genes of Plutella xylostella (Lepidoptera: Plutellidae) through double stranded RNA oral delivery

        Plutella xylostella (L.), Diamondback moth (DBM), is a notorious pest disrupting cruciferous crops across the world. Overuse of chemically synthesized insecticides as well as application of bio-pesticide like Bacillus thuringiensis has gained progressive resistance against them, resulting in DBM management failures. To overcome this, a more potential, non-chemical and safe technology like Ribonucleic acid interference (RNAi) is to be implemented. RNAi, is a reverse genetic tool with high sequence specificity, elicited by double-stranded RNA (dsRNA) for silencing the target genes. The objective of this study was to silence two candidate genes fromDBMviz., Juvenile Hormone Epoxide hydrolase (PxJHEH) and Ecdysteroid receptor (PxEcR) involved in regulating metamorphosis and molting respectively. In this regard, we have custom designed the dsRNAs (500 bp) by cloning and sequencing the candidate genes and delivered it orally (non-invasive mode). Further, we have assessed the extent of down regulation of target genes with five concentrations of dsRNA (1, 2, 5, 10 and 20 μg/μl) at four time points (24, 48, 72 and 96 h). The extent of gene silencing and mortality recorded for both genes were proportional to the dsRNA concentration. Our study projects that PxJHEH and PxEcR genes would be good target for dsRNA mediated gene silencing in management of insect pests such as DBM in near future.

        Cytokine expression pattern in milk somatic cells of subclinical mastitis-affected cattle analyzed by real time PCR

        The expression profiles of inflammatory cytokines viz. interleukins (IL)-6, IL-8, IL-12, granulocyte macrophage-colony stimulating factor, interferon-${\gamma}$ and tumor necrosis factor-${\alpha}$ in response to subclinical mastitis in indigenous cattle breed Kankrej (n = 6), Gir (Bos indicus) (n = 12) and crossbred (Bos taurus${\times}$Bos indicus) (n = 7) were investigated using quantitative real time PCR. Significant correlation (p < 0.05) was observed between total bacterial load and somatic cell count (SCC) in all three breeds of cattle. All the cytokines were observed to be up-regulated compared to cows with healthy quarters, however, level of their expression varied among three breeds of cattle. In Kankrej most cytokines were found to be transcribed to higher levels than in other two breeds; the milk had higher load of bacteria but not so high SCC, implying that Kankrej has a higher inherent resistance against mastitis. The results of present study indicated that mammary glands of crossbred cattle are more sensitive to bacterial infection than indigenous breed of cattle as they elicit immune response at lower bacterial load and result into higher SCC. Research on identification of factors responsible for differentially expressed cytokines profiles and use of cytokines as immunomodulatory tools can pave way for formulating control strategies against bovine mastitis.

        At Present times, A common person focus on their health issues not only for themselves but also for their family because health is a wealth. In the modern days most of the people are working and they spending 40% to 50% of the income on their health issues not only for themselves but also for their family. By this the rate of population arrival to the Hospitals are increasing day-by-day. So the hospitals have to be reliable towards the patients. The records of the patients and the medication details are to be maintained correctly. There may be many risks in the software that they use. So any health organizations are struggles to display the patient electronically measured health records and what they use medication outside. So every time common people are lose their health information. If the common people know the about their previous health records they reduce their money and take the first aid immediately. In this paper we suggest a new approach to risk assessment for health care systems. As risk assessment is a Qualitative approach, there are no exact techniques to solve such risks. Lack of risk assessment techniques leads to failure in the system and organization as the people don’t find interest to come if once the organization fails in any of the issues. Our new method mainly focuses on the risk management for health care organizations. Today’s health care industry faces a number of emerging risk issues related to health care reform, the shift from fee-for-services to outcomes-based compensation, the use of electronic medical records and the management of human capital. Therefore our method has put forth a Risk management technique for the Risk assessment.
