        국내 거베라 육종계통 형질전환 기초 조건 확립

        본 연구에서는 거베라 형질전환 기초기술개발을 위한 연구를 수행하였다. 경남원예화훼시험장에서 육성된 국내 거베라 18품종 중, Agrobacferium 감염에 순응적인 품종을 선발하기 위하여 18개의 국내 거베라 품종을 스크린하여 011, 016, GM023 등 12개의 순응형 계통을 스크린 하였으며 효율적인 형질전환을 위한 제균 항생제로는 cefotaxime이 선택되었다. 더 많은 캘러스 유도를 위한 전처리 배지의 호르몬 농도는 BA 2 ppm, Zeatin 2 ppm, IAA 0.2 ppm (2 x media)이 효율적이었고 재분화 배지의 호르몬 농도도 전처리 배지 호르몬의 농도와 동일하게 BA 2 ppm, Zeatin 2 ppm, IAA 0.2 ppm (2 x media)에서 효율적이었으나 GUS 발현에는 다른 차이가 없었다. 고효율 형질전환 체계 확립을 위하여 네 가지 다른 전처리 기간 처리와 두 가지 다른 Agrobacterium tumerfaciens 사용, 두 가지 다른 공배양 기간을 처리한 결과 전처리 하지 않은 엽병과 엽신을 LBA4404 Agrobacterium tumerfaciens을 사용하여 5일 간 공배양 기간을 가졌을 때와 dipping 처리하여 접종한 엽병과 엽신에서 높은 GUS 발현빈도를 나타냈다. Gerber (Gerbera hybrida) is a valuable ornamental species grown as a potted plant and cut flowers. However, genetic variability within the gerbera genus is very limited. So it is absolutely needed to introduce and widen genetic resources into gerbera lines by genetic transformation. For the purpose, 18 Korean gerbera lines were screened to establish Agrobacterium-mediated genetic transformation procedure. In an experiment to select Korean gerbera lines which are amenable to Agrobacterium-inoculation, 12 lines turned out to be positive in Agrobacterium-inoculation. More callus were produced from BA 2ppm, Zeatin 2ppm, IAA 0.2ppm in pre-culture and regeneration medium (2X media) but there was no difference in the frequency of GUS expression rate. In another experiment to find out optimal condition for highly efficient Agrobacterium-inoculation, petiole and leaf explants have been treated with four different pre-culture periods, two different co-culture periods and two different Agrobacterium strains. As a result, high GUS expression has been showed from petiole and leaf explants treated no pre-culture period with LBA4404 Agrobacterium tumerfaciens, 5 day co-culture period and dipping treatment.

        Structural and Electronic Properties of an Alkali Fulleride, $Rb_1C_{60}$

        Structural and electronic properties of an alkali metal fulleride, Rb1C60, was studied. In spite of the chain structure with shortdistance between balls along the crystallographic a-direction, the electronic structure calculation study with the X-ray defined crystal structure shows that Rb1C60 is a three-dimensional metal at room temperature. This result is different from the magnetic experiments in which the compound was found to behave as a quasi-one-dimensional metal. Partial Fermi surface nesting is supposed to be the reason for the metal-insulator transition found in Rb1C60 at ∼50 K.

        뇌성마비아동 어머니의 스트레스 및 대처방식 조사

        Purpose: The goal of this study was to analyze the degree of stress and methods of coping with stress in mothers of cerebral palsied children and to provide a basic resource for the development of policies to improve the mental health of mothers with cerebral palsied children. Methods: 85 mothers with cerebral palsied children and 77 mothers of normal children completed a self-administered questionnaire that evaluated the degree of stress and methods of coping with stress. Cronbach's ${\alpha}$ score was used to determine the internal consistency of the acquired data and the discriminated validity was estimated by Pearson's correlation coefficient. Independent t-tests were conducted to compare the degree of stress and method of coping with stress between mothers of cerebral palsied children and mothers of normal children and one-way ANOVA was performed to analyze the effect of generalized characteristics on stress in mothers of cerebral palsied children. Results: The degree of stress in terms of anxiety response and roles as mother for mothers with cerebral palsied children was higher than mothers with normal children. However, there was no significant difference between two group in regards to the methods of coping with stress during a stressful episode. The degree of stress for mothers with cerebral palsied children was greater when the child was younger. Conclusion: Mothers of cerebral palsied children had higher stress than that of mothers with normal children and felt more stress when their child was younger. Therefore, these results suggest that health-based policies should be developed to improve the mental health of mothers with cerebral palsied children.

      • Purification and Characterization of the Active Site of Phospholipase D

        양배추의 phospholipase D(PLD)는 아세톤 침전물로부터 10%의 회수율로 정제하였다. 정제방법은 Sephadex G-200 젤 여과법, DEAE-셀루로즈 크로마토그래피, ${\gamma}$-aminopropane agarose hydrophobic 크로마토그래피법을 순서대로 사용하였다. PLD의 순도는 SOS-PAGE로 확인되었고 분자량은 89,000으로 측정되었다. 활성도는 MES 완충용액 pH 6.3에서 $428{\mu}mol/min/mg$ 이었다. PLD의 열에 대한 안정성은 활용도에 필요한 칼슘이온과는 무관한 것으로 나타났다. Active site의 특성을 조사하기 위하여 N-ethylmaleimide, p-cholromercuribenzoate, p-bromophenacylbromide 및 diethylpyrocarbonate 등의 화학적 변형시약을 검토하였다. 효소의 활동도는 cysteine 변형시약으로 완전히 없어졌으며 active site에는 적어도 한 개의 histidine 기가 있을 것으로 밝혀졌다. Phospholipase D has been partially purified from an acetone powder of savoy cabbage in an overall yield of 10%. The purification involves gel filtration on Sephadex G-200, ion-exchange chromatography on DEAE-cellulose, and hydrophobic affinity chromatography using ${\gamma}$-aminopropane agarose gel. The purity of about 80% and molecular weight of 89,000 were estimated on the basis of gel electrophoresis. The specific activities of the partially purified enzyme was $428{\mu}mole/min/mg$ protein in MES buffer, pH 6.3, containing 4 mM phosphatidylcholine, 2 mM SDS and 10 mM $CaCl_2$ at $37^{\circ}C$. Calcium ion is essential to the enzyme activity but is not involved in the heat stability of the enzyme. To characterize the active site of the enzyme, the following chemicals were examined; N-ethylmaleimide, p-chloromercuribenzoate, p-bromophenacylbromide, and diethylpyrocarbonate. The enzymatic activity was lost completely by cysteine group modifying reagents and it was found that there was at least one histidine group in the active site.

