      • LED광을 이용한 눈꽃동충하초 생산

        최근 다양한 파장의 LED를 이용한 작물의 생장과 기능성 물질의 변화에 대한 연구가 활발히 진행되고 있다. 본 연구에서는 LED 조사가 눈꽃동충하초 생산에 미치는 영향을 알아보고자, 청색LED, 적색LED, 형광등 아래에서 눈꽃동충하초를 재배하여, 자실체 형성율, 자실체 개수, 자실체 길이 등을 관찰하였다. 그 결과, 적색 LED 광에서 재배한 눈꽃동충하초의 자실체 발생율은 100%에 달하였으며, 뿐만 아니라 형성된 자실체의 개수가 37%까지 증가하는 등 자실체 무게, 자실체 길이 등 모든 항목에서 우수한 결과를 보여주었다. 하지만 청색 LED광에서는 오히려 생육을 저해하는 결과를 나타내었다. 따라서, 눈꽃동충하초 재배 시, 적색 LED를 조사한다면 농가의 소득 향상에 도움을 줄 것으로 여겨진다. The effects of LED irradiation on growth and functional components of crops are actively being studied. We investigated the effects of red LED on the cordyceps cultivation because Cordyceps is very difficult to find in the wild and to cultivation. The synnemata formation rate of P. tenuipes was 100% under red LED. Also weight, length, and number of the synnemata were significantly increased. We expect that the red LED treatment will be of help to improve farm income.

      • 인체적용을 위한 실크 스펀지의 제조 및 특성

        Fibroin and sericin are natural proteins obtained from cocoon and one of the spotlight materials for medical device. Medical device made of these proteins also has the advantage that this material is biodegradable in to amino acid. In this study, we prepared silk sponges using fibroin, sericin and additives. The characterizations of the silk sponges such as morphology, stability, and blood absorbency were observed. The structural stability of the silk sponge decreased significantly by increasing sericin contents. The effect on the concentrations of ethanol to induce crystallization was observed to be superior to 70% ethanol. Structural stability of silk sponges containing additives was very lower than those not containing additives. The blood absorbency of the silk sponges was found to be excellent, regardless of the composition of sericin and fibroin. The resilient power of these sponges was also very good, in spite of the repeated soaking and drying. Therefore, we expect that the silk sponges can be used medical supplies such as plastic implants and hemostatic cotton.

      • 누에 품종에 따른 눈꽃동충하초 배양 특성

        동충하초는 곤충의 몸속에 들어가 버섯이 발생하는 곤충 기생성 약용 버섯으로 세계적으로 인체에 유용한 활성물질이 확인되었으며, 이를 누에를 이용하여 인공재배할 수 있는 기술이 개발되어 활용되고 있다. 그러나 누에 품종에 따른 동충하초의 생산성에 대한 연구 보고는 없는 실정이다. 본 연구에서는 장려품종 3품종(백옥잠, 금옥잠, 대성잠)과 특수품종 2품종(연록잠, 골든실크)을 이용하여 눈꽃동충하초의 재배조건과 특징을 조사하였다. 눈꽃동충하초 균주(P. tenuipes)에 대한 감염율은 금옥잠이 가장 높았으며, 백옥잠, 대성잠, 골든실크, 연록잠 순으로 감염율이 높았다. 눈꽃동충하초를 재배하기 위한 최적조건은 어두운 장소에서 번데기 당 약 4 cm2의 재배밀도로 22oC ± 1를 유지해 주는 것이었다. 또한 우리는 농가소득 향상을 위해, 동충하초의 수요가 있을 때 마다 수시로 재배할 수 있는 연중재배 기술을 개발하였다. 연중재배 방법은 먼저 동충하초균에 감염된 번데기 체내의 수분함량을 자연건조로 3% 이하로 낮추고, 이를 진공포장 후 4oC 이하에서 보관하는 3단계의 과정이 필수적이다. 이렇게 저온에 보관되어 있는 감염번데기는 언제든지 물에 침지하여 번데기 내의 함수율을 원래대로 복원 시킨 다음 최적 재배조건에서 재배하여 동충하초를 생산할 수 있게 함으로 연중생산이 가능하다. 그러므로 우리는 동충하초 재배농가가 누에 품종 중 금옥잠을 이용하고 연중생산 기술을 활용하여 소득증대를 이룰 수 있을 것으로 기대한다. There is no specific data on Entomogenous fungus (DongChungHaCho) in accordance with the silkworm varieties, despite of very high value industrial use as functional materials at domestic and abroad. In this study, we investigated culture condition and characteristics of Paecilomyces tenuipes in 3 silkworm cultivars and 2 specific silkworm breeds. Infection rate of P. tenuipes for silkworm varieties was the highest Kumokjam, followed by Baegokjam, Daeseongjam, Golden silk, Yeonnokjam in that order. The optimum culture conditions were as follows: dark condition, 22oC ± 1, and about 4 cm2 in planting density per pupa. Also, we have developed annual production technologies of P. tenuipes. First, in order that the moisture content of the infection pupa became 3% or less in the natural condition. Second, the dried pupa must be vacuum-packed and was kept under 4oC. Finally, by restoring the moisture content of the dried pupa and culture in optimal condition, the technique for annual production of the P. tenuipes was achieved. Therefore, we expect that the use of Kumokjam and annual production technique will contribute significantly to farm income.

