      • 가령에 따른 백서의 하악와 섬유층 세포내 vimentin 면역양성 중간세사의 출현

        백서에 있어서 하악와에 가해지는 기계적 부하의 증가와 관련하여 나타나는 조직의 반웅중 특히 하악와 섬유층의 섬유모세포의 세포질내에 다량으로 나타나는 중간 세사가 무엇인지, 또한 이 중간 세사의 출현이 가령 및 기계적 부하의 증가와 상관관계가 있는가를 규명하기 위하여 백서를 생후 14일, 28일 55일, 성쥐(체중 300g 이상)군으로 나누어 악관절 부위를 적출하여 면역염색을 실시한 뒤 광학 현미경을 이용하여 관찰하였으며 그 결과는 다음과 같다. 생후 14일군에서 vimentin 면역양성반응을 나타내는 세포는 전혀 관찰되지 않았다. 생후 28일군에서는 하악과 관절면의 섬유층에서 vimentin 면역양성반응을 나타내는 세포를 다수 관찰할 수 있었다. 특히 하악와 관절면의 중심부에서 많이 관찰되었으며 모양은 섬유모세포와 비슷하게 방추형으로 나타났고 핵의 주위에서 반응이 관찰되었다, 55일군에서는 하악와 관절면의 섬유층, 특히 하악와의 중간 부위에서 다량의 vimentin 면역 양성 세포를 관찰할 수 있었으며 이들 세포들은 방추형을 나타내어 섬유모세포와 유사한 양상을 보였다. 면역양성반응은 주로 핵주의의 세포질에서 핵을 둘러 싸고 있는 양상으로 나타났다. 성쥐군의 경우에도 하악과 관절면의 섬유층에서 다량의 vimentin 면역 양성 세포를 관찰할 수 있었으며 하악와의 중간부위에서 반응이 잘 나타났고 반응은 핵 주위의 세포질에서 관찰되는 등 대체로 55일군과 비슷한 양상을 보였다. 본 실험의 결과를 통해 가령에 따라 또한 incision 및 저작에 따른 기계적 부하의 증가와 관련하여 백서의 하악와에서 vimentin 면역양성 세포가 나타난다는 것을 알 수 있다. The purpose of this study was to investigate what the intermediate filaments in the cells of rat mandibular fossa fibrous fibrous layer are and any relationships between the presence of these filaments and aging. Mandibular fossae of 4 groups of rats(14-day, 28-day, 55-day and adult groups) were removed en bloc and processed for immunostaining and were subjected to light microscopic examination. The results were as follows: In 14-day group, there were no immunoreactive cells in fibrous layer of mandibular condyle articular surface. But in 28-day group, many immunoreactive cells were seen in fibrous layer, especially central portion of articular surface of mandibular fossa. These cells were fusiform shaped and immunoreactivities were seen in the cytoplasm around the nucleus. In 55-day group many immunoreactive cells were seen in fibrous layer of mandibular fossa. These cells were fusiform shaped and distributed evenly in central portion of this fibrous layer. Immunoreactivities were sseen in the cytoplasm around the nucleus. In adult group, the results were similar to 55-day group. Many immunoreactive cells were seen in fibrous layer of mandibular fossa especially central portion. According to these results, vimentin immunoreactive filaments appear with aging and increment of mechanical load associated with incision or mastication.

        하악골 상행지의 수직 골절단술과 시상분할 골절단술을 이용한 하악골 전돌증의 외과적 치험례

        This is a case report of mandibular prognathism corrected by orthognathic surgery with pre-and post op. orthodontic treatment. The patient was 17years old with severe skeletal class Ⅲ malocclusion (<ANB, -6.5, Mx-Mn differential, 53mm) and mild mandibular asymmetry due to overdeveloped mandible. After the precise clinical, cephalometric and model analysis, the patient was treated with vertical ramus osteotomy on Rt. side, intraoral sagittal spilt ramus osteotomy on Lt. side and the treatment results analyzed radiographically and clinically. The results were as follows; The mandible was moved backward 8mm on Lt. side, 11mm on Rt. side at occlusal plane level and it was produced marked profile change. The preop. and postop. orthodontic treatment improved the esthetic and functional results of orthodontic surgery.

