        Dental Panoramic Radiographic Indices as a Predictor of Osteoporosis in Postmenopausal Saudi Women

        Background: Many oral presentations of osteoporosis-a bone metabolic disease-were recorded. Thus, we aimed to assess panoramic radiomorphometric indices with bone mineral density (BMD) values among Saudi postmenopausal women and its importance in the prediction of osteoporosis. Methods: A total of 431 Saudi women were enrolled in this study. Panoramic radiographs were obtained at the time of BMD measurement. Subjects were fatherly classified into; normal BMD, osteopenia, and osteoporosis groups. Serum follicle-stimulating hormone, luteinizing hormone (LH), estradiol (E2), 25-hydroxy-vitamin D (25[OH]D) and intact-parathyroid hormone were measured. Moreover, serum creatinine, calcium, and phosphate, together with serum osteocalcin (s-OC), procollagen type I N-terminal propeptide (s-PINP) and cross-linked C-terminal telopeptide of type 1 collagen (s-CTX) were measured. Receiver-operator curve (ROC) curve analysis for use of mandibular cortical width (MCW), panoramic mandibular index (PMI), and maxillary-mandibular ratio (M/M ratio) to differentiate women with osteoporosis or osteopenia from normal subjects was calculated. Cut off values of 4.6 at T score <-1 and 4.1 at T score ≤-2.5 were used. Results: Body mass index is significantly low in the osteoporotic group. There is no significant difference in serum levels of LH, E2, calcium, phosphate, and 25(OH)D between the studied groups. Moreover, s-OC, C-terminal propeptide of procollagen type I, s-PINP, s-CTX, and urinary-CTX are significantly higher in osteoporosis than normal and osteopenia groups. ROC curve analysis revealed that MCW and PMI showed significant data while M/M ratio is non-significant. Conclusions: It could be concluded that MCW as an important panoramic radiographic parameter can be used for prediction and diagnosis of osteoporosis in postmenopausal Saudi women with low BMD.

      • <i>Commiphora molmol</i> Modulates Glutamate-Nitric Oxide-cGMP and Nrf2/ARE/HO-1 Pathways and Attenuates Oxidative Stress and Hematological Alterations in Hyperammonemic Rats

        <P>Hyperammonemia is a serious complication of liver disease and may lead to encephalopathy and death. This study investigated the effects of <I>Commiphora molmol</I> resin on oxidative stress, inflammation, and hematological alterations in ammonium chloride- (NH<SUB>4</SUB>Cl-) induced hyperammonemic rats, with an emphasis on the glutamate-NO-cGMP and Nrf2/ARE/HO-1 signaling pathways. Rats received NH<SUB>4</SUB>Cl and <I>C. molmol</I> for 8 weeks. NH<SUB>4</SUB>Cl-induced rats showed significant increase in blood ammonia, liver function markers, and tumor necrosis factor-alpha (TNF-<I>α</I>). Concurrent supplementation of <I>C. molmol</I> significantly decreased circulating ammonia, liver function markers, and TNF-<I>α</I> in hyperammonemic rats. <I>C. molmol</I> suppressed lipid peroxidation and nitric oxide and enhanced the antioxidant defenses in the liver, kidney, and cerebrum of hyperammonemic rats. <I>C. molmol</I> significantly upregulated Nrf2 and HO-1 and decreased glutamine and nitric oxide synthase, soluble guanylate cyclase, and Na<SUP>+</SUP>/K<SUP>+</SUP>-ATPase expression in the cerebrum of NH<SUB>4</SUB>Cl-induced hyperammonemic rats. Hyperammonemia was also associated with hematological and coagulation system alterations. These alterations were reversed by <I>C. molmol</I>. Our findings demonstrated that <I>C. molmol</I> attenuates ammonia-induced liver injury, oxidative stress, inflammation, and hematological alterations. This study points to the modulatory effect of <I>C. molmol</I> on glutamate-NO-cGMP and Nrf2/ARE/HO-1 pathways in hyperammonemia. Therefore, <I>C. molmol</I> might be a promising protective agent against hyperammonemia.</P>

        Nω-Nitro-L-Arginine Methylester Ameliorates Myocardial Toxicity Induced by Doxorubicin

