        다채 (Brassica campestris var. narinosa) 유묘의 형질전환 및 일시발현의 정량적 분석

        Brassica campestris var. narinosa는 새싹채소로 애용되는 식물로서 간단한 용기에서 손쉽게 재배할 수 있다. 본 연구에서는 vacuum-infiltration을 통한 Agrobacterium-mediated transformation에 의해서 B. campestris var. narinosa 싹에서의 GUS일시발현을 성공적으로 확인하였다. 발아전의 종자가 발아를 이미 시작한 종자보다 그 형질전환 효율이 높게 나타났다. 한편, hydrogen peroxide를 2일 생장시킨 싹에 처리한 후 형질전환 하였을 때 GUS발현이 증가되는 것을 관찰하였다. 간염항원유전자의 형질전환과 ELISA에 의한 항원단백질의 발현량 분석 시 hydrogen peroxide 처리 싹이 비처리 형질전환 싹에서 보다 2배 이상의 발현량이 측정되었다. Tah tasai chinese cabbage (Brassica campestris var. narinosa), a vegetable plant popularly consumed as several-days-old seedlings in oriental countries, can be easily cultivated using a simple appliance. We demonstrated that Agrobacterium-mediated transformation via vacuum infiltration (agroinfiltration) resulted in a successful transient GUS gene expression in tah tasai chinese cabbage seedlings. Pre-germinated seeds were found to be more susceptible to Agrobacterium infection than one-day-old or two-days-old seedlings. We also demonstrated that hydrogen peroxide (HPO) treatment increased GUS expression especially for two-days-old seedlings. In ELISA using seedlings transformed with hepatitis B surface antigen (HBsAg) DNA by agroinfiltration, HBsAg protein synthesis increased more than two folds by HPO treatment to two-days-old seedlings in comparison to the mock-treated pre-germinated seeds.

        Sodium Hypochlorite Solution As a Chemical Wounding Agent for Improving Agrobacterium-mediated Chinese Cabbage Seed Transformation

        배추 종자 및 유묘에 대하여 GUS발현 또는 hepatitis B surface antigen (HBsAg)발현 벡터를 지니는 Agrobacterium tumefaciens LBA4404 세포를 이용하여 진공침윤(agroinfiltration)에 의한 형질전환을 시도하였다. 특히 ELISA를 이용한 HBsAg발현의 정량적 분석에서 agroinfiltration방법은 형질전환효율이 매우 저조하게 나타났다. 그러나 차아염소산나트륨 용액을 발아 전 또는 발아 중인 배추종자에 처리한 후 agroinfiltration을 실시한 경우 형질전환 효율이 $2\~5$배 증가하였다. 따라서 차아염소산나트륨 등의 화학연마제에 의한 종자의 상처발생이 Agrobacterium의 감염을 용이하게 함으로써 배추유묘에서의 일시유전자발현을 증대시키는 것으로 제안되고있다. Chinese cabbage (Brassica campestris ssp. napus var. pekinensis Makino) seeds/seedlings were transformed via vacuum-infiltration with recombinant Agrobacterium tumefaciens LBA4404 cells. The agroinfiltration method was determined to be unsuccessful for Chinese cabbage transformation during the analysis of hepatitis B surface antigen expression by ELISA. However, treatment of sodium hypochlorite solution, prior to agroinfiltration, to pregerminated or germinating 1 day- or 2 days-old seeds was proven effectively to enhance transformation efficiency, suggesting that chemical wounding caused by sodium hypochlorite reaction might facilitate Agrobacterium infection and, therefore, transient gene expression in Chinese cabbage sprouts.

        단보 : NaOH처리에 의한 Agrobacterium이용 팽이균사체 형질전환

        Agrobacterium harboring vector pCHBs with hygromycin phosphotransferase(hph) and hepatitis B virus surface antigen(HBsAg)gene was transformed into the mycelial culture of Flammulina velutipes. In particular, mild NaOH solution was treated to the mycelia before Agrobacterium infection step. This was purposed to generate putative surface wounds in the mycelial cell walls. The results showed that hygromycin-resistant(hygr) mycelia could be obtained only from NaOH-treated mycelia but not from intact mycelia. The integration of hygr gene in fungal genome was confirmed by PCR. In addition, a single transgene integration and heterologous protein expression in F. velutipes could be verified by Southern blot hybridization and western blot analysis, respectively. This study demonstrated an efficient tool for the Agrobacterium-mediated transformation of F. velutipes mycelia.

