[PROTOCOL] Structural analysis of N-/O-glycans assembled on proteins in yeasts

Protein glycosylation, the most universal and diverse posttranslational modification, can affect protein secretion, stability, and immunogenicity. The structures of glycans attached to proteins are quite diverse among different organisms and even within yeast species. In yeast, protein glycosylation plays key roles in the quality control of secretory proteins, and particularly in maintaining cell wall integrity. Moreover, in pathogenic yeasts, glycans assembled on cell-surface glycoproteins can mediate their interactions with host cells. Thus, a comprehensive understanding of protein glycosylation in various yeast species and defining glycan structure characteristics can provide useful information for their biotechnological and clinical implications. Yeast-specific glycans are a target for glyco-engineering; implementing human-type glycosylation pathways in yeast can aid the production of recombinant glycoproteins with therapeutic potential. The virulenceassociated glycans of pathogenic yeasts could be exploited as novel targets for antifungal agents. Nowadays, several glycomics techniques facilitate the generation of species- and strain-specific glycome profiles and the delineation of modified glycan structures in mutant and engineered yeast cells. Here, we present the protocols employed in our laboratory to investigate the N- and O-glycan chains released from purified glycoproteins or cell wall mannoproteins in several yeast species.
The effect of social network sports community consciousness on sports attitude

The purpose of this study is to determine the impact of social network sports community consciousness onloyalty and sports attitude. In order to achieve this research purpose, the population of the study was selectedas adult men and women over the age of 20 who are active in the social network sports community in 2022. The sampling method used cluster random sampling to select a total of 300 people, 150 men and 150 women,as research subjects. The survey tool used was the questionnaire method, and the questionnaire whosereliability and validity had been verified in previous studies at home and abroad was used by requoting,modifying, or supplementing it to suit the purpose of this study. It was also structured on a 5-point scale. Frequency analysis, factor analysis, reliability analysis, simple regression analysis, and multiple regressionanalysis were performed on the collected data using the statistical program SPSS Windows 20.0 Version. Theresults obtained through this process are as follows. First, social network sports community consciousnesswas found to have a partial effect on loyalty. Second, social network sports community consciousness wasfound to have a partial effect on sports attitudes. Third, social network sports community loyalty was found tohave a partial effect on sports attitudes. Considering these results, various activities such as decision-makingprocess, relationship formation, and opinion expression of modern people are carried out by the O-linecommunity. In addition, while in the past it was a format that led from offline activities to online activities,currently, there are more and more formats that lead from online activities to offline activities. Therefore,modern people's SNS sports community activities provide many experiences, which creates a sense ofcommunity and sports attitudes are formed based on this. This can be said to lead to loyal activities.

The Purpose of this study to investigate the relationship of sports activity participation to subjective euphoria of office workers. To achieve this purpose, the subjects were selected by cluster random sampling of adults, in Daejon in 2002 at present. There were 273 male, 215 female used for the analysis. The total was 489. Data were collected by employing a questionnaire. The variables of this analysis were demographic characteristics, sports activity participation, subjective happiness(positive affect scale and negative affect scale). The questionnaire for Life sathisfaction had been developed by Diener. Emmons, Lersen & Griffin(1992) and standardized in Korean language by Lee Jong kil(1992). The questionnaire for Positive and Negative Affectby Watson, Clark & Tellgen(1988) and standardized in Korean language by Lee Ju Sung(1991). The collected data analyzed statistic program SPSS Windows 10.0 Version. The statistical analysis are descriptive statistics analysis, multiple regression analysis and analysis of covariance. From the analyses of the data, this study reached he following conclusions: First, subjective euphoria is partially different by sports activity participation among employees. Second, degree of sports activity participation is positively influence on subjective euphoria.

Understanding the characteristics and regulation mechanisms of cell wall integrity (CWI) in yeast is important not only for basic research but also in biotechnological applications. We found significantly different CWIs in two representative strains of the thermotolerant methylotrophic yeast Hansenula polymorpha. Compared to the A16 strain (classified as Ogataea polymorpha), the DL1-L strain (classified as Ogataea parapolymorpha) has a thinner cell wall that was found to be more fragile following long-term cultivation and more sensitive to zymolyase. To gain a deeper insight into this difference, we compared the characteristics of the Mpk1pmediated CWI signaling pathway in the two strains. While a DL1-L mutant deficient in Mpk1p (mpk1Δ) showed severe growth retardation at both normal and high growth temperatures and in the presence of cell-wall disrupting agents, the A16 mpk1Δ mutant displayed only a mild defect in cell growth. Sorbitol effect on rescuing growth retardation was different in the two mpk1Δ strains, which could partly be ascribed to subtle differences in the activation of HOG pathway. Among the cell wall disruptors evaluated, only caffeine clearly increased phosphorylation of Mpk1p in DL1-L, but not in A16. A transcriptome analysis of the DL1-L strain revealed that caffeine significantly increased the expression of a subset of cell-wall related genes in an Mpk1p-dependent manner, but not the expected Rlm1-target genes. Taken together, our data support an essential role for Mpk1p in maintaining CWI in H. polymorpha, although the requirement for Mpk1p and its regulation under diverse stress conditions varies depending on the strain background.

In 2016 and 2017, as part of a comprehensive investigation to identify the prokaryotic species in Korea, a total of 12 bacterial strains were isolated from the gastrointestinal tract and/or fecal samples of four endangered species, including reptile, bird, and marine and terrestrial mammals. Phylogenetic analysis with the 16S rRNA gene sequence was used to assign these strains to the phyla, Firmicutes, Actinobacteria or Proteobacteria. Furthermore, most of the strains Firmicutes belonged to the order Lactobacillales. Interestingly, 12 of the isolated strains have not been previously reported from the Korean Peninsula. Also, based on their high 16S rRNA gene sequence similarities (>98.7%) and formation of strong monophyletic clades with the closest type species, each isolated strain of isolates was assigned to an independent, predefined bacterial species. Gramstain reaction, colony and cell morphology, biochemical characteristics, isolation source, and NIBR IDs are described in the species description section.
