        ODP Leg 204 Site 1249C와 Site 1251B 퇴적물의 유기물 기원 및 지화학적 특성

        오레곤 대륙 주변부 남부 하이드레이트 릿지에서 실시한 ODP Leg 204에서 회수한 코어 Site 1249C와 Site 1251B의 유기물 기원 및 유기지화학적 특성을 규명하기 위하여 원소분석과 Rock-Eval 열분석 및 동위원소 비 분석을 실시하였다. 분석결과 Site 1249C와 Site 1251B의 유기물은 해양기원 유기물이 우세하며 이러한 결과는 퇴적물의 퇴적당시 환경이 일반적인 해양 환경이었음을 시사한다. 그러나 Rock-Eval 열분석 결과는 다른 분석결과와 반대의 결과를 나타내는데 이러한 결과는 열적으로 미성숙한 유기물이 열분석 과정에서 유기물의 특성이 제대로 반영되지 못하기 때문이다. 따라서 열분석 결과 해석은 다양한 지화학적 결과를 종합하여 해석할 필요가 있다. 코어의 유기물 기원이 동일함에도 불구하고 Site 1249C는 전 구간에서 가스 하이드레이트의 집적이 높은 반면, Site 1251B는 가스 하이드레이트가 일부 구간에서만 소량 발견되었다. 이러한 결과는 하이드레이트 정상부에 위치한 Site 1249C는 가스 하이드레이트 생성에 필요한 심부 열기원 가스가 천부 퇴적물로 이동할 수 있는 이동 통로(Horizon A, BSR2)가 존재하는 것을 시사한다. 그러나 가스 하이드레이트 집적이 미비한 분지 지역의 Site 1251B는 열기원 가스가 심부로부터 이동할 수 있는 가스 이동 통로가 제한적이고 그에 따른 가스 공급이 원활하게 이루어지지 않기 때문에 현장 퇴적물의 속성작용에 의한 생물기원 가스가 우세하여 가스 하이드레이트 생성이 제한적인 것으로 추정된다. To study biogeochemical characteristics and origin organic matter, sediment samples were taken from Site of 1249C and Stie 1251B of ODP Leg 204. Data of Rock-Eval, isotope, and element analysis generally indicate dominance of marine organic matter in sediments deposited under marine sedimentary environment. Only Rock-Eval data are somewhat different from those of others owing to under-maturation of organic matter. Samples of Site 1249C show high content of gas hydrate, whereas Site 1251B low content of gas hydrate in some intervals of the core. This result may be accounted to different location of two cores and presence of transportation passage (Horizon A, BSR 2) of thermogenic gas in the core, 1249 C. However, Site 1251B Located in the basin of low accumulation of gas hydrate is presumed to be limited in the gas hydrate production. Because not only transportation passage is limited to move thermogenic gas from the core, but also gas supply was not enough. Therefore, the biogenic gas that resulted from diagenesis of there sediment is superior.

        제마성향정기산과 열다한소탕의 항염증 효능 비교 연구

        Objectives : Jema-sunghyangjungkisan (JSGS, Jima Xingxiang Zhengqi san) and yeoldahanso-tang (YDHST, Reduo hanshao decoction) are traditional herbal formulas which commonly used to prevent and treat stroke in traditional korean medicine. However, JSGS and YDHST extracts have not been previously reported to have anti-inflammatory effects. Therefore, We measured the anti-inflammatory effects of JSGS and YDHST extracts on lipopolysaccharide (LPS)-stimulated murine macrophage cell line, RAW 264.7 cells. Methods : To investigate the anti-inflammatory activities of JSGS and YDHST extracts, tumor necrosis factor-alpha (TNF-α), interleukin (IL)-6, nitric oxide (NO) and prostaglandin E2 (PGE2) were examined in RAW 264.7 cells with LPS of 1 ㎍/㎖. Results : JSGS and YDHST extracts did not have any cytotoxicity in RAW 264.7 cells. TNF-α concentration decreased 49.67% at 500 ㎍/㎖ by JSGS but, YDHST has no statistically significant effect at all concentration. IL-6 accumulation on JSGS and YDHST extracts in LPS-stimulated RAW 264.7 cells reduced 22.03% and 41.44% at 500 ㎍/㎖ respectively. In addition, JSGS has no inhibitory effects on NO accumulation and YDHST reduced 10.08% at 500 ㎍/㎖. Moreover, JSGS and YDHST treatment does-dependently reduced the PGE2 production. In particular, YDHST (500 ㎍/㎖) extract was more effective compared with 10 ng/㎖ of indomethacin which is the PGE2 positive control. Conclusions : Our results suggest that treatment of JSGS and YDHST extracts decreased the LPS-stimulated inflammation. Therefore, in the present study, we demonstrated that JSGS and YDHS may be used as a potential anti-inflammatory therapeutic agent.