        Korean Examinees’ Perceptions of Testing Spoken English: A Movement Toward Proficiency

        The purpose of this study is to investigate Korean test-takers’ perceptions on ACTFL Proficiency Guidelines and Oral Proficiency Interview-computer rubrics, and to understand Korean examinees’ beliefs regarding OPIc preparation and test results currently in use. The source of data for this research is a questionnaire filled in by test-takers. Results are presented in the following categories: ACTFL and ACTFL proficiency guidelines, OPIc in general, OPIc rubrics and test items, OPIc criteria, OPIc test preparation, OPIc tests in use, and English speaking tests. It was found that Korean examinees did not have enough information on ACTFL, proficiency levels, OPI and proficiency movement, although they had positive perceptions regarding OPIc rubrics. It is suggested that test-takers need to acquire sufficient information on ACTFL Proficiency Guidelines and OPIc preparation and speaking strategies from the test institution. (Chung-Ang University)

        내러티브 탐구 학습을 통한 교육 경험의 성찰

        본 논문은 내러티브 탐구를 새롭게 학습한 연구자들이 텍스트를 통해 이해했던 것을 개인의 삶에 적용시키며 그 교육적 경험의 성찰을 기록한 내용을 재구성한 것이다. 기존의 연구방법과는 달리 개인의 체험이 이야기 형식으로 시공간을 통해 사회적 의미로 심화, 확대 될 수 있다는 점에서 내러티브 탐구는 삶의 적용과 실천을 가능하게 하였다. 강독으로 시작한 모임이 점점 개인의 삶 속에서 교육적 성장과 성찰을 이끌었다는 체험을 세 명의 연구자들이 각자의 이야기로 서술하였다. 학교 가는 길로 시작하는 연구자의 이야기는 강의하러 가는 길목에서 마주치는 학생들을 보며 과거 학생 시절의 연구자 모습을 떠올리게 하였고 이것은 다시 현재로 돌아와 교실에서의 교수자와 학생의 관계 그리고 전달과 소통의 문제로 연결되었다. 내러티브 탐구를 통해 학교라는 공간에서 시작한 교육 경험의 성찰은 다시 연구자의 삶으로 확대되어 연구자들이 삶과 앎을 하나로 묶는 기회를 갖기도 하였다. The purpose of this study is to investigate three researchers` reflections on their educational experience and development through narrative inquiry. After studying Narrative Inquiry (Clandinin & Connelly, 2000) for seven months, we have still tried to apply the qualitative knowledge into our lives. To us, narrative inquiry was not just a research method. Rather, it leads us to reflect on our educational experience and development as teachers and researchers. Acquiring narrative research tradition helps us pursue our own professional career on the right track. This research paper illustrates how our reflections evolve on our educational fields and lives through narrative inquiry. Participants` journals were collected and classified into mainly two: one is our narratives on our educational fields and the other is our reflections on our overall lives through narrative inquiry. Narrative research was judged to be the appropriate way for evaluating the educational experience of teachers at school. On the way to teaching, we looked back on the past and could see the present. Moving from outward to inward in a situated place, we could see our past and present, and our students through us.

        언어교육 분야의 정체성 연구에 관한 경향성 탐색

        The purpose of this study is to explore a research trend of academic research papers published in Korea on the topic of language education and identity. A total number of 40 papers were collected, classified and coded into 1) research subjects, sub-categorized into language learners and language (native and non-native) teachers and 2) the type of language use, sub-categorized into English (ESL, EFL), Korean (KHL) and Bilingual. A majority of the research papers was carried out on English language education in terms of language learners' identity construction. It showed that these studies contribute to better understanding of the complex relations between language education and identity formation. Most of the research findings revealed that language learner's identity is dynamic, contradictory and multiple over time and space in sociocultural context all of which is based on post-structuralistic theory of identity. It was confirmed that the understanding of social identity in language learning and teaching could play a crucial role in helping learners and teachers in diverse educational contexts.
