      • Ficin의 酵素化學的性質에 관한 硏究

        One of ficin, isolated from Fig latex, was conducted to investigate some chemical properties of enzyme. The results are as follows; 1. The activity of crude ficin was 2.3 unit/mg and milk clotting activity was 8.33x10-²unit/mg 2. The hydrolytic activity to substrate was 6.8%, which is thought to be of low value. 3. While activities of enzyme after adding 10?? Mcysteine and those concentration of 2-mercaptoe thanol were high. 10-³M-Na₂S₄O?? was the strongest inhibitor among others. 4. The value of enzyme activity was the highest when the concentration of casein solution was 3%.

      • 리놀레산 과산화물이 β-amylase 활성도에 미치는 영향

        자동산화 지질과 단백질과의 상호 작용을 알아보기 위하여 모델 시스템에서 리놀레산 과산화물이 β-amylase의 활성도에 미치는 영향을 실험한 결과 여러 온도와 pH 조건하에서 리놀레산 과산화물에 의해 β-amylase 활성도는 감소되었다. 또한 각종 라디칼 소거제와 금속이온 존재하에서 β-amylase 활성을 측정한 결과와 전기영동 결과에 비추어 볼 때 효소의 활성 감소는 시스템내에 존재했던 라디칼에 의해서 효소 자체에 손상을 입혀 비롯된 것으로 사료되었다. For a better understanding the interactions of autoxidizing lipids with enzyme proteins, the effect of linoleate hydroperoxides (LAHPO) on β-amylase activity was tested. The activity of β-amylase was largely decreased by LAHPO at various temperatures and pH. The inactivation of β-amylase was due to radicals in system because ascobate and metal ion enhanced the inactivation of enzyme by LAHPO. The courses of SDS-PAGE of β-amylase in system showed that LAHPO caused scission of the enzyme.

      • 방아풀(Isodon japonicus)의 精油成分에 關한 硏究

        방아풀(Isodon japonicus)의 一般成分 및 精油成分을 分析한 結果는 다음과 같다. 가용성 무질소물은 30.5%로 가장 많았으며 精油의 含量은 生體試科 100g당 0.6ml였으며 精油의 Liquid chromatography에서 9개 peak가 존재하였으며 그중 9번 peak가 97.48%였다. Liquid chromatogram의 9번 peak분획 精油의 比重은 0.97, 屈扮率은 1,513沸點은 221℃ 였으며 uv의 吸光은 226nm와 276nm에서 강한 吸光이 나타났고 Elemental analyzer의 分析 결과 C는 81.0%, H는 18.8%, N은 0.2%였다. Infrared spectra로 보아 Liquid chromatogram의 9번 peak의 物質은 piperitone 類似物質로 R基가 -CH₂=CH₂-CH₃인 것으로 思料된다. To get the basic informations for the utilization of spicery in Isodon japonicus, the chemical compotition and essential oil of that were analyzed by chromatography. The results obtained were as follows; 1. The contents of soluble nitrogen-free substance was 30.5%, and essential oil was 0.6ml/100g fresh weight basis. 2. In analyzing the Isodon japonicus by the liquid chromatography, there were 9 peaks, of which the 9th peak was 97.48%. 3. In the 9th prak of essential oil, the spesific gravity was 0.97, 1.513 in reflective index, 221℃ in boiling point, 226nm and 276nm in Uv absorbance. 4. The ratio of C>N. and H were 81.0%, 0.2% and 18.8%, respectively. 5. It is assumed that the 9th peak's substance was piperitione analogue substance and the radical group of that was -CH₂=CH₂-CH₃.

        A Colorimetric Method for Estimation of Fermented Degree in Black Tea Processing

        This study was conducted to determine which color attribute change the more during fermentation in tea processing and if relationship between a tea pigment compound and the color index by fermentation level. Fermentation was performed after rolling. Hunter’s color value was measured either by using colorimeter or spectrum color meter. The liquor of finished tea was subjected to chemical analyses for total theaflavins, thearubigin, caffeine,catechin compounds. Hunter color b-value, yellowness, decreased during fermenting process while it was slightly increased at earlier stage. Hunter color a-value, redness, increased from minus value to plus during fermentation. The color b-value was higher than a-value in an earlier fermentation process. The difference Hunter value between b and a decreased gradually as fermentation time was increased, which values meet on a point at a certain fermentation time. After this time, color a-value was higher than the b-value. The result suggested that the color difference value between Hunter b and a can be applied on estimating fermented degree in black tea processing, with a relationship by the equation, ‘Fermentated Degree (%) = Hunter value b - a’. Total theaflavin increased until 30 min fermentation by 91.39 ± 4.80 mg% and then decreased by 36.79 ± 1.90 mg% at 180 min fermentation, while thearubigin increased until 120 min fermentation by 63.1 ± 1.92 mg% and then decreased by 30.8 ± 1.48 mg% at 180 min fermentation level. Caffeine and catechin compounds such as EGCg and ECg decreased as fermentation levels were high. These results was in agreement with the result that tea catechins changed to thearubigins through theaflavins. These results of chemical analysis also showed that the color difference data, Hunter value b-a, can be used to determine fermentation-stop point during black tea processing. If ICT technology is applied on automation of tea processing, the control of fermentation process can possibly be controlled and standardized by monitoring with automatic color difference meter.

