        다형연쇄구균이 유치우식발생에 미치는 영향

        The aim of this study was to clarify connection between S. mutans and caries pattern of deciduous dentition. For this purpose 330 6-years old children and 152 8-years old children were selected. The selected subjects were surveyed by calibrated examiner under natural light with dental mirror and explorer using the WHO criteria. The S, mutans level in the saliva was estimated using peptone solution rinsing method. According to S. mutans level examined children were classified into three groups - low(CFU=O), moderate (CFU= 1,000-50,000), high (CFU>50,000). The examined tooth surfaces were categorized into three patterns- fissure pattern, posterior buccal/lingual smooth pattern, posterior proximal pattern. The caries data were analyzed using "Caries Analysis System" proposed by Douglass. The result obtained were as follows; 1. The rate of caries positive children was 69% in low S. mutans level, 92% in moderate S. mutans level, 96% in high S. mutans level. 2. The rate of dfs significantly increased with S. mutans level. 3. The prevalence and severity in all patterns significantly increased with S. mutans level. 4. Distribution of fissure pattern significantly decreased with S. mutans level, but distribution of posterior proximal and posterior buccal/lingual smooth surface patterns significantly increased.

        The statistical observation on fat amount by various measurement methods. In 30 of them anthropometry, densitometry and skinfold thickness(triceps)measurement were made. I The total body fat was calculated predicted from the equation by Keys and Brozek's. As a result of this studies, the following conclusion were obtained: (j) The correlation coefficient of body weight and body volume was highly significant as r=+0.96 (p-0.001) and its induced regression equation predicted from Keys & Brozek's was shown as the following: Fat(kg)=0.111M+2.285 (Where M is body weight in kg) (2) The correlation cefficient among the body fat amount(kg) as determined by indirection method of volumetry and othe various methods which that skin-fold thickness and relative weight methods was r=+0.870, r=+0.459 and r=+0.974 in each method.

        쥐에서 Taurocholate 부하에 의한 간의 Glyoxalase I의 유도

        The possible mechanisms of increased glyoxalase I (GLO-I) activity in cholestatic rat liver and serum were studied. These hepatic and serum enzyme activities were determined from the experimental rats with choledocho-caval shunt (CCS) or bile duct obstruction (BDO). The Michaelis-Menten constants in this hepatic enzyme were also measured. The activities of hepatic and serum GLO-I as well as the Vmax value of this hepatic enzyme were found to be increased significantly in both CCS plus taurocholic acid (TCA) injected group, and BDO plus TCA injected group than in each control group such as CCS alone and BDO alone. In addition, these serum and hepatic enzyme activities did not change in both the CCS plus tauroursodeoxycholic acid injected group and the BDO plus tauroursodeoxycholic acid injected group. On the other hand, the value of Km of the above hepatic enzyme did not change in any experimental group. Above results suggest that TCA induce the biosynthesis of GLO-I in the liver. The elevated activity of the serum GLO-I is believed to be caused by the increment of membrane permeabiliry of hepatocytes upon TCA mediated liver cell necrosis.

      • Transformation of Potato (Solanum tuberosum L.) Tuber Cells with Agrobacterium tumefaciens and Ti Plasmid DNA

        본 연구에서는 Agrobacterium tumefaciens 및 Ti-plasmid에 의한 감자 괴경세포의 형질전환 여부를 조사하여 다음과 같은 결과를 얻었다. Agrobacterium tumefaciens KU-12의 감염에 의하여 감자 괴경의 원반에 tumor가 형성되었으며, 이 원반을 호르몬이 없는 배지에서 배양한 결과 tumor에서 완전한 식물체가 재생되었다. 또한 tumor에서 재생된 식물체 내에는 lysopine dehydrogenase(LpDH) 활성이 있음을 확인하였다. 감자의 원형질체에 Ti-plasmid DNA(pTi-12)를 처리하여 호르몬이 결여된 배지에서 배양한 결과, 165개의 callus를 얻었으며, 그 중 일부에서는 뿌리가 형성되었다. 뿌리를 형성한 callus와 형성하지 않은 callus을 임의로 선택하여 LpDH 활성을 조사한 결과 둔 경우 모두 octopine을 합성함으로 형질전환체임을 확인하였다. 형질 전환된 식물의 genome내의 T-DNA 부위를 분석하기 위하여 DNA-DNA 혼성화실험을 성시한 결과, pTi-12 DNA내에 존재하는 T region내에는 Sma I fragment들 중 분자량이 134 kb, 37 kb 및 4.75 kb인 것이 완전히 내포되어 있으며, T region의 양쪽 끝은 분자량이 각각 9.7 kb와 4.55 kb인 Sma I fragment에 연관되어 있음을 알 수 있었고, 또한 T-DNA에 내포된 Sma I fragment들은 pTi-12 DNA 분자내에서 서로 연결된 상태로 존재하고 있음을 확인하였다. In order to analyze the T-DNA region maintained in transformed plant genome, the DNA-DNA hybridization experiment was performed. As a result, it was found that Sma I fragments of Ti plasmid (pTi-12) 13.4 kb, 3.7 kb, and 4.75 kb in size, were completely contained within T-region, and each fragment of 9.7kb and 4.55 kb was joined to opposite ends of T-region. Also, it was identified that Sma I fragments which were contained within T-DNA region were covalently linked together within the pTi-12 DNA molecule.

