        <i>Dendropanax morbifera</i> Prevents Cardiomyocyte Hypertrophy by Inhibiting the Sp1/GATA4 Pathway

        <P>An extract of <I>Dendropanax morbifera</I> branch exerts antioxidant, anti-inflammatory, antithrombotic, and anticancer activities. The purpose of this study was to investigate the effect of the extract in isoproterenol-induced cardiac hypertrophy. Phalloidin staining showed that treatment with the extract dramatically prevents isoproterenol-induced H9c2 cell enlargement and the expression of cardiac hypertrophic marker genes, including atrial natriuretic peptide (ANP) and B-type brain natriuretic peptide (BNP). Further, pretreatment with the extract decreased isoproterenol-induced GATA4 and Sp1 expression in H9c2 cells. Overexpression of Sp1 induced the expression of GATA4. The forced expression of Sp1 or its downstream target GATA4, as well as the co-transfection of Sp1 and GATA4 increased the expression of ANP, which was decreased by treatment with the extract. To further elucidate the regulation of the Sp1/GATA4-mediated expression of ANP, knockdown experiments were performed. Transfection with small interfering RNAs (siRNAs) for Sp1 or GATA4 decreased ANP expression. The extract did not further inhibit the expression of ANP reduced by the transfection of GATA4 siRNA. Sp1 knockdown did not affect the expression of ANP that was induced by the overexpression of GATA4; however, GATA4 knockdown abolished the expression of ANP that had been induced by Sp1 overexpression. The extract treatment also attenuated the isoproterenol-induced activation of p38 MAPK, ERK1/2, and JNK1. Hesperidin, catechin, 2,5-dihydroxybenzoic acid, and salicylic acid are the main phenolic compounds present in the extract as observed by high performance liquid chromatography. Hesperidin and 2,5-dihydroxybenzoic acid attenuated isoproterenol-induced cardiac hypertrophy. These findings suggest that the <I>D. morbifera</I> branch extract prevents cardiac hypertrophy by downregulating the activation of Sp1/GATA4 and MAPK signaling pathways.</P>

      • Effects of increased soil fertility and plant growth-promoting rhizobacteria inoculation on biomass yield, energy value, and physiological response of poplar in short-rotation coppices in a reclaimed tideland: A case study in the Saemangeum area of Korea

        <P><B>Abstract</B></P> <P>This study aims to investigate the physiological response, biomass yield and its bio-energy value to charcoal mineral fertilizer and <I>Bacillus subtilis</I> strain JS, a plant growth-promoting rhizobacteria, under nine different soil-enrichment procedures, and to provide a reference for suitable management strategies for short-rotation coppices on the marginal soils of reclaimed tidelands in Saemangeum area of Korea. The treatments used in this experiment were as follows: 1) 1:100 (volume basis) diluted rhizobacteria inoculation {B1, (<I>B. subtilis</I>: double-distilled water, v/v)}, 2) 1:50 diluted rhizobacteria inoculation (B2), 3) mineral fertilization {F1, (charcoal 200 kg ha<SUP>−1</SUP>)}, 4) additional mineral fertilization {F2, (charcoal 300 kg ha<SUP>−1</SUP>)}, 5) F1+B1, 6) F2+B1, 7) F1+B2, 8) F2+B2, and 9) control plot with no soil enrichment (C). The plant species used in this study is <I>Populus euramericana</I>. Net photosynthetic assimilation using leaves acclimated for 2 min at 1500 μmol m<SUP>−2</SUP> s<SUP>−1</SUP> was the highest in F1+B2, followed by B2+F2 and B2. Total biomass yield in F2+B2 was the highest by a significant margin among the nine soil-enrichment procedures, whereas that in the control soil was lowest. Plants in all soil conditions provided similar net calorific values of dry mass, and they all fell within the third grade of wood pellet quality criteria of the National Institute of Forest Science standard specification (Korea), within the premium category in the Pellet Fuels Institute standard specification (US), and within the A1 category in the EN-Plus standard specification (EU). Moreover, it was confirmed that the poplar was suited to produce wood pellets for heating energy.</P> <P><B>Highlights</B></P> <P> <UL> <LI> The effect of <I>B. subtilis</I> JS and charcoal fertilizer was efficient to increase biomass yield in reclaimed land. </LI> <LI> Biomass in the F2 (charcoal 300 kg ha<SUP>−1</SUP>) + B2 (JS bacterial culture diluted 1:50) was significantly highest among treatments. </LI> <LI> Photosynthetic rate, leaf N content and the T-N in soil had a positive correlation. </LI> <LI> Poplar grown on marginal soil was suitable for wood pellet production. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        The Relationship among Engagement in Workplace Learning Activities, Openness to Learning, and the Extent of Participant Involvement in Informal Learning