      • 실크피브로인 용해조건에 따른 생체막의 물성 변화

        Silk is a natural polymer that has the advantages of the biocompatibility, excellent mechanical strength, low immune rejection, and molding facility. But silk does not dissolve easily in water or general solvent. To investigate the characteristics of silk biological membranes according to dissolving condition of silk fibroin, we made the silk biological membranes using silk fibroin solutions with different amount and dissolving time of silk. The characterizations of the silk biological membranes such as morphology, structure, and mechanical strength were observed. Although each biological membrane has the same fibroin content, there was a significant difference in the thickness and transparency. But there was no significant change in the molecular weight of the silk fibroin solutions and morphology of silk biological membranes. We were established the manufacturing condition for silk fibroin biological membrane. So we expect that the conditions will help in the development of medical supplies in the future.

        Silk fibroin is a natural biomaterial that has the biocompatibility and other many advantages. But as a silk fibroin membrane thickness increases, the transparency becomes more opaque. Because the transparency of membranes tissue such as the cornea and dura mater are necessary, transparent membrane is required to replace these transparent membranes. In this study, we fabricated blending silk fibroin membranes that made by mixing the various inorganic salts or polymer in an aqueous solution of silk fibroin. The transparency of the membranes were analyzed. the transparency of these membranes is very different, depending on the mixed materials. Inorganic salts mixed silk membrane was more transparent than the polymer mixed one. Especially, the silk fibroin membrane with calcium chloride was very transparent. We showed the possibility of blending silk fibroin membrane, which can be used in perfect transparent membrane such as the cornea. In the future, we expect that the transparent blending silk fibroin membrane can be used in various medical applications.

        Silk fibroin, a natural protein produced by silkworm, is a good biomaterial which has biodegradability and biocompatibility. To ascertain the effects of silk fibroin on cell growth, silk fibroin films were prepared using silk fibroin aqueous solutions of various concentrations. We investigated the attachment, proliferation, morphology of the cells and the expression levels of genes related to cell attachment and growth on the silk fibroin films. When the cells were cultured on the 0.1 and 1% silk fibroin film, the cell adhesion ability was very excellent. Particularly, overall cell growth on the 1% silk fibroin film was definitely superior to the others. Also, expression levels of genes related cell growth were increased on the 0.1 and 1% silk fibroin film. These results suggest silk as a material for medical applications.

        A number of researcher have studied biomaterials for cartilage regeneration and are now proceeding. Silk protein was attempted for use as biomedical materials by many researchers because it is natural polymer with biocompatibility and excellent mechanical strength. In this study, we want to know a possibility of silk protein on the cartilage regeneration. We isolated chondrocytes from nasal cartilage and confirmed optimal culture condition of the cells. To observe the effects of silk fibroin on chondrogenesis, we added silk fibroin solutions to the culture medium of chondrocyte and detected gene expression levels related chondrogenesis such as col2, col10. The chondrocytes showed optimal growth when they were cultured in DMEM medium supplemented with 10% FBS 100 ·?M ascorbic acid. The levels of col2 gene expression were increased in non-autoclaved silk fibroin, but decreased in autoclaved one. Also the gene expression levels of col10 were increased in silk fibroin, particulary at 3D culture. Based on the results of this study, we had seen the possibility of silk fibroin for cartilage regeneration. In future studies, we should know more clearly the relationship between cartilage regeneration and the silk protein.

        집단미술치료가 한부모 가정 아동의 자아존중감, 사회성과 부모표상 지각에 미치는 효과

        본 연구는 집단미술치료 활동이 한부모 가정 아동의 자아존중감, 사회성, 부모표상 지각에 미치는 효과를 연구한 것이다. 연구 대상으로 초등학생 16명을 실험집단과 통제집단으로 각각 8명씩 할당하였으며, 실험집단은 주 1회 1시간씩 14주 동안 16회기의 집단미술치료 활동에 참여하였고 자아존중감, 사회성, 부모표상 지각 등의 종속변수 측정이 집단미술치료 활동 사전과 사후에, 그리고 활동 종결 한 달 후에 실시되었다. 반면 통제집단은 사전, 사후, 추후 검사에만 참여하였고 특별한 처치를 받지 않았다. 연구 결과, 실험집단은 통제집단과 사전점수는 비슷한 수준이었으나 사후검사에서 통제집단 보다 자아존중감, 사회성, 부모표상 지각에서 유의한 점수의 증가를 보였으며 이러한 효과는 추후검사에서도 유지되었다. 또한 프로그램 초기와 종결기에 실시된 동적 집-나무-사람 그림검사(KHTP), 동적가족화(KFD), 학교생활화(KSD)의 질적인 변화를 분석한 결과, 크기와 내용과 상징에서 많은 변화를 보여 자아존중감, 사회성, 부모표상 지각이 긍정적으로 향상되었음을 나타내었다. 자신감과 성취감의 경험으로 자신을 가치 있는 사람으로 생각하고, 또래관계가 원만해지고 부모를 바라보는 시선이 긍정적으로 변화된 것으로 확인되었다. This study is designed to investigate the effects of group art therapy on self-esteem, sociability and the parent`s image perception of the children in single parent families. The subjects were 16 elementary school students, 8 of whom participated in 16 one-hour sessions of group art therapy over 14 weeks, while the remaining 8 students were allocated to the waiting-list control group with no treatment. The results showed that group art therapy significantly increased the levels of the children`s self-esteem, sociability, and parent`s image perception at the post- and follow-up tests. Besides, the qualitative data including projective picture drawings (KHTP, KFD, KSD) at the pre- and post-tests showed meaningful changes in the size, contents, and symbolic meanings of the drawings, which can be interpreted as positive changes in self-esteem, sociability and the parent`s image perception of the children. The qualitative data implied that the positive atmosphere and the interaction, recognition and support from group members helped them to feel self-confident and self-worthy, and to have a positive perspective towards their parents` image. The limitations and implications of the study were discussed.