        스마트한 지능형 교통체계 구축을 위한 ITS 관련법령 정비에 관한 연구

        우리나라의 지능형 교통체계(ITS)는 "국가통합교통체계효율화법"에 근거한 ITS 사업시행지침 및 각 ITS 표준에 의하여 구축되고 있다. 그러나 주요 ITS 설비에 대한 기술적 특징과 시공 구축에 대한 기술기준이 마련되어 있지 않은 채, 단 몇 개의 사업시행지침과 정보교환기술에 치우쳐 있는 ITS 표준만으로는 스마트한 지능형 교통체계(ITS) 구축을 기대하기 어렵다. 따라서 본 논문에서는 현행 법적 제도의 현황 및 문제점을 파악하고 스마트한 지능형 교통체계(ITS) 구축을 위한 기술기준 제정의 필요성에 대하여 살펴보기로 한다. In Korea, it is constructing the Intelligent Transportation System(ITS) in according with the ITS project implementation guideline and each ITS standard on the basis of "National Integrated Transportation System Efficiency Act". However, only a few ITS project implementation guideline and ITS standard for the exchange of information technologies are not able to construct a smart ITS, without technical regulation for the technical characteristics of the main ITS equipment. Therefore, in this paper, we identify the current status and problems of the legal system and look for the necessity of technical regulation enactment for the smart ITS construction.

      • 대장균에서 융합벡터를 이용한 인체 tau 유전자의 발현

        다량의 인체 tau 단백질을 얻기 위하여 융합벡터(pGEX-2T) 시스템을 이용하였다. 그 결과 인체 tau 단백질은 Escherichia coli에서 tac promoter의 조절하에 glutathione S-transferase(GST;EC 2.5.1.18) 단백질과의 융합체(GST-tau)로 대량으로 발현되었으며, 또한 분리 정제하기 쉬운 용해성(soluble form)으로 존재함을 알 수 있었으나 세포내에 존재하는 단백질가수분해효소의 공격에 의해 tau 단백질의 일북가 절단됨을 확인하였다. 그리고 GST단백질의 C-말단 인접부위를 인식하는 thrombin의 절단부위가 tau 단백질 내에도 존재함을 확인하였다. A fusion vector(pGEX-2T) system was used to obtain the human tau protein in abundance. As a result a large amount of human tau protein was expressed as soluable fusion protein(GST-tau) with glutathione S-transferase(GST;EC 2.5.1.18) protein under the control of tac promoter in Escherichia coli, however some of tau fragments were cleaved by the endogenous protease activities. It was also identified that the human tau protein contains the additional internal cleavage sites recognized by thrombin which digests the site immediately adjacent to the C-terminus of GST protein.

      • 재조합 tau 단백질과 그의 C-말단 합성 peptide 단편의 필라멘트 형성능

        Tau 단백질은 포유동물 뇌의 미세소관(microtubule)에 결합하여 있는 단백질 중의 하나로 Alzheimer 병의 경우 그것은 특정한 신경세포의 체세포수지상조직(somatodendrite) 부위에 축적되어 이중나선섬유(paired helical filament; PHF)를 형성하며 neurofibrillary tangle을 일으키는 주성분으로 작용한다. PHF를 형성하는데 관여하는 tau 단백질의 역할을 알아보기 위한 일환으로 사람의 tau 단백질을 Escherichia coli에서 발현시켜 분리하였고 또한 이 tau 단백질의 C-말단쪽 아미노산 잔기에 해당하는 5가지의 peptide 단편들(peptide 1 - peptide 5)을 합성하여 그들의 필라멘트 형성능을 조사하였다. 그 결과 재조합 tau 단백질은 짧은 필라멘트 구조를 형성한 반면, 하나의 합성 peptide(peptide 1)는 긴 필라멘트를 다량으로 형성하였으며 간혹 PHF와 유사한 구조도 형성하였음을 관찰하였다. Tau protein is one of the microtubule-associated proteins in mammalian brain. In Alzheimer's disease tau protein is immobilized in the somatodendritic compartment of certain nerve cells, where it forms part of paired helical filament(PHF), major constituent of the neurofibrillary tangle. To understand a role of tau protein in the formation of PHF, a recombinant human tau protein expressed in Escherichia coli and five kinds of synthetic peptide fragments(peptide 1 to peptide 5) which correspond to the parts of C-terminal half of the tau protein were prepared and examined their ability of self-assembly to form filamentous structures. Recombinant human tau proteins formed short filamentous structures, while a synthetic peptide fragment(peptide 1) could assemble in vitro into long filamentous structures in abundance and PHF-like structures sometimes.