        일터학습에 있어서 비공식학습의 중요성이 대두함에 따라, 많은 기업들은 직원들의 비공식 학습을 촉진할 수 있는 다양한 학습전략을사용하고 있다. 기존의 연구들은 기업 내 공식학습과 비공식학습은 상호 연관되어 있어, 공식학습을 제공할 경우 비공식학습도 동시에 활성화된다고 주장한다. 본 연구는 직원들이 기업에서 행해지고 있는 일터학습에 참여하는 정도가 비공식학습활동에 어떠한 영향을 미치는지 연구함으로써, 일터학습의 참여도와 비공식학습의 관계를 알아보고자 하였다. 일터학습에 참여하고 있는 제약회사의 직원들을 대상으로 한 설문조사에 대하여 회귀분석을 실시한 결과, 일터학습에 참여하는 것 자체는 직원들이 비공식학습에 몰입하는 정도에 유의미한 영향을 미치지 못하는 것으로 나타났다. 특히 실천공동체와 온라인 학습의 경우 비공식학습에 부정적 영향을 미치는 것으로 나타났다. 이는 기업이 일터학습을 제공하더라도, 체계적인 형태를 갖추지 못했거나 직원들 간의 상호작용을 촉진하지 못할 경우 일터학습이 오히려 비공식학습을 저해할 수 있다는 것을 보여준다. 그러나 개인의 학습개방성은 비공식학습의 정도에 영향을 미치는 것으로 나타나, 일터학습의 참여도가 비공식학습의 정도에 미치는 영향에 조절효과를 보이는 것으로 나타났다. 끝으로 연구결과는 관리자 직급에 있는 근로자들의 경우 그렇지 않은 직원들에 비하여 비공식학습활동에 더 많이 참여하고 있다는 것을 보여준다. 관리자 직급의 근로자들은 학습에 대한 경험이 풍부하고, 학습의 필요성에 대해서도 충분히 인식하여 비공식학습활동에도 적극적으로 참여하고 있다고 볼 수 있다. 따라서, 기업 내에서 비공식학습을 활성화시키기 위하여, 종업원들에게 일터학습을 제공하고 이것에 참여하도록 유도하는 것뿐만 아니라, 직원들 스스로 학습에 참여할 수 있는 개방적 태도를 지닐 수 있도록 지원하는 것이 중요하다는 것을 보여준다. Informal Learning in the workplace is becoming more important for improving employees` competencies and organizational performance. Many business organizations use a variety of workplace Learning strategies in order to facilitate informal Learning among employees. Research on workplace Learning explains that formal Learning and informal Learning interrelate and facilitate each other. This study investigated the relationship among engagement in workplace Learning activities, openness to learning, and the extent of participant involvement in informal Learning. As a result of quantitative research in a pharmacy company, this study showed the following: (a) openness to Learning affects participant involvement in informal Learning more than engagement in workplace Learning activities does; (b) among types of workplace Learning, communities of practice negatively influence internal informal Learning, and web-based Learning also has a negative effect on involvement in internal, external informal Learning and informal Learning among newly hired employees, but traditional classroom training positively affects participants` involvement in informal learning among new employees; and (c) employees at the managerial Level involve in informal Learning activities more than other employees. These results suggest that business organizations and corporate management should support and facilitate employees to learn with openness as well as provide them with workplace Learning opportunities.

