        We aimed to investigate the effect of in vitro allergen challenge on activation of eosinophils and expression of intercellular adhesion molecule-1(ICAM-1) in the epithelial and endothelial cells of the nasal mucosa of allergic rhinitics. Materials and methods . Inferior turbinate mucosal specimens which were obtained from 16 perennial allergic rhinitics and sensitized to D. farinae during conchotomy operations were used. Each mucosal specimen was divided into six pieces. The first two pieces were used as basic control group without culture, the second two pieces as culture-control group which was cultured with diluent only, and the last two pieces as antigen-challenge group cultured with D. farinae solution for 24 hours. Specimens were immunohistochemically stained using monoclonal anti- body against eosinophil cationic protein(EG1 for all eosinophils and EG2 for activated forms) and ICAM-1. Results .' Average counts of EG1 cells and EG2+ cells within 1 mm of lamina propria of nasal mucosa were 14 and 11 respectively in the basic control group, 17 and 19 in culture control group, and 16 and 16 in the antigen-challenge group. ICAM-1 expression on vessels was observed more than half of t,he vessels in the nasal mucosa and there was no significant difference between groups, however, its expression on the epithelial cells was most prominent in the antigen-challenge group (2.1 in basic control group, 3.3 in culture control group, and 6.2 in antigen-challenge group). Conclusion: Most of the eosinophils infiltrat,ing into the nasal mucosae out of vessels in allergic rhinitics are activated forms. Significant up-regulation of epithelial ICAM-1 expression by antigen-challenge suggests that ICAM-1 contributes to the transmigration of eosinophils from the subepithelial tissue into the epithelial cell layer and then finally into the nasal secretion.

        돼지에서 단일 전신 방사선 조사 용량에 따른 혈액변화 관찰

        4주령의 돼지에서 여러 용량의 방사선 조사를 하고 경시적으로 혈액 수치의 변화, helper T-cell 및 cytotoxic T-cell에 대한 변화를 관찰 하였다. 6 Gy 이상을 조사한 돼지에 3일이후 백혈구와 혈소판의 수의 감소되어 회복 되지 않았으며, 식욕 부진, 설사 등의 부작용을 보이며, 조사 후 10일부터 15일 사이에 모두 폐사 하였다. 4 Gy 전신 단일 조사한 돼지에서는 조사 후 임상적인 부작용을 보이지 않았으며, 9일 이후 백혈구, 혈소판이 증가하여 회복되는 경향을 보였다. 본 연구의 결과는 돼지를 사용한 방사선 생물학적 실험에서 혈액변화 및 면역억압 관찰에 대한 기초 자료로 활용 될 수 있을것이다. We examined total body irradiation (TBI)-induced effects by complete blood count (CBC) and fluorescence-activated cell sorter analysis (FACS) in the piglet following radiation irritation. A CBC included red blood cell count, white blood cell count, and platelet cell count. Four piglets were examined in this study and each piglet was divided by irradiation dose, two piglets with 4 Gy, two with 6 Gy, one with 8 Gy. All piglets showed leukopenia, thrombocytopenia after irradiation. In 6 and 8 Gy group, three piglets showed severe hemostatic disorder and gastrointestinal disorder suchas diarrhea and anorexia, and they died between 10 and 15 days after radiation irritation. In 4 Gy, two piglets showed no clinical sign after radiation injury, but persistent leukopenia was shown in blood examination. We suggest that a single TBI dose less than 6 Gy is adequate for conditioning piglet for bone marrow transplantation.

      • 알레르기비염에서 비내 부데소니드와 경구 테페나딘의 비교 : 증상 및 비즙 호산구에 미치는 영향 effect on the symptom score and nasal secretion eosinophils

        Background : Eosinophilia in the nasal secretion and mucosal tissues is the characteristic finding of allergic rhinitis. We compared the effects of nasal budesonide dipropionate and oral terfenadine on the symptom score and nasal secretion eosinophils in perennial allergic rhinitics. Materials and Method : Study subjects consisted of 81 patients with perennial allergic rhinitis and nasal eosinophilia. Fifty-seven patients were treated with nasal budesonide and 24 patients were treated with oral terfenadine for two weeks respectively. Nasal secretion eosinophils were measured as the percentage of total leukocytes under microscope. Symptom scores for sneezing, rhinorrhea, obstruction and itching were graded from 0 (no symptom) to 3 (severe symptom). Results : In budesonide treatment group, symptom score reduction (total symptom score before treatment-total symptom score after treatment) was 5.1, while it was 2.3 in the terfenadine treatment group, which revealed a statistically significant difference (p$lt;0.01). Reduction of eosinophils was 50% in the budesonide treatment group and 28% in terfenadine treatment group (p=0.045). There was statistically significant correlation between total symptom score and % eosinophils (r= 0.668 , p$lt;0.01 ) Conclusion : Although both the nasal budesonide and oral terfenadine substantially reduced the proportion of eosinophils in the nasal secretion as well as symptoms in allergic rhinitics with eosinophilia, budesonide was superior to terfendine. Nasal eosinophils correlated with reduction of allergic symptoms in a statisticaly significant manner.