      • 갈겨니(Zacco temminki)의 진화에 관한 연구 VII.갈겨니 2 Type의 Mitochondrial DNA변이

        한국산 담수 어류인 갈겨니(Zacco temminki)는 형태적으로 동일하거나 전기영동법에 의한 s-Mdh에 2type(MM과 MS type)이 있음을 (1987)이 주장하였으며 2type간 유전적 차이 정도를 분석한 결과 자매종으로 밝힌바 있다. 본 연구에서는 상기 결과를 토대로 2type의 mtDNA를 4개 집단에서 추출하여 11가지 제한 요소로 처리한 다음 fragment양상을 비교 분석하였다. 갈겨니 mtDNA의 전 genome크기는 약 16.7Kb였으며 frabment homolgy(F)에서 MS type 간 F값은 0.464,MM type간 F값은 0.762로 높은 값을 나타냈다. 또한 MM과 MS 이형간 평균 F값은 0.312(0.258-0.345범위)로 동형간보다 낮은 유사성을 나타내고 있다. 집단 또는 type간 nucleotide sequence divergence(p)를 산출한 결과 MS type 간의 P=0.128에 비해 동형간의 P값은 0.045로 매우 낮은 mtDNA sequece 변이를 나타내었다. 그러나 이형간인 2type평균 P값은 0.195(0.177-0.226범위)로 차이를 나타냈다. 이와 같은 2type간의 차이는 isozyme연구 분석 결과와 일치하고 있어 갈겨니 2type간의 종분화를 확신시키는 새로운 자료가 되었다. Mitochondrial DNAs of two Mdh allelotypes of the dark chub, Z. temmincki inhabiting in Korean fresh water, were analysed. Samples of each type were collected from four populations, and the fragment patterns for mtdNA of each type were explained from 7 of the eleven restriction enzymes with hexanucleotide recognition site. Genome size was approximately 16.7 kilobases. The highly typical mtdNA fragments of each type were discovered in digestion profiles produced by Eco RI and Pst I enzyrnes. The comparisons of restriction fragment patterns and relative digestion maps permitted the estimation of fragment homology (F) and nucleotide sequence divergence(p). Between the two identical types, sequence divergence(p) was 0.128(MS), and 0.045(MM), ; between the two different types, 0.195 (range 0.177-0.226). These result may provide a distinct difference more than the value derived from allozyrne analysis, and a powerful new molecular approach for assessing genetic-evolutionary relationship among fishes.

        학령기 정상 아동의 호흡 특성과 신체 조건에 관한 상관분석

        Purpose: Respiratory is an essential vital component for conservation of life in human, which is controlled by respiratory muscles and its related neuromuscular regulation. The purpose of this study is to assess lung capacity and respiratory pressure in healthy children, and to investigate relationship and predictability between respiratory pressure and other related respiratory functions. Methods: A total of 31 healthy children were recruited for this study. Demographic information and respiratory related factors were assessed in terms of body surface area (BSA), chest mobility, lung capacity, and respiratory pressure. Correlation between respiratory pressure and the rested variables was analyzed, and multiple regression using the stepwise method was performed for prediction of respiratory muscle strength, in terms of respiratory pressure as the dependent variable, and demographic and other respiratory variables as the independent variable. Results: According to the results of correlation analysis, respiratory pressure showed significant correlation with age (r=0.62, p<0.01), BSA (r=0.80, p<0.01), FVC (r=0.80, p<0.01), and FEV1 (r=0.70, p<0.01). In results of multiple regression analysis using the backward elimination method, BSA and FVC were included as significant factors of the predictable statistical model. The statistical model showed a significant explanation power of 71.8%. Conclusion: These findings suggest that respiratory pressure could be a valuable measurement tool for evaluation of respiratory function, because of significant relationship with physical characteristics and lung capacity, and that BSA and FVC could be possible predictable factors to explain the degree of respiratory pressure. These findings will provide useful information for clinical assessment and treatment in healthy children as well as those with pulmonary disease.

        Design, Fabrication and Electrical Tests of Pad Sensors for the Electromagnetic Calorimeter of the Future Linear Collider Experiment

        We have designed and fabricated silicon pad sensors for the electromagnetic calorimeter, to be used in the future linear collider experiment. The pad sensors have pixels with sizes of 0.98 × 0.98 cm², arranged in an active area of 6 × 6cm². Such a pixel size is chosen so that the calorimeter will meet its design goal in the resolution of the energy measurement. The pad sensors also have a structure of three guard rings outside of the active area, which have been proven to function well without high leakage current and breakdown. We present the design, fabrication, and electrical tests of the pad sensors.
