Inhibitory Effects of Acanthopanax chiisanensis Ethanolic Extracts on FcεRI α Chain Expression

호염기구와 비만세포는 FcεRI을 매개로 한 알러지 반응에 있어 효과세포로서 중요한 역할을 담당한다. 인간 유래 호염기구성 세포주 KU812F 세포의 FcεRI α chain 발현에 있어 지리 오갈피의 저해 효과에 대해 연구하였다. 지리 오갈피의 뿌리 및 줄기를 에탄올로 추출하여, FcεRI α chain 저해 활성 실험에 이용하였다. 세포 표면의 FcεRI α chain 발현량을 flow cytometry로 분석한 결과, 지리 오갈피 뿌리 및 줄기 추출물에서 세포표면의 FcεRI 발현을 억제하는 효과를 나타냈다. 또한 지리 오갈피 뿌리 및 줄기 추출물은 FcεRI α chain mRNA 발현을 감소시켰으며, FcεRI을 매개로 한 히스타민 유리를 감소시켰다. 이러한 결과는 지리 오갈피의 뿌리 및 줄기 추출물이 FcεRI α chain 발현의 저하 조절 및 히스타민과 같은 염증매개인자의 분비를 저하시킴으로서 항 알러지 활성을 갖는데 중요한 역할을 할 것으로 판단된다. Basophils and mast cells play an important role in FcεRI-mediated allergic reaction as effector cells. We studied the effects of Acanthopanax chiisanensis on FcεRI α chain expression in human basophilic KU812F cells. Ethanol extracts from root and stem of A. chiisanensis were tested for inhibitory effects of FcεRI α chain expression. The cell surface FcεRI α chain expression was examined by flow cytometric analysis. All of the extracts of A. chiisanensis reduced the cell surface FcεRI α chain expression. Furthermore, A. chiisanensis extracts caused a decrease in the level of FcεRI α chain mRNA level and FcεRI-mediated histamine release. These results suggest that root and stem extracts of A. chiisanensis play an important role in anti-allergic activity via down-regulation of FcεRI α chain expression and decrease in release of inflammatory mediator such as histamine.

Pandanus fascicularis Lam has been shown to exert to a variety of physiological effects on edema, tumors, leprosy, spasm, inflammation, pain, and rheumatoid arthritis. In this study, the effects of a P. fascicularis extract on the production of lipopolysaccharide (LPS)-induced pro-inflammatory cytokines interleukin (IL)-1β, IL-6, and tumor necrosis factor (TNF)-α expression and on the production of the inflammatory mediator, nitric oxide (NO), in RAW 264.7 cells were examined. The P. fascicularis extract decreased LPS-induced NO production. Moreover, enzyme-linked immunosorbent assays revealed that the P. fascicularis extract concentration-dependently suppressed LPS-induced production of the pro-inflammatory cytokines IL-1β, IL-6, and TNF-α. These results suggest that the P. fascicularis extract has potential as an anti-inflammatory therapeutic substance for use in the prevention of the inflammatory disorder.
Achyranthes japonica Nakai Water Extract Suppresses Binding of IgE Antibody to Cell Surface FcεRI

Achyranthes japonica Nakai (AJN) water extract has a variety of physiological properties, including anti-diabetic, anti-cancer, anti-inflammatory, anti-microbial, and anti-oxidative activities. In the present study, the inhibitory effects of AJN extract were investigated in high affinity immunoglobulin E receptor (FcεRI)-mediated KU812F cells activation. AJN extract showed suppressive effects on histamine release and intracellular calcium [Ca<SUP>2+</SUP>]i elevation from anti-FcεRI antibody (CRA-1)-stimulated cells in a dose-dependent manner. Flow cytometric analysis showed that AJN extract treatment caused a dose-dependent decrease in the cell surface FcεRI expression and the binding between the cell surface FcεRI and the IgE antibody. Moreover, reverse transcription-polymerase chain reaction analysis showed that levels of the mRNA for the FcεRI α chain was decreased by treatment with AJN extract. These results indicate that AJN extract may exert anti-allergic effects via the inhibition of calcium influx and histamine release, which occurs as a result from the downregulation of the binding of IgE antibody to cell surface FcεRI. This mechanism may occur through FcεRI expression inhibition.

