      • Impaired nucleolus biogenesis during pre-implantation development of mouse nuclear transfer embryos

        핵인의 가장 널리 알려진 기능 중에 하나는 리보솜의 합성을 수행한다는 것이다. 접합 유전자의 수정 이후에 핵인의 재구축은 포유 동물의 착상 전 배아의 성공적인 발달을 위해 필수 적인 요소이다. 그러나 핵 이식 배아에서 핵인의 발달이 지연 되거나 비정상적으로 발달하는 양상을 보인다. 이번 연구에서는 쥐의 난자를 이용해 정자미세주입법과 단위발생 그리고 난구세포를 이용해 핵이식을 수행했을 각각의 경우에 착상 전 배아가 발달하는 동안 핵인의 발달과 핵인의 전구체 형성 그리고 핵인의 미세구조와 핵인을 조절하는 부위의 활성화 정도를 살펴보고 비교해보기 위한 실험을 수행하였다. 결과를 살펴보면 난구세포를 이용해 핵이식을 수행한 난자에서는 정자미세주입법과 단위발생 시킨 난자보다 전핵당 전구체의 개수가 더 많은 결과를 보였다. 그리고 난구세포를 이용하여 핵이식을 수행한 난자의 4cell 단계에서 손상된 핵인이 활성화 되는 것을 살펴볼수 있었고 미세구조 단계에서부터 질이 덜 좋은 핵인이 발달하고 또 한 염색체상에서 핵인을 조절하는 부위의 활성화 정도가 떨어지는 것을 살펴볼 수 있었다. 그리고 난구세포를 이용하여 핵이식을 수행한 난자의 경우 정자미세 주입법이나 단위 발생 난자에 비해 배반포 형성 비율이 억제되는 것을 살펴볼 수 있었다. 결론적으로 핵이식 난자에서 손상된 핵인의 발달이 수행되며 핵이식 된 난자의 착상 전 발달 능력이 현저히 떨어지는 데 영향을 미치는 요인중에 하나가 핵이식 된 난자에서 손상된 핵인이 형성되기 때문이라고 설명할 수 있다. The most well-known function of nucleolus is ribosome biogenesis. The re-establishment of functional nucleoli after zygotic genome activation is essential for successful development of mammalian pre-implantation embryos. However, in somatic cell nuclear transfer (SCNT) embryos, the nucleoli always showed delayed developed or malfunction. In current study, mouse intracytoplasmic sperm injection (ICSI), pathenogenetic activated (PA) and cumulus cell nuclear transfer (CCNT) embryos were generated to compare the nucleoli development during pre-implantation development, including nucleolar precursor bodies formation, nucleolar ultrastructure and active nucleolar organizer region (NOR) numbers. The results showed that, CCNT embryos had more nucleolar precursor bodies (NPBs) per pronucleus than ICSI and PA embryos at pronuclear stage (2.82 v.s. 0.98 and 1.19). At 4-cell stage, the CCNT embryos showed impaired nucleolar activity, including inferior nucleolar development at ultrastructure level and less active NORs on chromosomes (6.68 v.s. 7.44 and 7.56). Moreover, CCNT embryos showed significant lower blastocyst rate than ICSI and PA embryos (62.0% v.s. 97.1% and 98.0%). The results demonstrated that mouse CCNT embryos had impaired nucleolar development, which may account for their low pre-implantation developmental competence.

