순수 분리된 Fur 단백질과 Aerobactin Operon의 작동자 부분을 포함하는 DNA 절편간의 결합하는 성질을 Nitrocellulose filter binding assay에 의하여 측정하였다. Fur 단백질은 $Fe^{2+}$ 이외에도 많은 전이 금속을 보조 억제 인자로서 수용하는 것으로 보인다. 얻어진 Fur-작동자 결합의 해리상수는 $Mn^{2+}$을 보조 억제 인자로 사용하였을 경우$(3{\sim}6){\times}10^{-12}M^{2}$이었으며 Fur 단백질은 Dimer로서 작동자와 결합하는 것으로 나타났다. Fur-작동자 결합을 $43^{\circ}C$에서 수행 하였을 때는 $23^{\circ}C$ 또는 $37^{\circ}C$ 때와 동일한 해리상수를 얻었다. 그러나 Fur 단백질은 고온에서는 동량으로 첨가된 작동자로부터 더 높은 포화 수치(saturation level)를 보여준다. Stability of Fur-operator complex depends on the divalent transition metal involved in the complex. Binding affinity of the protein was not altered by the different incubation temperature. However, at higher temperature the repressor showed higher saturation level with the same amount of operator.
