Genetic Relationship of Hibiscus syriacus L. Clarified by AFLP and Morphological Evaluation

Genetic diversity and relationship of Hibiscus syriacus L. were analyzed using 127 cultivars and 7 landraces by the amplified fragment length polymorphism (AFLP) and the assessment of morphological characteristics. The AFLP analysis was carried out by using capillary electrophoresis with 28 fluorescent labeled primer combinations. Morphological evaluation was executed by the 11 parameters. A multivariate data analysis was performed using similarity coefficients, Euclidean distance, unweight pair group method with arithmetic averages (UPGMA) clustering method, and principal coordinate analysis (PCoA) of NTSYS-pc program. In the AFLP analysis, a total of 2, 842 bands were produced in the 28 primer sets, and 43.4% (1,236 bands) of them were identified to be polymorphic. Similarity coefficient values ranged between 0.49 and 0.98. Scattered distribution of the 127 cultivars and 7 landraces was identified, and they were divided into three major groups based on PCoA and UPGMA clustering analysis. In the morphological evaluation, Euclidean distance values ranged from 0.28 to 10.32. The distribution showed that there was a little difference compared with the result of AFLP. The pedigrees of several cultivars were cross-correlated using the results of AFLP and morphological analysis.
RPL36 as a prognostic marker in hepatocellular carcinoma

<P>Ribosomal proteins (RP) play key roles in the regulation of apoptosis, multidrug resistance and carcinogenesis. The aim of this study was to investigate the expression of ribosomal protein L36 (RPL36) in hepatocellular carcinoma (HCC) and to correlate it with clinicopathological parameters and clinical outcome. Liver specimens were obtained from 60 HCC patients who had undergone a partial hepatectomy. Expression of RPL36 in tumor tissue and surrounding non‐tumorous tissues was evaluated on a tissue microarray by immunohistochemistry. RPL36 was expressed in 45 of 60 (75%) HCC by immunohistochemistry, but was not detected in corresponding non‐tumors. RPL36 expression correlated significantly with serum levels of albumin (<I>P</I>= 0.044) and prothrombin time (<I>P</I>= 0.026), which reflect liver synthetic function. Moreover, expression of RPL36 was found to be higher in patients with early tumor stages (I/II) (<I>P</I>= 0.038) or without portal vein thrombosis (<I>P</I>= 0.005). In univariate analysis, patients with RPL36 expression revealed better overall survival (<I>P</I>= 0.037). By multivariate survival analysis, RPL36 expression was found to be an independent prognostic factor for overall survival (<I>P</I>= 0.026). Our data suggest that RPL36 may be involved in the early stage of hepatocarcinogenesis, and it can be used as an independent and potential prognostic marker for resected HCC.</P>

A blue protein was purified from the Methylobacillus sp. strain SK1 that is grown on methanol in the pres-ence of copper ion. This protein was found to be a monomer with a molecular weight of 13,500. The Isoelectric point of the protein was estimated to be 8.8. The spectrum of the protein that was treated with fer-ricyanide showed a broad peak around 620 nm, but that of the dithionite-treated protein revealed no peaks. It contained 0.83 mol of EDTA-stable copper per mol protein. Under air, the protein accelerated the inacti-vation of methanol dehydrogenase (MDH). The pro-tein was reducible by phenazine methosulfate or by active MDH that was prepared from cells that were grown in the absence of added copper, but not by methanol, dichlorophenol indophenol, or inactive MDH that was prepared from cells that were grown in the presence of added copper. It was also reducible by active MDH in the presence of methanol. The absorp-tion peak at 340 nm of the active MDH disappeared after the enzyme was treated with ferricyanide, hy-drogen peroxide, or the purified blue protein. The in-active MDH also showed no peak at 340 nm. The 340-nm peak was not recovered after incubation of the inactive MDH and blue protein-treated active MDH with dithionite or methanol. The inactive MDH and blue protein-treated active MDH co-migrated with the active MDH preparation on nondenaturing poly-acrylamide gel, and contained two non-identical sub-units with molecular weights that were identical to those of the active MDH. The N-terminal amino acid sequence of the protein was Ala-Gly-Cys-Ser-Val-Asp-Val-Glu-Ala-Asn-Asp-Ala-Met-Gln-Phe. An analysis of the amino acid composition revealed that the pro-tein contained no tryptophan. It contained three cys-teines per mol protein. The blue protein in Methyloba-cillus sp. strain SK1 was produced only in the cells that were grown in the copper-supplemented medium.

The aim of this study was to evaluate the effects of tumbling condition (time and temperature) on the quality characteristics of restructured chicken breast ham. Tumbling conditions were 10, 30, and 60 min at 3 or -3oC, respectively. After tumbling, quality characteristic regarding the treatments processed by each condition were measured. There were no significant differences in pH levels among tumbling time or temperature. Treatments involving 60 min of tumbling had significantly higher water holding capacity (WHC) than that tumbled for 10 min, but WHC was not affected by the tumbling temperature. The tumbling condition at -3oC for 60 min demonstrated the most effective cooking yields for restructured chicken breast ham. Myofibrillar protein solubility of treatments tumbled for 60 min had the highest value of all, regardless of tumbling temperature. The hardness of treatments tumbled at -3oC were lower than those at 3oC; however, the springiness of treatments tumbled for 60 min were significantly higher than those regarding the treatments tumbled for 10 and 30 min. Therefore, tumbling technology for 30 min or more at -3oC can produce restructured chicken breast ham of excellent quality.

Bowen's disease was originally described by Bowen in 1912 and is believed to be a squamous cell carcinoma in situ. It develops on any part of the .3kin surfaces or mucous membranes. But the lesions on the vulvar region are relatively ran. The gross appearance of vulvar lesion, which shows erosion, ulceration or pigmentation, is different from that of typical Bowen's disease. We, herein, report a case of Bowen's disease arising on a vulvar region with a review of the literature.(Korean j Dermatol 2003:41(1) : 112∼115)

<P>Langerhans cell histiocytosis (LCH) and Langerhans cell sarcoma (LCS) are clonal proliferations of Langerhans-type cells. Unlike in LCH, the pathophysiology and clinical course of LCS are unclear due to its rarity. Here, we report the case of a 73-year-old male patient who was diagnosed with cutaneous LCH and pulmonary LCS at the same time. Pathological review of these 2 tumors revealed similar immunohistochemical findings. However, the tumor cells in LCS had more aggressive cytological features than those in LCH. Results of BRAF mutation analysis using real-time PCR were negative for both tumors. In whole-exome sequencing (WES), stop-gain mutations in TP53 gene were discovered only in LCS cells. The mechanism of development of LCS from various progenitor cells is currently unclear. According to the results of the WES study, changes in TP53 gene might have contributed to the malignant features of LCS.</P>