      • 임실지역 당뇨병 환자들의 관리 실태

        연구배경: 당뇨병의 관리에 있어서 만성합병증 예방 및 치료는 당뇨병 환자의 예후와 밀접한 관계가 있다. 그러나, 이는 연령, 지역 및 경제적 여건에 따라 많은 차이를 보일 수 있고 또한 시골지역 고령 환자들의 관리 소홀로 반영될 수 있다. 본 연구에서는 시골지역 당뇨병 환자들의 관리 실태 및 문제점을 알아보고자 하였다. 방법: 임실 지역 환자들 중 최근 6개월간 임실의료원에 내원한 107명의 당뇨병 환자들을 대상으로 혈당 및 혈압조절 정도를 조사하였다, 또한 신경병증 및 망막병증 등을 포함한 만성합병증에 대한 검사여부 및 유병률, 그 외 운동 및 식이 조절행태에 대해 알아보았다. 결과: 대상환자들의 평균연령 및 당뇨병 이환 기관은 각각 63.7±8.9, 6.8±3.7년이었고 공 복시 평균혈당은 128.8±41.7mg/dl, 식후2시간 평균혈당은 233.7±73.9mg/dl, 평균 당화혈색소는 7.3±0.7%로 조사되었다. 고혈압이 동반된 환자는 58.9%, 단백뇨 및 신경병증증상을 보인 환자는 각각 23.4%와 30.8%였다. 75.7%에 달하는 많은 환자들은 한번도 망막병증에 대한 안과적 검진을 받은 적이 없었다. 그 외 운동 및 식이조절을 시행하고 있는 환자들은 각각 43%와 34.6%로 낮게 조사되었다. 결론: 시골지역 당뇨병 환자들의 관리에 있어서 혈당 및 혈압 조절 외에 소홀하게 여겨지고 있는 만성병증의 교육, 예방 및 치료를 위해 좀 더 적극적인 정책적 변화가 필요하리라 사료된다. Background: In the management of diabetic patient, the prevention and the treatment of chronic complications are strongly related with prognosis. But management status of chronic complications is affected by age, living area and economic condition of patients, so it is possible that the patients living in rural district might be managed carelessly. Therefore, this study was performed to estimate the management status and problems in caring of diabetic patients in the country. Methods: We studied 107 diabetic patients who visited Im-Sil public Medical Center during the period from September 2004 to March 2005 in Im-Sil district. We investigated blood glucose level, blood pressure, the prevalence of the chronic complications and whether the evaluations of chronic complications including peripheral neuropathy and retinopathy were done. Results: The average age was 63.7±8.9yrs and average disease duration was 6.8±3.7yrs. The mean fasting and 2-hr blood glucose level were 128.8±41.7mg/dl and 233.7±73.9mg/dl, respectively. The mean HbAlc level was 7.3±0.7% and 44.7% of the patients had < 7.0% of HbAlc level. Hypertension, proteinuria, peripheral neuropathy and retinopathy were combined 58.9%, 23.4%, 30.8% and 1.9%, respectively. Exercise trial and diet control were performed only 43% and 34.6% of the patients, respectively. Conclusion: We suggest that positive medical systems for diabetic patients living in the country should be prepared for prevention and treatment of chronic complications.

      • Verification of autophagy in porcine oocytes and investigation of its roles

        Autophagy is well-conserved cellular recycling process. However, autophagy in in-vitro porcine oocyte maturation have not been discovered yet. In this study, we confirmed autophagosome in-vitro culturing porcine oocytes by detection of LC3 which is promised marker protein of autophagosome. Then, we also examine the relationship between autophagy and oocyte maturation by treatment of autophagic inhibitors or nuclear maturation inhibitors. Autophagy activation was regarded to amount of autophagosome in this study. To autophagosome detection in porcine oocytes, Immunochemistry and westernblotting using LC3 antibody was conducted by culturing time of 0 h, 14 h, 28 h, and 42 h. Then, we measured LC3-II levels using western blotting after inhibiting nuclear maturation of oocytes via cAMP treatment in an in vitro culture, to clarify whether nuclear maturation affects autophagy. To clarify whether autophagy affects nuclear maturation, we also counted mature oocytes after inhibiting autophagy by treating with wortmannin or a mixture of E64d and pepstatin A. Both groups of cAMP, which were applied for different treatment times, showed the same levels of LC3-II, while their maturation rate was approximately four times higher in 22h treatment than that of the cAMP 42h treatment group. This indicated that neither cAMP nor nuclear status affected autophagy. Autophagy inhibition during in vitro maturation with wortmannin treatment decreased oocyte maturation rates by approximately half, but autophagy inhibition in the E64d/pepstatin A mixture treatment did not significantly affect the oocyte maturation rate. Thus, wortmannin itself, or the autophagy induction step, but not the degradation step, is involved in the oocyte maturation of porcine oocytes. Overall, we confirmed autophagic dynamic during porcine in-vitro maturation. Moreover, we suggest that oocyte maturation does not exist upstream of autophagy, but autophagy may exist upstream of oocyte maturation.
