        피복물 종류에 따른 더덕의 생육 및 항산화 물질 비교

        Background: This study determined the effects of mulching, an environment friendly organic cultivation method, on antioxidant compound contents and growth in Codonopsis lanceolata, commonly known as Deodeok. Methods and Results: C. lanceolata was treated by mulching with several different methods (a non-woven fabric, biodegradable film, or rice husks) and also treated with hand weeding. A non-treatment plot was used as a control. The growth and levels of weed control in C. lanceolata were better in plants cultivated under mulching treatments (non-woven fabric, biodegradable film, and rice husks) than in those under non-mulching treatments (hand weeding and non-treatment). The contents of antioxidant compounds, such as total flavonoids, phenolics, and anthocyanins, were highest under the biodegradable film treatment, followed by the nonwoven fabric treatment, rice husks treatment hand weeding, and non-treatment. There were identifiable differences in DPPH and ABTS activity in comparison to antioxidant compound content by solvent fractions. Mulching treatments resulted in higher DPPH scavenging activity in water and ethyl ether fractions and ABTS scavenging activity in n-butanol fractions than in other fractions, as opposed to hand weeding and non-treatment groups, although total activity of DPPH and ABTS did not increase with mulching treatments. Conclusions: Mulching C. lanceolata with biodegradable film and non-woven fabric is an effective method for improving plant growth and inhibiting the occurrence of weeds as well as for increasing antioxidant compound content and altering antioxidant activity.

        타액반 피검물에서 개인식별을 위한 DNA의 유전자형 검사

        Cigarette butts from 5 smokers were gathered and then, place in room temperature for 1, 3, 5, 7, 15 days. The possible use of the cigarette butts for individual identification was evaluated in sex determination, amplification of D1S80 locus, polymorphisms of HLA-DQA1 gene from the extracted DNA. 1. DNA extraction was possible in cigarette butts left in room temperature for 15 days, so it can be applicable to individual identification by polymerase chain reaction (PCR). 2. Amplification of X-Y homologous amelogenin gene by PCR made it possible to identify the sex in saliva stains (cigarette butts). 3. Amplification of D1S80 locus can be acquired from adding the bovine serum albimine and hot start PCR procedures from forensic samples such as saliva stains (cigarette butts), so the AMP-FLPs examining is possible. 4. Genotype could be determined simply and rapidly using Amplitype?? HLA-DQα forensic kit in examining the HLA-DQA1 gene. From the investigation, DNA extraction, sex determination, amplification of D1S80 locus, polymorphisms of HLA-DQA1 gene were successfully done even though the cigarette butts were left for 15 days at room temperature. Therefore cigarette butts are highly reliable and applicable as molecular biological samples for individual identification.

        홍합 섬유실 처리가 침수 상태의 눈개승마 생장 및 생리적 반응에 미치는 영향

        이 연구는 침수 상태의 눈개승마에 대한 홍합 섬유실 처리의 생태적 영향을 평가하는 목적으로 수행하였다. 실험을 위해 홍합 섬유실 함량을 0%, 3%, 5% 및 10%로 조절하였고, 침수 조건은 토양 표면에서 1cm의 수위를 유지하였다. 눈개승마는 침수 스트레스에 따라 최대광합성속도, 최대카르복실화율 및 최대전자전달율 등 광합성 활성이 감소하는 것으로 나타났으며, 특히 홍합 섬유실을 처리하지 않은 대조구가 광합성 활성 및 생장특성이 가장 불량한 경향을 보였다. 그러나 광합성속도와는 달리 수분이용효율은 침수 처리 후 12일경 다소 증가하였는데 이를 통해 기공닫힘으로 인한 수분 및 이산화탄소 교환이 원활하게 이루어지지 않아 광합성의 감소가 나타난 것을 알 수 있었다. 엽록소 형광분석 결과 역시 홍합 섬유실 처리구에 비해 무처리구의 기능지수 감소가 뚜렷하였는데, 이는 광계 2 활성의 감소가 두드러진 것을 의미하며, 이와 대조적으로 홍합 섬유실 3% 처리구는 광합성 활성 및 생장특성이 가장 양호하였고 홍합 섬유실 5% 처리구 역시 유사한 경향을 나타냈다. 결과적으로 홍합 섬유실 처리는 눈개승마의 침수 내성 증가와 생장에 도움을 주는 것을 알 수 있었다. This study aimed to assess ecological effects of byssal thread application on Aruncus dioicus var. kamtschaticus under waterlogging condition. The treatments were regulated with the four different byssal thread contents (bt 0%, bt 3%, bt 5% and bt 10%). And also waterlogging condition were maintaining the water level 1 cm above soil surface. Under waterloggins stress, photosynthetic activity such as maximum photosynthesis rate (PN max), maximum carboxylation rate (Vcmax) and maximum electron transport rate (Jmax) was significantly reduced. Particularly, the bt 0% showed relatively lower growth performances and photosynthetic activity than others treated. However, water use efficiency (WUE) was increased slightly at 12days. This implies that photosynthetic rate is reduced due to an inability to regulate water and CO2 exchange through the stomatal. According to OKJIP analysis, The bt 0% exhibited performance index (PIabs) were dramatically decreased, which reflects the relative reduction state of the photosystem II activity. In contrast, Photosynthetic activity and growth performances were highest in the bt 3% treatment, and similar result occurred in the 5% treatment. Similarly, the bt 5% treatment also showed relatively higher than others treated. Thus, byssal thread, when mixed with soil, promotes the growth and waterlogging stress tolerance of A. dioicus var. kamtschaticus.