        Bioactivity-Guided Fraction from Viscera of Abalone, Haliotis discus hannai Suppresses Cellular Basophils Activation and Anaphylaxis in Mice

        Basophils and mast cells are specialized effector cells in allergic reactions. Haliotis discus hannai (abalone), is valuable seafood. Abalone male viscera, which has a brownish color and has not been previously reported to show anti-allergic activities, was extracted with acetone. Six different acetone/hexane fractions (0, 10, 20, 30, 40, and 100%) were obtained using a silica column via β-hexosaminidase release inhibitory activity-guided selection in phorbol myristate acetate and a calcium ionophore, A23187 (PMACI)-induced human basophils, KU812F cells. The 40% acetone/hexane fraction (A40) exhibited the strongest inhibition of PMACI-induced-β-hexosaminidase release. This fraction dose-dependently inhibited reactive oxygen species (ROS) production and calcium mobilization without cytotoxicity. Western blot analysis revealed that A40 down-regulated PMACI-induced MAPK (ERK 1/2, p-38, and JNK) phosphorylation, and the NF-κB translocation from the cytosol to membrane. Moreover, A40 inhibited PMACI-induced interleukin (IL)-1β, IL-6, and IL-8 production. Anti-allergic activities of A40 were confirmed based on inhibitory effects on IL-4 and tumor necrosis factor alpha (TNF-α) production in compound (com) 48/80-induced rat basophilic leukemia (RBL)-2H3 cells. A40 inhibited β-hexosaminidase release and cytokine production such as IL-4 and TNF-α produced by com 48/80-stimulated RBL-2H3 cells. Furthermore, it’s fraction attenuated the IgE/DNP-induced passive cutaneous anaphylaxis (PCA) reaction in the ears of BALB/c mice. Our results suggest that abalone contains the active fraction, A40 is a potent therapeutic and functional material to treat allergic diseases.

        COMPONENT-BASED DEVELOPMENT OF OBSERVATIONAL SOFTWARE FOR KASI SOLAR IMAGING SPECTROGRAPH

        In this paper, we have made the component-based development of observational software for KASI solar imaging spectrograph (KSIS) that is able to obtain three-dimensional imaging spectrograms by using a scanning mirror in front of the spectrograph slit. Since 2002, the KASI solar spectrograph has been successfully operated to observe solar spectra for a given slit region as well as to inspect the response functions of narrow band filters. To improve its capability, we have developed the KSIS that can perform sequential observations of solar spectra by simultaneously controlling the scanning mirror and the CCD camera via Visual C++. Main task of this paper is to introduce the development of the component-based software for KSIS. Each component of the software is reusable on the level of executable file instead of source code because the software was developed by using CBD (component-based development) methodology. The main advantage of such a component-based software is that key components such as image processing component and display component can be applied to other similar observational software without any modifications. Using this software, we have successfully obtained solar imaging spectra of an active region (AR 10708) including a small sunspot. Finally, we present solar $H{\alpha}$ spectra ($6562.81{\AA}$) that were obtained at an active region and a quiet region in order to confirm the validity of the developed KSIS and its software.

        배초향 잎의 유용성분과 특성

        배초향 잎에 함유된 유용성분을 개발하고 식품에 적용시키기 위해 기호성에 미치는 각종 성분을 분석하였다. 시기별 수분과 회분의 함량은 각각 74~82%, 1.5~3.4%에 달했으며 유리당의 주요 성분은 fructose, glucose, sucrose, maltose로 나타났다. 주요 구성아미노산은 glutamic, aspartic acid, leucine이었으며 특히 유엽과 여름철 시료에서 높은 함량을 나타냈다. 유리아미노산 중에서는 histidine이 가장 높은 함량을 보였으며 methionine은 적게 함유된 것으로 나타났다. 무기질의 주요 성분은 칼륨, 칼슘, 나트륨이었며 마그네슘을 포함하여 대부분의 성분이 다른 식물체에 비해 높은 것으로 평가되었다. Ascorbic acid의 함량은 6.6~9.2㎎% 범위를 나타냈으며 시기별로는 큰 차이가 없었다. 시기별 배초향 잎에 함유된 폴리페놀류의 함량은 109~181㎎%에 달했으며 유리형보다는 결합형이 다량 검출되었고, 주요 페놀화합물은 caffeic, syringic, ferulic acid로 나타났다. In order to develope useful components and to apply in food additives, various components in the leaves of baechohyang were determined according to picking date. Moisture and ash were reached to 74~82%, 1.5~3.4%, respectively, and major sugars were fructose, glucose, sucrose, and maltose. Glutamic, aspartic acid, and leucine constituted the main component amino acids and their contents were relatively higher in the samples of summer and young leaves. Among free amino acids, histidine showed the highest components and methionine were the lowest. Potassium, calcium, and sodium were main mineral components. The content of ascorbic acid were 6.6~9.2㎎% and seasonal variations were negligible. Muture leaves contained more polyphenols as much as 108~181㎎% and the amount of bound fractions were much more. Periodic evaluation of phenolic compounds were made and major components of bound fractions were caffeic, syringic, and ferulic acid.