        The effects of No-nitro-L-arginine methylester (L-NAME) and L-arginine on cardiotoxicity that is induced by doxorubicin (Dox) were investigated. A single dose of Dox 15mgkg i.p. induced cardiotoxicity, manifested biochemically by a significant elevation of serum creatine phosphokinase (CPK) activity [EC 2.7,3.2]. Moreover, cardiotoxicity was further confirmed by a significant increase in lipid peroxides, measured as malon-di-aldehyde (MDA) in cardiac tissue homogenates. The administration of L-NAME 4 mg/kg/d p.o. in drinking water 5 days before and 3 days after the Dox injection significantly ameliorated the cardiotoxic effects of Dox, judged by the improvement in both serum CPK activity and lipid peroxides in the cardiac tissue homogenates. On the other hand, the administration of L-arginine 70mg/kg/d p.o. did not protect the cardiac tissues against the toxicity that was induced by the Dox treatment. The findings of this study suggest that L-NAME can attenuate the cardiac dysfunction that is produced by the Dox treatment via the mechanism(s), which may involve the inhibition of the nitric oxide (NO) formation. L-NAME may, therefore, be a beneficial remedy for cardiotoxicity that is induced by Dox and can then be used to improve the therapeutic index of Dox.

        Growth, carcass traits, immunity and oxidative status of broilers exposed to continuous or intermittent lighting programs

        Objective: An experiment was conducted to investigate the continuous and intermittent lighting program effects on terms of the productive performance, carcass traits, blood biochemical parameters, innate immune and oxidative status in broiler chicks. Methods: A total of 600 Cobb-500 one day old chicks were randomly allocated into six equal groups (100 chicks per treated group with five replicates of 20 chicks each) based on lighting program; 22 continuous lighting (22 C), 11 h lighting+1 darkness twice daily (11 L/1 D), 20 h continuous lighting (20 C), 5 h lighting+1 darkness four times daily (5 L/1 D), 18 h continuous lighting (18 C) and the final group subjected for 3 h lighting+1 h darkness six times daily (3 L/1 D). The experimental period lasted 42 days. Results: Compared with those under the intermittent light program, broiler chicks exposed to continuous lighting for 22 h had significant improvement in live body weight and carcass (dressing and breast percentage) measured traits. Though reducing lighting hours significantly reduced feed intake and feed conversion ratio values. Different lighting programs revealed no significant effect on all blood biochemical parameters. Oxidative stress and innate immunity parameters significantly enhance by reducing lighting hours (3L/1D). Conclusion: The findings suggest that reducing lighting hours up to 3L/1D would be more useful in enhancing feed efficiency, innate immunity, and oxidative status compared with continuous lighting programs on broilers.

        Genetic parameters and principal components analysis of breeding value for birth and weaning weight in Egyptian buffalo

        Objective: The objectives of the current study were to study the main environmental factors affecting birth weight (BW) and weaning weight (WW), estimate variance components, genetic parameters and genetic trend and to evaluate the variability and relationships among breeding value of BW and WW using principal components analysis (PCA). Methods: A total of 16,370 records were collected from 8,271 buffalo calves. Genetic parameters and breeding values were estimated using a bivariate animal model which includes direct, maternal and permanent maternal effects. These estimates were standardized and used in PCA. Results: The direct heritability estimates were 0.06 and 0.41 for BW and WW, respectively whereas direct maternal heritability values were 0.03 and 0.14, respectively. Proportions of variance due to permanent environmental effects of dam were 0.455 and 0.280 for BW and WW respectively. The genetic correlation between BW and WWs was weak approaching zero, but the maternal correlation was 0.26. The first two principal components (PC1 and PC2) were estimated utilizing the standardized breeding values according to Kaiser method. The total variance explained by the first two PCs was 71.17% in which 45.91% and 25.25% were explained by PC1 and PC2, respectively. The direct breeding values of BW were related to PC2 but those of WW and maternal breeding values of BW and WWs were associated with PC1. Conclusion: The results of genetic parameters and PCA indicate that BW and WWs were not genetically correlated and improving growth traits of Egyptian buffaloes could be achieved using WW without any adverse effect by BW.

      • Biological Screening of Novel Derivatives of Valproic Acid for Anticancer and Antiangiogenic Properties

        Background: Valproic acid (VPA) is a potent anticancer and antiangiogenic agent. However, design and synthesis of chemical derivatives with improved antiangiogenic and anticancer activities are still necessary. In this study a library of novel derivatives of VPA was synthesized and tested. Methods: A human liver cancer cell line (HepG2) and a human normal embryonic kidney cell line (HEK 293) were exposed to various concentrations of VPA derivatives for 24 hours and cell viability was checked by MTT colorimetric assay. Anti-angiogenic properties were evaluated in transgenic zebrafish embryos. Results: N-valproylglycine derivatives suppressed survival almost 70% (p value 0.001) in HepG2 cells but only 10-12% in HEK 293 cells (p value 0.133). They also suppressed angiogenic blood vessel formation by 80% when used between $2-20{\mu}M$ in zebrafish embryos. Valproic acid hydrazides showed moderate level of anticancer activity by affecting 30-50% (p value 0.001) of cell viability in HepG2 cells and 8-10% in HEK293 cells (p value 0.034). Conclusion: The majority of compounds in this study showed potent and stronger antiangiogenic and anticancer activity than VPA. They proved selectively toxic to cancer cells and safer for normal cells. Moreover, these compounds inhibited developmental angiogenesis in zebrafish embryos. Based on the fact that liver is a highly vascularized organ, in case of liver carcinoma these compounds have the potential to target the pathological angiogenesis and could be an effective strategy to treat hepatocellular carcinoma.

