      • 당근 절편을 이용한 Agrobacterium spp. 분리, 동정과 선발균주에 의한 연초의 형질전환

        These studies were carred out to obtain the transformant from tobacco cells by Agrobacterium spp. from crown gall and soil at the natural field in Korea, and identified their virulence. Kodo's and Clark's selective media were used for isolation of Agrobacterium spp. In these media, total of 99 strains were characterized based on the morphological characteristics of colonies. Among them 34 strains were able to induce on carrot discs. And hypervirulent strains C23-1 and K29-1 were identified as Agrobacterium tumefaciens biotype 1 and biotype 2, respectively. These strains formed fast growing, larger gall as compared to those induced by other strains on the carrot discs. Transformed tobacco callus was initiated on the phytohormone free MS medium with 250$\mu\textrm{g}$/ml carbenicillin after co-cultivation of tobacco stem explants and Agrobacteria. On the phytohormone free media, shoot was rarely formed from transformed callus. However, these shoot were teratoma shoots which were not grown as normal shoot, and teratoma shoot from transformant by C23-1 was smaller than that of K29-1.

        栽植位置 年生에 따른 人蔘의 形質變異

        This study was carried out to obtain the basic information for development of ginseng varieties. Ginseng plants of one to five years were used for this study. Agronomic characters, such as stem diameter, stem length, stem weight, leaf length, leaf width, leaf weight, petiole length, number of leaves, number of leaflets, root length, main root length, root diameter, and root weight were determined. All of the characters of two-year-old plant showed low variability as compared with those of three-, four-and five-year-old plants. On the variations of characters according to the planting position, stem diameter, number of leaves, number of leaflets, root length, and main root length were found to be low variable at all of the plant ages, while the other characters were highly variable in three-, four-, and five-year-old plants. Generally, the growth of aerial portion of ginseng plants at above three years planted on the 1st and 5th lines were poorer than that of the 2nd, 3rd, and 4th lines, while the root growth of ginseng plants planted on the 1st and 2nd lines was better than that of the 3rd, 4th, and 5th lines. Root weight and root diameter related to yield were increased with the increase of plant age, but the length of main root was decreased.

      • 연초 Crown Gall Callus유래 Teratoma Shoot의 생육과 발근에 미치는 식물호르몬과 활성탄의 영향

        These studies were conducted to investigate effects of phytohormone and activated carbon on the growth and rooting of teratoma shoots induced from Nicotiana tabacum cv. NC2326 transformed by Aerobacterium tumefaciens C58. GA was effective for shoot elongation and reduction of multiple shoots from teratoma shoot, however, leaves of teratoma shoot cultured on the medium with GA were pointed. ABA was also effective in promoting shoot elongation, but was not for reduction of multiple shoots. Teratoma shoot cultured on the medium with 1 n activated carbon promoted shoot elongation and inhibited the number of shoots differentiated, but was grown as abnormal shoot. Addition of 1% activated carbon and 0.5mg/l BA to culture media was effective for shoot elongation and reduction of multiple shoot and for formation of round leaves as normal leaves. Though these shoots were inoculated on the rooting medium, they could not from roots but formed multiple shoots. Boric acid, myo-inositol and sucrose were also ineffective on the rooting of teratoma shoots.

        栽培條件을 달리한 水稻變異系統의 農耕形質의 變異

        Present studies were carried out to clarify the variation of agronomic characters of rice mutant lines when grown under different cultivation conditions. In general the variations of culm length, panicle length, number of days to heading, number of panicles per ㎡, number of grains per panicle, and weight of 1,000 grains of mutant lines were found to be more variable as compared with those of mother variety. In mutant lines as well as in the mother variety, the variability of panicle number per unit area and grain number per panicle was high, while the other agronomic characters showed comparatively low variability. No correlation was found among various agronomic traits when grown under conditions above-mentioned.

        水稻高蛋白突然變異系統 No. 398에 關한 硏究

        High protein mutant No.398 (M_9 in 1973) was obtained from X-rayed Hokwang variety, and their grain characters were examined. The protein content of mutant No. 398 was increased 30% than that of mother variety. Besides the protein content, heading date was 25 days earlier and culm length was shortened 20㎝. Grain size and shape, weights of embryo and endosperm were similar to the control variety, while protein content of embryo of high protein mutant line was higher. Although the protein amount of embryo in the mutant was higher, it is negligible because in rice the endosperm was more than 35 times heavier than the embryo. The increase of protein in No.398 is attributable to the increment of protein in the endosperm. The mutant has more protein than mother line in the peripheral starch layer of endosperm. In developing grain the difference of protein amount per grain in mutant line was outstanding from three weeks after heading.

        放射線에 의한 水稻의 蛋白質育種

        The studies were carried out on change of protein content of high protein mutants under different cultivation conditions, correlation between protein content and agronomic characters of mutant lines, and other related works. As for the influence of sowing date, mutants behaved variously showing no definite tendency, and variation was wider than that of the mother varieties. In the mutants and their mother varieties alike, the tendency of variation of protein under different planting spaces was uniform, and the variation was not large in mutants as well as in their mother lines. Among the high protein mutant lines grown under different cultivation conditions, no correlations were found between protein percent and panicle number per unit area, between protein percent and grain size, and between grain size and protein amount per grain. In relation between protein content and some agronomic traits, there were correlations between culm length and protein, grain weight and protein content, culm length and grain weight, and between grain weight and protein per seed. High protein mutants were attributable to the corresponding increase of glutelin fraction which accounted for more than 70% of protein. Usually high protein mutant lines had lower basic amino acids per gram protein than low protein lines when estimated by DBC.
