酸棗仁이 CT105에 의한 신경세포 상해 및 白鼠의 기억에 미치는 영향

Objective: This study was designed to research whether demonstrate neuroprotective and memory enhancing effects of Z1BU, suggesting its beneficial actions for the treatment of AD. Methods: The present study investigated effects of the dichloromethane extract of Semen Zizyphi Spinosae(ZIBU) on neurotoxicity and reactive oxygen species(ROS) and nitric oxide(NO) formation in PC 12 cell line overexpressed with carboxyl-terminal 105 amino acid peptide fragment of amyloid precursor protein(CT105). In addition, the present study evaluated its radical scavenging activity and effects on acetylcholinesterase(AChE) activity. Furthermore, effects on neuronal loss in CAl area and cognitive deficits induced by scopolamine treatment were also evaluated in rats. Result: we found in this study that ZIBU significantly inhibited apoptotic neuronal death induced by CT105 overexpression in PC12 cells, with the IC_(50) value of 23 ㎍/ml. Based on phase-contrast and fluorescence microscopic examinations, ZIBU reversed morphological changes of CT105-expressed PC-12 cells. It was also found that Z1BU significantly induced neurite outgrowth and inhibited formation of ROS and NO.Z1BU scavenged DPPH radicals and inhibited AChE activity. Furthermore, it attenuated memory impairment induced by the injection of scopolamine in rats, based on passive avoidance test. Crystal violet staining of rat brain slices exhibited increase in the viability of pyramidal neurons in hippocampal CAI area. Conclusion: these results demonstrate neuroprotective and memory enhancing effects of ZIBU, suggesting its beneficial actions for the treatment of AD.

Knoellia aerolata sp. nov., isolated from an air sample in Korea

<P>An aerobic, Gram-positive, non-motile, non-spore-forming, rod-coccus-shaped bacterium, strain 5317S-21(T), was isolated from an air sample from Suwon city, Republic of Korea. The isolate was able to grow within a pH range of 5.0-9.0 and a temperature range of 5-35 degrees C and it tolerated up to 2 % (w/v) NaCl. The cell-wall peptidoglycan contained meso-diaminopimelic acid as diagnostic diamino acid. The predominant isoprenoid quinone was MK-8(H(4)). The major polar lipids were phosphatidylinositol, phosphatidylethanolamine and diphosphatidylglycerol; phosphatidylglycerol and several unknown phospholipids were also detected. Mycolic acids were absent. The only whole-cell sugar was glucose. The major cellular fatty acids were iso-C(16 : 0), C(17 : 1)omega8c and iso-C(15 : 0). 16S rRNA gene sequence analysis indicated that strain 5317S-21(T) was related phylogenetically to members of the genus Knoellia, with 97.4 % sequence similarity to the type strains of Knoellia sinensis and Knoellia subterranea. The G+C content of the genomic DNA of strain 5317S-21(T) was 73 mol%. Levels of DNA-DNA relatedness between strain 5317S-21(T) and the type strains of Knoellia sinensis and Knoellia subterranea were 37 and 41 %, respectively. It was concluded that strain 5317S-21(T) represents a novel species of the genus Knoellia, for which the name Knoellia aerolata sp. nov. is proposed. The type strain is 5317S-21(T) (=KACC 20583(T) =DSM 18566(T)).</P>
Massilia jejuensis sp. nov. and Naxibacter suwonensis sp. nov., isolated from air samples