      • 성선자극호르몬의 투여가 생쥐 난자의 각인유전자의 DNA메틸화에 미치는 영향에 관한 연구

        성선자극호르몬에 의한 포유동물의 과배란 유도는 많이 사용되어 온 효과적인 방법이다. 그러나 이러한 과배란 유도방법에 의한 난포의 성장 및 성숙은 난자의 성숙과 함께 이루어지기에 난자의 각인유전자 발현과 동일한 시기로서 각인유전자의 DNA 메칠화에 영향을 미칠 염려도 있다. 본 연구의 목적은 과량의 성선자극 호르몬으로 난소를 자극하였을 때 난자 각인인자의 DNA 메칠화에 영향을 미치는지를 관찰 하는 것 이다. 대조군은 6주령된 ICR 암컷 생쥐에 48시간 간격으로 PMSG와 hCG를 각각 5 IU씩 복강투여 하였고 처리군은 같은 방법으로 PMSG와 hCG를 각각 10 IU씩 투여하였다. 그리고 성숙된 (MII단계) 난자는 hCG투여한 13-15시간 뒤에 난관으로부터 회수하여 각인인자 Snrpn 와 Peg3의 DNA 메틸화를 Bisulfite 시퀀싱 방법으로 분석하였다. 분석결과, PMSG를 5 IU 투여한 암컷 마우스의 성숙난자는 Snrpn 와 Peg3 유전자 모두 정상적인 메틸화 상태를 나타냈다. 그러나, 정량 보다 두 배의 PMSG를 투여한 생쥐의 난자는 Snrpn 유전자는 정상적인 메틸화 양상을 나타냈지만, Peg3 유전자는 대조군에 비해 비정상적인 DNA 메틸화 양상을 나타냈다. 이 연구결과는 과량의 성선자극호르몬의 투여는 난자 각인유전자의 DNA 메틸화에 영향을 미친다는 것을 시사한다. Superovulation or ovarian stimulation with gonadotropin is an effective way to enhance the availability of oocyte. Therefore, it is currently an indispensable ART (assisted reproductive technology) base on its significant benefit. However, ovarian stimulation was performed during oocyte growth, thus it may likely affect imprinted methylation as genomic imprinting was established in oocytes during the same time window. In current study, we investigated the impact of superovulation with excessive gonadotropin on methylated imprints in oocytes. MII oocytes were collected from female ssuperovulated with single-dose and double-dose PMSG. DNA methylation profiles of two imprinted genes Snrpn and Peg3 were evaluated by bisulfite sequencing. Our results showed that Snrpn and Peg3 were hypermetylated in the oocytes from single-dose superovulation females. In contrast, although all clones of Snrpn were hypermethyled in oocytes from double-dose superovulation females while some clones of Peg3 were hypomethylated. Our data demonstrated that genomic imprinting was correctly acquired in oocytes from females superovulated with single-dose gonadotropin while excessive stimulation affected genomic imprinting in oocytes, suggesting that DNA methylation in oocytes may be more susceptible to disruption of methylated imprint when superovulated with excessive gonadotropin.

      • 肉鷄 飼料添加劑로서 柴胡莖葉의 利用

        Diets containing 0.0%, 0.2%, 0.5%, and 1.0% of SLPBFL were fed to broiler chickens at 7 weeks age. The results of SLPBFL feeding are as follows; Body weight gain was significantly improved in 0.5% SLPBFL diet group ( p < 0.01 ). Because feed consumption was increased with adding of SLPBFL, broilers taste was not considered in this experiment. Feed requirement was analyzed same score to 1.99 in 0.2% and 0.5% SLPBFL added rations be improved about 8% than the control group. The amounts of carcass, drumsticks and breast meat were significantly(p<0.05) plenty in 0.5% treatment, and also the carcass ratio was enhanced to the highest score in 0.5% SLPBFL, In 0.5% treatment, birds were improved of growth rate, the head and neck weights, leg and shank weight, and amounts of blood and feather were higher appeared than other treatments. The weight of internal organs (liver, heart, spleen, gizzard) were appeared to be heavy in 0.5% treatment. The fat accumulation (abdominal and gizzard surrounding) was observed from the broiler chickens fed 0.5%, tended to increase with fat contents agreeably to live weight gain (P<0.05). Total serum protein, serum bilirubin and LDL-cholesterol concentrations in non- SLPBFL were lower than those in SLPBFL treatments. The concentrations of serum GPT, serum GOT, serum albumin and HDL- cholesterol in non-SLPBFL bird were the lowest in treatment. The concentrations of serum HDL-cholesterol and albumin in 0.5% treatment to be were appeared to be highest score and the concentration HDL-cholesterol was analyzed the lowest score, and total serum protein, serum GPT, serum GOT, serum bilirubin, total serum cholesterol, HDL-cholesterol and serum triglyceride concentrations were analyzed to be mean score in experimental diets.         Studies on the utilization added in diet with a Stem and Leaf Powder of Angelicae Gigas Nakai(SLPAGN) in the broiler chickens. Diets added with 0.0%, 0.2%, 0.5%, and 1.0% of SLPAGN were fed to broiler chickens at 7 weeks age. The results of SLPAGN feeding are as follows; The body weight gain was significantly improved in 1.0% SLPAGN diet (P<0.01), and feed efficiency was effected with 1.0% SLPAGN ration(P<0.05). The amount of carcass was the heaviest in 0.2% SLPAGN diet. The amounts of breast meat and drumsticks product were the highest in 0.2% SLPAGN, and the weights of head and neck, leg and shank were lower than other treatments. The fat accumulation (abdominal and gizzard surrounding) was observed in broilers fed diets added with 0.2% SLPAGN. The weights of liver, spleen and gizzard were tended to increase with adding SLPAGN rations than control diet. The degradation of fabricius sac was delayed to non-SLPAGN treatment than SLPAGN groups. The blood contents were analyzed to be the highest level to the total serum, GOT, total serum cholesterol and LDL-cholesterol concentrations in the broiler chickens fed 1.0% SLPAGN diets, and then the level of total serum protein and HDL-cholesterol concentrations were appeared to be the lowest. In the birds fed non-SLPAGN treatments the concentration of serum GPT, serum albumin, total serum triglyceride, total serum cholesterol and LDL-cholesterol were analyzed to be the lowest levels. In 0.2% SLPAGN treatment, the contents of serum GOT and serum bilirubin contents were analyzed the lowest levels. Key words: Angelicae Gigas, feed efficiency. HDL-cholesterol.