        가령에 따른 백서의 하악와 섬유층 세포내 Vimentin 면역양성 중간세사의 출현

        협부에 전이된 지방육종의 치험례

        지방육종은 주로 근간 간엽조직으로부터 발생되어지며, 중년이상의 남성에서 호발하고 주된 발병부위는 하지 및 후복막강이고 구강 주위조직에 이환되는 경우는 극히 드문 악성종양이며, 본 증례에서는 4년 전 좌측 하지에서 발생되어 우측 하지 및 좌측 협부에 전이된 것으로 생각되어지는 점액양의 지방육종으로 종물의 외과적 절제술 및 화학요법을 시행하였고 장기적인 관찰이 요구된다. Liposarcoma is one of the most common malignant mesenchymal neoplasm, comprising approximately 15% of all soft tissue sarcoma. This is a tumor with an incidence peak between age 40 and 60 years, and is slightly more common in men than women. Although all body region may be involved, the most frequent sites are lower extremities and retroperitoneum but rare in the head and neck region. Liposarcoma can be classified to four subtypes ; myxoid, well-differentiated, round-cell, pleomorphic. The myxoid type is the most common and accounts for almost one half of all liposarcoma. Wide surgical excision with or without radiation therapy has been used to manage this lesion. We report a case of 50-year old man with soft tissue swelling on the left cheek, previously diagnosed as myxoid liposarcoma in the left lower extremity. After radiologic and ultrasonic study and surgical excision with biopsy, the lesion of cheek was diagnosed as myxoid liposarcoma with round cell differentiation.

        화학적 발암화에 따른 Protein Kinase C의 발현 변화

        Protein kinase C (PKC) is known to play a pivotal role in neoplastic transformation cells and its high expression is often found in a variety of types of tumors including oral cancer. While PKC is associated with the altered signal transduction pathway of the tumor cells, it is still unclear which isoform is involved in the carcinogenesis process. Since the cellular distributions and the roles of PKC are isoform-specific, it is very important to identify the specific target molecules to improve our understanding of the carcinogenesis processes. Thus, the present study attempted to perform chemical carcinogen-induced neoplastic transformation of human epithelial cells and analyze the specific isoform of PKCs involved in the cellular transformation. The study analyzed overall PKC responses upon MNNG(N-Methyl-N'-nitro-N-nitroso guanidine) exposure with [3H] PDBu binding assay. PKC translocation was observed at high doses of MNNG treatment in the presence of extracellular calcium. Such effects were not observed in the absence of extracellular calcium. Translocational effects with exposure of MNNG was further enhanced in the presence of hydrocortisone. The result suggests that the type of PKC involved may be Ca2+-dependent classical isoform and steroid hormone enhances PKC activation. Among cPKC isoforms examined, only PKC-α and r showed significant translocation of protein levels from cytosolic fraction to membrane fraction, as analyzed by immunoblot. PKC-ε in nPKC class showed an inch·eased translocation, but other forms in this class did not show the effect. None of isoforms in aPKC class was affected by MNNG treat-ment. The study demonstrated that there was a certain specificity in the patterns of isoform induc-tion follwong chemical carcinogen exposure and helped identify all the types of PKC isoforms expressed in human epithelial cells. It was revealed that PKC isoforms were activated in an early resonse to chemical carcinogen, suggesting that PKC be associated with carcinogenesis process from an early stage in this particular cell system. The study will contribute to improving our understanding of chemical-induced carcinogenesis in human cells and may provide a scientific basis to introduce the specific PKC inhibitors as an anticancer drug of epithelial cell-origin cances including oral cancer.
