Iron as well as aluminum is reported to accumulate in neurons with neurofibrillary tangles (NFTs) of Alzheimer's disease(AD) brain. Previously we demonstrated that aluminum(Ⅲ) shows phosphate-dependent binding with hyperphosphorylated τ(PHFτ), the major constituent of NFTs, thereby inducing aggregation of PHFτ. Herein we report that iron(Ⅲ) can also induce aggregation of soluble PHFτ to occur, iron in the oxidized state (Ⅲ) is essential since iron in the reduced state (Ⅲ) lacks such ability. Furthermore, iron (Ⅲ)-induced aggregation is reversed by reducing iron (Ⅲ) to iron (Ⅱ). Thus the iron-participating aggregation is mediated not only by τ phosphorylation but also by the transition of iron between reduced (Ⅱ) and oxidized (Ⅲ) states. Further incubation of insoluble PHFτ aggregates isolated from AD brain with reducing agents produced liberation of solubilized PHFτ and iron (Ⅱ), indicating that PHFτ in association with iron (Ⅲ) constitutes the insoluble pool of PHFτ. These results indicate that iron might play a role in the aggregation of PHFτ leading to the formation of NFTs in AD brain.
Regulation of Wnt signaling by protein-protein interaction and post-translational modifications

The Wnt signaling pathway is conserved in various species from worms to mammals, and plays im-portant roles in cellular proliferation, differentiation, and migration. Wnt stabilizes cytoplasmic β-catenin and then the accumulated β-catenin is translocated into the nucleus, where it activates the transcriptional factor T-cell factor (Tcf)/lymphoid enhancer factor (Lef), and thereby stimulates the expression of genes including c-myc, c-jun, fra-1, and cyclin D1. Tight regulation of this response involves post-trans-lational modifications of the components of the Wnt signaling pathway. Phosphorylation, ubiquitination, and sumoylation have been shown to affect the half-life of β-catenin and the transcriptional activity of Tcf/Lef. The precise spatio-temporal patterns of these multiple modifications determine the driving force of various cellular responses.
Peptides from Bombyx Fibroin Counter D-Galactose-induced Hair Aging in Mice

Using proteases, we produced a peptide mixture from fibroin of the silkworm $Bombyx$ $mori$. Mice received D-galactose by subcutaneous injection for 8 weeks to accelerate aging, and then received the peptide mixture orally for 7 weeks. In the mice aged with D-galactose, the coefficient of friction of hair increased significantly up to 1.6-fold, but in the mice subsequently given the peptide mixture, it was normal. Scanning electron microscopy showed improved hair cuticles in the latter mice too. These results indicate that oral administration with peptides from $Bombyx$ fibroin counters the aging of hair cuticles.
Jumping Pattern Generation for One-Legged Jumping Robot

This paper describes a one-legged robot capable of jumping that consists of a waist, thigh, a shin, and foot. The jumping robot’s height is 1200 [mm], and its weight is 90 [kg]. Two kinds of pneumatic artificial muscle, the mono-articular muscle and the bi-articular muscle are installed. Also, the jumping pattern that is composed of an air-supplying and an air-exhausting pattern of the pneumatic actuator is discussed. The jumping pattern is generated offline taking into account actuator dynamics. A jumping experiment was conducted, and the effectiveness of the mechanism and jumping pattern was verified.

<P>Circadian rhythms are the daily oscillations of multiple biological processes regulated by an endogenous clock. The Period2 gene is essential in controlling the circadian rhythm and plays an important role in tumor suppression. We examined whether the overexpression of the mouse Period2 gene (mPer2) in cultured tumor cells from human tissues inhibits cell growth, using the recombinant adenovirus vector AdmPer2. The overexpression of mPer2 in human pancreatic cancer cells (Panc1, Aspc1) reduced cellular proliferation and induced apoptotic cell death. Infection with AdmPer2 also inhibited cell-cycle progression, inducing arrest at the G(2)-M phase. Western blotting analyses confirmed that infection with AdmPer2 reduced Bcl-X(L), Cdc2 and cyclin B1 protein, whereas it increased Bax protein in Aspc1 cells. The overexpression of mPer2 suppressed Cdc2 kinase activity. Moreover, infection with AdmPer2 resulted in dose-dependent synergic cell killing effects with the anticancer agent cisplatin (CDDP) in human pancreatic cancer cells. This synergic effect might be related to the reduction of Bcl-X(L) induced by infection with AdmPer2. Our results suggest that the circadian gene Period2 may play an important role in suppression of cell proliferation in human cancer, and additionally Period2 gene expression level may influence the sensitivity to cisplatin depending on Bcl-X(L) expression level.</P>
