        발프로익산이 인체 테논낭 섬유아세포의 생존에 미치는 영향

        목적: 발프로익산이 배양된 인체 테논낭 섬유아세포의 생존에 미치는 영향에 대해 알아보고자 하였다. 대상과 방법: 일차배양한 섬유아세포에 0, 0.25, 0.5, 1.0 mM 발프로익산과 0, 1.0, 2.5 μg/mL 미토마이신 C에 각각 또는 동시에 5일간 노출시킨 후 MTT (3-[4, 5-dimethylthiazol-2-yl]-2, 5-diphenyltetrazolium bromide) assay를 이용하여 세포의 생존을 측정하였고, annexin-V/propidium iodide 이중염색 후 유세포 분석을 이용하여 세포고사의 정도를 측정하였다. 결과: 발프로익산은 농도에 비례하여 섬유아세포의 생존을 유의하게 저하시켰으며, 미토마이신 C를 추가한 경우 섬유아세포의 생존을 더욱 저하시켰다. 두 약제 모두 섬유아세포의 세포고사를 유발하였으나 발프로익산은 미토마이신 C에 비해 세포고사를 적게 유발하였다. 결론: 발프로익산은 섬유아세포의 생존을 저하시키며 세포고사를 유발한다. 또한 미토마이신 C를 병용한 경우 섬유아세포에 대한항증식효과가 더 나타날 것으로 생각된다. Purpose: To investigate the effects of valproic acid on the survival of cultured human Tenon’s capsule fibroblasts (HTFBs). Methods: Primary cultured HTFBs were exposed to 0, 0.25, 0.5, and 1.0 mM valproic acid with or without 0, 1.0, 2.5 μg/mL mitomycin C, and incubated for 5 days. Cell survival was assessed using an MTT (3-[4, 5-dimethylthiazol-2-yl]-2, 5-diphenyltetrazolium bromide) assay and the degree of apoptosis was assessed by flow cytometry using annexin-V/propidium iodide double staining. Results: Valproic acid decreased the survival of HTFBs in a dose-dependent manner, and survival was further decreased by adding mitomycin C to valproic acid. Both valproic acid and mitomycin C induced apoptosis of HTFBs. Valproic acid induced less apoptosis than mitomycin C. Conclusions: Valproic acid decreased the cellular survival of HTFBs and induced apoptosis. The antiproliferative effects of valproic acid were further enhanced by the addition of mitomycin C.

        유세포분석을 이용한 레즈브라트롤이 인체 테논낭 섬유아세포의 생존에 미치는 영향에 대한 연구

        목적: 레즈브라트롤은 세포의 종류에 따라 세포보호 효과를 나타내거나 독성을 나타낸다. 레즈브라트롤이 배양된 인체 테논낭 섬유아세포의 생존에 미치는 영향에 대해 알아보고자 하였다. 대상과 방법: 일차배양한 섬유아세포에 0, 10, 100 μM의 레즈브라트롤에 3일간 노출시킨 후 MTT assay를 이용하여 세포의 생존을 측정하였고, annexin-V/propidium iodide 이중염색 후 유세포분석을 시행하여 세포고사에 미치는 영향을 측정하였다. 결과: 레즈브라트롤은 10 μM의 농도에서부터 섬유아세포의 생존을 유의하게 저하시켰다(p=0.04). 유세포분석에서 10 μM의 농도에서는 섬유아세포의 세포고사를 유의하게 유발하지 않았으나(p=0.89), 100 μM의 농도에서는 섬유아세포의 세포고사를 유의하게 증가시켰다(p=0.003). 10 μM와 100 μM의 농도에서 세포괴사에는 유의한 영향을 미치지 않았다(p=0.74, 0.33). 결론: 배양된 인체의 테논낭섬유아세포에 대해 레즈브라트롤은 세포의 생존을 저하시키며 세포고사를 유발하므로 인체의 테논낭섬유아세포에 대해 레즈브라트롤은 항증식작용을 나타낼 수 있을 것이다. Purpose: Resveratrol exerts cytoprotective or cytotoxic effects according to cell type. This study was performed to evaluate the effects of resveratrol on the survival of cultured human Tenon's capsule fibroblasts (HTFBs). Methods: Primarily cultured HTFBs were exposed to 0, 10, or 100 μM resveratrol for 3 days. Cellular survival was assessed us-ing the MTT assay and degree of apoptosis was analyzed with flow cytometry using annexin-V/propidium iodide double staining. Results: Resveratrol decreased the survival of HTFBs after exposure to 10 μM (p = 0.04). In flow cytometric analysis, 10 μM re-sveratrol did not affect the degree of apoptosis (p = 0.89), but 100 μM resveratrol increased the degree of apoptosis (p = 0.003). Both 10 and 100 μM resveratrol did not affect the degree of necrosis (p = 0.74, 0.33). Conclusions: Resveratrol decreased cellular survival of cultured HTFBs and induced apoptosis. Thus, resveratrol may exert an-tiproliferative effects on HTFBs.

        품질지표기반 정치 후원금 지원을 위한 국회의원 추천시스템 연구

        Purpose: During 2015-2019, the average amount of political donation to the national assembly members in Korea was 1,000 won per person. Despite its benefits such as receiving tax credits, the donation system has not been actively practiced. This paper aims to promote political donations by suggesting a recommendation system of national assembly members by analysing the bills they proposed. Methods: In this paper, we propose a recommendation system based on two aspects: how similar the newly proposed or ammended bills are to the sponsors’ interest (similarity index) and how much effort national assembly members put into those bills (intensity index). More than 25,000 bills were used to measure the recommendation quality index consisted with both the similarity and the intensity indices. Word2vec was used to calculate the similarity index of the bills proposed by the national assembly member to the sponsor’s interest. The intensity index is calculated by diving the number of newly proposed or entirely revised bills with the number of senators who took part in those bills. Subsequently, we multiply the similarity index by the intensity index to obtain the recommendation quality index that can assist sponsors to identify potential assembly members for their donation. Results: We apply the proposed recommendation system to personas for illustration. The recommendation system showed an average f1 score about 0.69. The analysis results provide insights in recommendation for donation. Conclusion: n this study, the recommendation system was proposed to promote a political donation for national assembly members by creating the recommendation quality index based on the similarity and the intensity indices. We expect that the system presented in this paper will lower user barriers to political information, thereby boosting political sponsorship and increasing political participation.