Human basophilic KU812F cells express a high-affinity immunoglobulin (Ig) E receptor, $Fc{\varepsilon}RI$, which plays an important role in IgE-mediated allergic reactions. Houttuynia cordata Thunb (Family Saururaceae), which is rich in polyphenols, has been shown to have various physiological properties, including antiviral, antioxidative, anticancer, and anti-inflammatory activities. The effect of H. cordata extract on the expression of $Fc{\varepsilon}RI$ in human KU812F cells was examined. Flow cytometric analysis showed that the $Fc{\varepsilon}RI$ expression and the IgE binding activity were suppressed when the cells were cultured with H. cordata extract. Reverse transcription-polymerase chain reaction analysis showed that levels of the mRNAs for $Fc{\varepsilon}RI{\alpha}$- and $\gamma$-chains were decreased by the treatment of H. cordata extract. Addition of H. cordata extract to culture medium was also observed to result in a reduction in the release of histamine from the cells. These results suggest that H. cordata extract may exert its anti-allergic activity through down-regulation of $Fc{\varepsilon}RI$ expression and a subsequent decrease in histamine release.

In an ongoing search of compounds from natural sources that exhibit anti-allergic properties, 3 flavonoids were isolated from the ethyl acetatefraction of methanol extracts of gujeolcho (Chrysanthemum zawadskii var. latilobum), with guided histamine release inhibitory effects. The isolated compounds were identified as eriodictyol-7-O-${\beta}$-D-glucuronide, 5,7-dihydroxy-4-chromene, and 6-methoxyluteolin based on spectroscopic evidences including NMR and MS spectra. These results suggest that gujeolcho extracts and 3 compounds would be candidates as therapeutic or preventive agents for allergic reaction.
Induction of Rice Allergen-Specific IgE Synthesis by KU812 Cells

CD40L를 발현하는 KU812세포와 CD40를 발현하는 항원 특이적 B 세포의 배양을 통해 IgE 클라스 스위치가 유도된다. 본 연구자는 체외면역법에 의해 건강인의 말초혈액을 이용하여 쌀 알레르겐 특이적 IgM 항체를 생성하는 B 세포주를 수립하였다. 본 연구에서는 KU812세포에 의한 쌀 알레르겐 특이적 IgE 항체의 유도를 시도하였다. 배양 전, 쌀 알레르겐 특이적 B 세포주, RA9G11과 PMA와 ionomycin을 6시간 처리하여 활성화된 KU812세포에서 CD40와 CD40L 발현량을 flow cytomerty를 통화여 각각 확인하였다. RA9G11세포와 활성화된 KU812세포는 높은 수준의 CD40와 CD40L를 각각 발현하였다. 쌀 알레르겐 특이적 IgE항체 합성 유도를 위해, 여러 가지 농도의 IL-4 존재하에, RA9G11세포와 활성화된 KU812세포를 12일 동안 배양하였다. IgE 정상영역의 mRNA 발현량과, 쌀 알레르겐 특이적 IgE 항체 생성 세포가 모든 조건에서 확인되었으며, 특히 50U의 L-4 농도 하에서 같은 비율의 RA9G11과 활성화된 KU812세포를 배양했을 때, IgE 합성이 가장 높았다. 따라서 활성화된 KU812세포에 의한 쌀 알레르겐 특이적 IgE 합성은 알레르기 질환의 치료 및 예방에 관한 연구에 기여할 것으로 사료된다. In vitro IgE class switching could be induced through co-culture of CD40L-expressing KU812 cells and CD40-expressing B cells in the presence of IL-4 or IL-13. It has been generated several B cell lines, which produce rice allergen (RA)-specific IgM antibody by in vitro immunization (IVI) using peripheral blood lymphocyte (PBL). In this study, induction of RA-specific IgE antibody by KU812 cells was attempted. Before co-culture, we determined the CD40 expression in RA-specific B cell lines, RA9G11 and the CD40 ligand (CD40L) expression in activated KU812 cells by treatments with phorbol myristate acetate (PMA) and ionomycin for 6 hrs. Flow cytometric analysis shown that RA9G11 and activated KU812 cells expressed high level of CD40 and CD40L, respectively. RA9G11 cells were cultured with activated KU812 cells for 12 days in the presence of IL-4 for IgE class switching. Mature Cε mRNA level and RA-specific IgE spot forming cells (SFC) were observed in all culture condition, and especially, high level of RA-specific IgE synthesis was determined the same ratio of RA9G11 and activated KU812 cells in the presence of 50U IL-4. Therefore, induction of RA-specific IgE synthesis by activated KU812 cells can be contributed in the application for allergic therapy and prevention.