<P>Two Gram-negative, motile, rod-shaped bacteria (strains 5317J-18<SUP>T</SUP> and 5414S-25<SUP>T</SUP>) were isolated from air samples collected in the Jeju Island and Suwon region of Korea, respectively. Phylogenetically, strain 5317J-18<SUP>T</SUP> was grouped with the genus <I>Massilia</I> with <I>Massilia brevitalea</I> byr23-80<SUP>T</SUP> as the closest relative (98.8 % sequence similarity). Strain 5414S-25<SUP>T</SUP> was affiliated with the genus <I>Naxibacter</I> with <I>Naxibacter haematophilus</I> CCUG 38318<SUP>T</SUP> as the closest relative (98.8 % sequence similarity). The mean DNA-DNA relatedness values between strain 5317J-18<SUP>T</SUP> and <I>M. brevitalea</I> DSM 18925<SUP>T</SUP> and <I>Massilia aurea</I> DSM 18055<SUP>T</SUP> were 43 and 36 %, respectively. The mean DNA-DNA hybridization values between strain 5414S-25<SUP>T</SUP> and <I>N. haematophilus</I> KACC 13771<SUP>T</SUP>, <I>M. brevitalea</I> DSM 18925<SUP>T</SUP>, <I>Massilia timonae</I> DSM 16850<SUP>T</SUP>, <I>Naxibacter varians</I> KACC 13770<SUP>T</SUP>, <I>M. aurea</I> DSM 18055<SUP>T</SUP>, <I>Massilia lutea</I> DSM 17473<SUP>T</SUP> and <I>Massilia albidiflava</I> DSM 17472<SUP>T</SUP> ranged from 33 to 42 %. Both novel strains had ubiquinone Q-8 as the predominant isoprenoid quinone and summed feature 3 (comprising iso-C15 : 0 2-OH and/or C16 : 1<I>ω</I>7<I>c</I>) and C16 : 0 as the major fatty acids. Both strains also showed similar polar lipid profiles with phosphatidylethanolamine, phosphatidylglycerol and diphosphatidylglycerol as the major polar lipids. The DNA G+C contents of strains 5317J-18<SUP>T</SUP> and 5414S-25<SUP>T</SUP> were 66.1 and 67.8 %, respectively. On the basis of their phenotypic, chemotaxonomic and genotypic characteristics, the new strains represent novel species in the genera <I>Massilia</I> and <I>Naxibacter</I>. Strain 5317J-18<SUP>T</SUP> (=KACC 12634<SUP>T</SUP>=DSM 21309<SUP>T</SUP>) is proposed as the type strain of <I>Massilia jejuensis</I> sp. nov. and strain 5414S-25<SUP>T</SUP> (=KACC 12635<SUP>T</SUP>=DSM 21311<SUP>T</SUP>) is proposed as the type strain of <I>Naxibacter suwonensis</I> sp. nov.</P>
Pseudomonas pohangensis sp. nov., isolated from seashore sand in Korea.

<P>A novel bacterium, designated strain H3-R18(T), was isolated from seashore sand collected from Homi cape, Pohang city, Korea. Cells were Gram-negative, aerobic, non-motile, cream-coloured, mesophilic and slightly halotolerant. 16S rRNA gene sequence analysis indicated that the organism was a member of the genus Pseudomonas, but the sequence showed < or =96.3 % sequence similarity to that of the type strains of all recognized Pseudomonas species. Highest sequence similarities were to Pseudomonas brenneri CFML 97-391(T) (96.3 %) and Pseudomonas migulae CIP 105470(T) (96.3 %). The major fatty acids were summed feature 3 and C(16 : 0), with lesser amounts of C(12 : 0), C(12 : 0) 3-OH, C(18 : 1)omega7c and C(14 : 0). The major isoprenoid quinone was Q-9. The DNA G+C content was 64.0 mol%. The phylogenetic, phenotypic and genetic properties of strain H3-R18(T) suggest that it represents a novel species, for which the name Pseudomonas pohangensis sp. nov. is proposed. The type strain is H3-R18(T) (=KACC 11517(T)=DSM 17875(T)).</P>
Paludibacterium yongneupense gen. nov., sp. nov., isolated from a wetland, Yongneup, in Korea.

<P>A cream-coloured bacterial strain, 5YN8-15(T), was isolated from a wetland, Yongneup, in the Inje region of the Republic of Korea. The bacterium was facultatively anaerobic, Gram-negative, motile with a single polar flagellum and curved-rod-shaped. Based on 16S rRNA gene sequence analysis, strain 5YN8-15(T) is a member of the Betaproteobacteria. Closely related taxa were Gulbenkiania mobilis E4FC31(T) (94.9% sequence similarity), Chromobacterium species (94.1-94.4%), Aquitalea magnusonii TRO-001DR8(T) (93.2%) and Aquaspirillum serpens IAM 13944(T) (92.5%). All other species with validly published names analysed showed sequence similarities of below 92%. Strain 5YN8-15(T) had ubiquinone 8 as the major isoprenoid quinone. The major fatty acids were summed feature 3 (C(16:1)omega7c and/or iso-C(15:0) 2-OH), C(16:0) and C(18:1)omega7c. The DNA G+C content was 63.0 mol%. Based on the data from the polyphasic study, strain 5YN8-15(T) represents a novel genus and species of the family Neisseriaceae, for which the name Paludibacterium yongneupense gen. nov., sp. nov. is proposed. The type strain is 5YN8-15(T) (=KACC 11601(T)=DSM 18731(T)).</P>
