        국내산 개암(Corylus heterophylla)의 항산화 활성 및 세포 보호 효과

        This study investigated the antioxidant activity and cellular protective effect of domestic cultivars of hazelnut (Corylus heterophylla) including Dureoksil (DRS), Yeomun (YM), HV, and an imported cultivar from Turkiye (Corylus avellana, CA). The DRS and HV cultivars showed the highest total phenolic content whereas the YM cultivar exhibited the highest 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activity. There were significant differences in the levels of kaempferol-3-O-rutinoside, (-)-epicatechin-3-O-gallate, quercetin-3-O-glucoside, and proanthocyanidin tetramers among the cultivars. The YM cultivar showed higher concentrations of bioactive compounds than other cultivars. All cultivars increased cell viability against H2O2-induced cytotoxicity in HT22, A549 and HT-29 cells, according to the 3-(4,5-dimethyl-thiazol-2-yl)-2,5-diphenyl tetrazolium bromide assay. Results of the 2',7'-dichlorofluorescein diacetate assay revealed that the YM and HV cultivars reduced the H2O2-induced formation of reactive oxygen species in HT22, A549 and HT-29 cells. These results suggest that domestic hazelnut could be used as a functional food material that inhibits oxidative stress.

        고당 및 올레산으로 유도된 HepG2 세포에서 국내산 호두(Juglans regia) 추출물의 비알코올성 지방간 질환 개선 효과

        This study was investigated to evaluate the in vitro anti-diabetic activity and lipid accumulation inhibitory effect of the domestic walnut (Junglans regia, Gimcheon 1ho cultivar) in high glucose- and oleic acid-induced hepatic HepG2 cells. Eighty percent of ethanolic extract from Gimcheon 1ho (GC) showed higher inhibitory effects against α-amylase and α-glucosidase activities when compared to other ethanolic extracts. GC significantly inhibited the formation of advanced glycation end products (AGEs). The GC increased cell viability against H2O2- and high glucose-induced cell death determined by 3-(4,5-dimethyl-thiazol-2-yl)-2,5- diphenyl tetrazolium bromide (MTT) assay. GC reduced the formation of reactive oxygen species (ROS) in H2O2- and high glucose-induced HepG2 cells determined by 2',7'-dichlorofluorescein diacetate (DCF-DA) assay. GC inhibited lipid accumulation in the oleic acid-induced HepG2 cells. The GC increased AMP-activated protein kinase (AMPK) phosphorylation and decreased the expression of lipid metabolism related proteins such as sterol regulatory element-binding proteins 1 (SREBP-1), fatty acid synthase (FAS), sterol regulatory element-binding protein 2 (SREBP-2) and 3-hydroxy-3-methyl-glutaryl-coenzyme A (HMGCR). The results suggest GC is a potentially functional food material which can improve nonalcoholic fatty liver disease by inhibiting lipid accumulation.

        산마늘 추출물의 알코올 유도 세포독성에 대한 간 세포 보호 효과

        This study aimed to evaluate the antioxidant and hepatoprotective effects of aqueous and 60% ethanol extracts of Allium ochotense Prokh. against alcohol- induced cytotoxicity as well as on the activities of alcohol-metabolic enzymes. Antioxidant effects of the extracts were analyzed using 3-ethylbenzothiazoline- 6-sulfonic acid, 1,1-diphenyl-2-picrylhydrazl, ferric reducing antioxidant power, and malondialdehyde assays, and found that both extracts exhibited considerable antioxidant activities. Additionally, both extracts showed synergistic effects on the activities of alcohol-metabolic enzymes, such as alcohol dehydrogenase, but not on the activity of aldehyde dehydrogenase. In addition, 2'-7'-dichlorodihydrofluorescein diacetate (DCF-DA) and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assays revealed that aqueous and 60% ethanol extracts reduced oxidative stress and increased cell viability. Moreover, both extracts regulated the expression of apoptosis-related proteins, namely B-cell lymphoma (BCl-2), BCl-2 associated X (BAX), and pro-caspase-3, in HepG2 cells. In conclusion, aqueous and 60% ethanol extracts of A. ochotense Prokh. might be valuable functional materials derived from natural resources for the prevention of ethanol-induced cytotoxicity.

        미세먼지로 유도된 산화적 스트레스에 대한 국내산 호두(Juglans regia) 아세트산 에틸 분획물의 뇌 해마세포 보호 효과

        본 연구에서는 국내산 호두 아세트산 에틸 분획물에 대한 생리활성 평가를 위해 총 폴리페놀 화합물, 총 플라보노이드 화합물, 총 탄닌 화합물, ABTS와 DPPH 라디칼 소거 활성, 철의 환원 능력, 지질과산화물 생성 억제 능력, AChE의 활성 억제 효과를 확인하였다. 특히 EFGC의 철의 환원 능력과 지질과산화물 생성 억제 능력은 양성대조군으로 사용된 비타민 C, 카테킨보다 높은 활성을 나타내었다. HT22 세포에서 PM2.5로 유도되는 세포독성에 대한 세포 생존율과 산화적 스트레스 생성 억제 능력을 평가해 보았을 때 우수한 세포보호 효과를 나타내었다. 또한, EFGC는 caspase-1, BAX, p-JNK, BCl-2 및 p-NF-κB와 같은 염증성 인자와 세포사멸 관련 인자의 발현을 개선함으로써 PM2.5로 유발되는 세포독성에 대한 보호 효과를 확인하였다. 따라서 본 실험에서는 EFGC가 PM2.5로 유도된 뇌 신경세포 독성에 대하여 도움을 줄 수 있는 건강기능식품 소재로서의 산업적인 가치를 확인하였다.

        떫은감 품종별 가공 형태에 따른 영양성분 분석

        This study was performed to analyze the nutritional components of fresh Diospyros kaki fruits of four persimmon cultivars (Bansi, Gabjubaekmok, Gojongsi, and Dungsi) processed in different ways. Fructose and glucose contents were higher in processed products than in fresh fruits. Sliced-dried persimmon had higher levels of total free sugar, palmitic acid, stearic acid, α-linolenic acid, and linoleic acid than other processed products. All Gojongsi products processed had elevated levels of unsaturated fatty acids. In the results of amino acid analysis, glutamic acid and aspartic acid were highest in cultivars. Sliced-dried persimmon had a higher essential amino acid content than other fresh fruits and processed products, and vitamin C contents were higher in all processed products than in fresh fruit. β-Carotene content was greatest for sliced-dried Gabjubaekmok. γ-Tocopherol and total tocopherol contents were higher in processed products than in fresh fruit, and sliced-dried Gabjubaekmok had a higher total tocopherol content than the other cultivars. Myricetin, 3′-hydroxy-5,6,7,8,4′-pentamethoxyflavone, 6,8-di-C-methylkaempferol 3,7-dimethyl ether, citric acid, malic acid, and gallic acid were detected as major physiologically active compounds by ultra-performance liquid chromatography/quadrupole time-of-flight tandem mass spectrometry (UPLC/Q-TOF/MS).