        어혈처방 3종이 지방전구세포 분화에 미치는 영향

        Objectives : Blood stasis (BS) is related to be caused by blood circulation and stagnation which are cancer, atherosclerosis and hyperlipidemia in traditional medicine. We extracted 3 kinds of BS formula; Seogakjihwang-tang (SGT), Tonggyuhawlhyul-tang (THT), Hyulbuchukeo-tang (HCT). This study was conducted to investigate whether the 3 kinds of herbal formula extracts have inhibitory efficacy association with anti-adipogenesis. Methods : To investigate the anti-adipogenesis, we used the mouse fibroblast cell line, 3T3-L1 which differentiated into adipocytes in response to insulin, IBMX and dexamethasone (MDI). Cytotoxicity of herbal formula extracts were examined by CCK-8 kit. Intracellular lipid droplets were detected by Oil-Red-O staining. Triglyceride (TG) and leptin were measure using elisa kit. Results : The yield of water extracts was 14.62% (SGT), 21.27% (THT), 20.02% (HCT). Lipid accumulation was reduced significantly by 3 kinds of herbal formula compared to control. Especially, THT and HCT decreased lipid droplet, respectively at all concentration. The TG and leptin were also inhibited by 3 kinds of herbal formula. The IC50 of TG were 280.51 ㎍/㎖ (SGT), 52.62 ㎍/㎖ (THT), 313.99 ㎍/㎖ (HCT). The IC50 of leptin were 348.76 ㎍/㎖ (SGT), 164.02 ㎍/㎖ (THT), 257.00 ㎍/㎖ (HCT). THT was better than other herbal formula on anti-adipogenesis. Conclusion : kinds of herbal formula inhibited adipogenesis in MDI-induced 3T3-L1 adipocytes, as indicated by the significant reduction in TG and leptin concentration without cytotoxicity. Therefore, 3 kinds of herbal formula for BS might act as a therapeutic agent for preventing lipid diseases, such as obesity and atherosclerosis.

        발효 균주에 따른 전복발효추출물이 피부 미백 및 주름개선에 미치는 영향

        The effect of fermented abalone hydrolysate by various strains was studied as a raw material for functional cosmetics. Chondroitin sulfate content of fermented extract was compared with normal extract to measure change of active component content. The anti-oxidant effect was researched by using DPPH radical inhibition activity assay. To confirm whitening and anti-wrinkle effects, fermented abalone extract was measured by melanin assay and sicol collagen assay. The cell viability of fermented abalone extract was measured to induce toxicity in NIH-3T3 and B16F10 cells using thiazolyl blue tetrazolium bromide (MTT). Also, subjects were tested on their irritation, elasticity, wrinkle etc. DPPH radical inhibition activity test of fermented abalone extract showed over 80% at 0.01%. It was higher than that of ascorbic acid. The cell viability was confirmed to no toxicity by the MTT assay. Anti-wrinkle effect showed 70% collagen increase at 1% and similar to adenosine (0.04%). Furthermore, fermented abalone extract were showed 30%-35% skin elasticity in clinical test and area of skin wrinkles was improved 20% in fermented abalone extract compared with normal extract. The pores and pophyrin activity were decreased significantly. These results suggest that fermented abalone extract was available as a natural ingredient of cosmeceutical by anti-wrinkle and elasticity effects.