      • 상토의 조성에 따른 느티나무 묘목의 생육특성

        This study was conducted to find out the optimum composition of nursery soil for raising seedling of Zelkova serrata. The treatments were regulated with the three different soil types : full peat moss(PM 100%), peat moss + perlite + vermiculite(1:1:1), peat moss + perlite + vermiculite(1:2:1). The full peat moss(PM 100%) showed the highest photosynthetic activity such as maximum photosynthesis rate(PN max), water use efficiency(WUE), Fv/Fm, PIabs and SPAD value. And also, full peat moss(PM 100%) showed relatively better growth performances. As a result of surveying the whole experiment, full peat moss(PM 100%) soil increases growth performances and enhances overall quality.

        NaCl 처리가 고들빼기의 생장과 생리적 특성에 미치는 영향

        Background: This study was conducted to investigate the effects of NaCl concentration on the photosynthetic parameters, chlorophyll fluorescence and growth characteristics of Crepidiastrum sonchifolium. Methods and Results: As treatments, we subjected C. sonchifolium plants to four different concentrations of NaCl (0, 50, 100 and 200 mM). We found that the photosynthetic parameters maximum photosynthesis rate (PN max), net apparent quantum yield (Φ), maximum carboxylation rate (Vcmax), and maximum electron transport rate (Jmax) were significantly reduced at an NaCl concentration greater than 100 mM. In contrast, there was an increase in water-use efficiency with increasing NaCl concentration, although in terms of growth performances, leaf dry weight, root dry weight, stem length, and total dry weight all decreased with increasing NaCl concentration. Furthermore, leakage of electrolytes, as a consequence of cell membrane damage, clearly increased in response to an increase in NaCl concentration. Analysis of the polyphasic elevation of chlorophyll a fluorescence transients (OKJIP) revealed marked decrease in flux ratios (ΦPO, ΨO and ΦEO) and the PIabs, performance index in response to treatment with 200 mM NaCl, thereby reflectings the relatively reduced state of photosystem II. This increase in fluorescence could be due to a reduction in electron transport beyond Q− A . We thus found that the photosynthetic parameters, chlorophyll fluorescence and growth characteristics of C. sonchifolium significantly increased in response to treatment with 200 mM NaCl. Conclusions: Collectively, the findings of this study indicate that C. sonchifolium shows relatively low sensitivity to NaCl stress, although photosynthetic activity was markedly reduced in plants exposed to 200 mM NaCl.

      • 토양 건조 및 침수처리가 박태기나무의 광계 II 활성에 미치는 영향

        This study was conducted to investigate the photosystem II activities of Cercis chinensis by soil water condition. Drought stress was induced by withholding water and waterlogging treatments was immerging the pots for 15 days. Results showed that the relative activities per reaction center such as ABS/RC, TRo/RC and Dio/RC were significantly increased compared with the control group after 12 days in waterlogging treatments. Particularly, Dio/RC increased substantially under waterlogging stress, indicating that excessive energy was consumed by heat dissipation. Furthermore, the performance index on absorption basis(PI<sub>abs</sub>) and responses to structural and functional PS II(SFI<sub>abs</sub>) were dramatically decreased after 15 days in both the drought and waterlogging treatments, which reflects the relative reduction state of the photosystem II. These results of chlorophyll a fluorescence by OKJIP analysis show that the sensitive changes photosystem II activity. Thus, on the basis of our results that Cercis chinensis was exhibited a strong reduction of photosynthetic activity to waterlogging stress, and OKJIP parameters such as ABS/RC, DIo/RC, PI<sub>abs</sub> and SFI<sub>abs</sub> could be useful indicator to monitor the physiological states of Cercis chinensis under soil water condition.