        Low cost nanoparticles derived from nitrogen fertilizer industry waste for the remediation of copper contaminated soil and water

        The goal of this study was to produce a novel nano-scale material from nitrogen fertilizer industry byproduct (nNFIB) and assess its capability to remediate Cu contaminated wastewater and soil. The novel nNFIB was produced using planetary mono mill and characterized. Equilibrium and kinetics studies of Cu sorption by nNFIB were performed in batch system. The effects of a variety of factors, including pH, coexisting ions and adsorption time on Cu adsorption were investigated. Furthermore, Cu sequestration mechanism onto nNFIB was investigated using sequential extraction technique and Fourier transform infrared (FTIR) spectra before and after nNFIB adsorption. The Cu sorption equilibrium and kinetics data were successfully described by Langmuir and first-order models, respectively. The calculated maximum Cu(II) adsorption capacity (qmax) of nNFIB (100 mg g−1) was four times higher than qmax of bulk NFIB. Copper removal by nNFIB was quite fast (around 86%) in the first 5 min and gradually slowed down until achieved 100% removal at equilibrium time. The FTIR spectra and Cu fractionation data in biosolid-amended soil demonstrated that Cu sequestration in contaminated water and soil is strongly related to CaCO₃ of nNFIB. The overall findings show the potential use of nNFIB as a best management practice for Cu removal from wastewater and Cu stabilization in contaminated biosolid-amended soils.

        Egyptian Honeybee Pollen as Antimicrobial, Antioxidant Agents, and Dietary Food Supplements

        In this study, three Egyptian bee pollen types from different plant sources, namely maize (Zea mays),clover (Trifolium alexandrinum), and date palm (Phoenix dactylifera), were evaluated as natural antimicrobial agents,antioxidants, and food additives. A methanolic extract of maize bee pollen exhibited the highest antibacterial activity,followed by clover and date palm bee pollen. Staphylococcus aureus and Escherichia coli were the most sensitive,followed by Listeria monocytogenes, Salmonella enteritidis,and Pseudomonas aeruginosa. In ghee, all methanolic extracts, especially that of maize bee pollen, showed high antioxidant activities as radical scavenger substances and as inhibitors of lipid peroxidation. Sensory evaluation of yoghurt supplemented with different bee pollens revealed that maize bee pollen improve texture, increased gel strength, and decreased syneresis and had favorable nutty flavor, while clover and date palm pollen added a sweet taste and bean-like flavor, respectively. These findings establish the potential of Egyptian maize and clover bee pollen as antioxidants, antimicrobial agents, promising natural food supplements, and natural preservatives.

        Antihyperglycemic Effect of Crude Extracts of Some Egyptian Plants and Algae

        Diabetes mellitus is a major global health problem. Various plant extracts have proven antidiabetic activity and are considered as promising substitution for antidiabetic drugs. The antihyperglycemic effect of 16 plants and 4 algae, commonly used in Egypt for the treatment of diabetes mellitus, was investigated. A diabetes model was induced by intraperitoneal injection of nicotinamide (120 mg/kg body weight [b.wt.]), then streptozotocin (200 mg/kg b.wt.) after 15 min. Hydroethanolic extracts (80%) of the plants and algae under investigation were prepared. The extracts were orally administered to nicotinamide-streptozotocin–induced diabetic mice by a gastric tube at doses 10 or 50 mg/kg b.wt. for 1 week. The antidiabetic activity was assessed by detection of serum glucose concentrations at the fasting state and after 2 h of oral glucose loading (4.2 mg/kg b.wt.). Extracts prepared from Cassia acutifolia, Fraxinus ornus, Salix aegyptiaca, Cichorium intybus, and Eucalyptus globulus showed the highest antihyperglycemic activity among the tested plants. Extracts prepared from Sonchus oleraceus, Bougainvillea spectabilis (leaves), Plantago psyllium (seeds), Morus nigra (leaves), and Serena repens (fruits) were found to have antihyperglycemic potentials. Extracts prepared from Caulerpa lentillifera and Spirulina versicolor showed the most potent antihyperglycemic activity among the tested algae. However, some of the tested plants have insulinotropic effects, all assessed algae have not. Identification of lead compounds from these plants and algae for novel antidiabetic drug development is recommended.
