        광어 고음 추출물의 생리활성

        양식 해산어의 효율적인 수급과 소비 방법을 다양화하기 위하여 광어를 이용한 고음 추출물을 제조하여 이의 치료식으로서의 이용가능성을 타진한 결과는 다음과 같다. 광어 고음 추출물들은 rat의 십이지장에 대해 수축활성을 나타냈고, in vivo상태에서 혈압강하작용이 있었다. 또한 이들 추출물들은 rat의 대동맥에 대해 농도 의존적으로 이완활성을 보였다. RM 60 추출물은 MCF7 세포에 대해 항암효과를 나타냈지 만 다른 추출물들은 항암효과와 항균활성을 지니지 않았다. 광어 고음추출물들을 사료로 하여 rat의 혈액성상효과를 측정한 결과 대조군에 비해 HDL-콜레스테롤의 증가 이외에는 현저한 차이를 보이지 않았다. 따라서 이러한 결과들은 광어 고음 추출물에 다양한 생리활성물질들이 포함되어 있다는 것을 제시한다. The pharmacological effects of hydrocooked(110℃, 5 hours) extracts of Bastard halibut have been investigated. All of the hydrocooked extracts showed the measurable contractile effect on the isolated rat duodenum and decreased the normal blood pressure in anesthetized rat. The hydrocooked extracts also exhibited a dose-dependent relaxation on the isolated rat aorta precontracted with phenylephrine. Only RM 60 fraction of these extracts had the cytotoxic effect against MCF7 cell(human breast adenocarcinoma cell line), but the other fractions showed neither antibacterial activity nor antitumor activity. Although fish extracts fed group of rat maintained their original body weight, there were no notable changes in the hematological parameters, except that the levels of high-density lipoprotein was significantly increased. These results suggest that the hydrocooked extracts of bastard halibut may contain a variety of bioactive materials.

        바지락(Ruditapes philippinarum) 아가미로부터 항균 펩타이드의 탐색 및 정제

        This study screened the biological activity of an acidified gill extract of the Manila clam Ruditapes philippinarum in-cluding antimicrobial, hemolytic, membrane permeabilization, and DNA-binding activity, and purified the antimicro-bial material. The acidified gill extract showed potent antimicrobial activity against Bacillus subtilis and Escherichia coli without significant hemolytic activity, but showed no membrane permeabilization or DNA-binding ability. An antimicrobial material was purified from the acidified gill extract using C18 reversed-phase and cation-exchange high-performance liquid chromatography (HPLC). Treatment of the purified material with trypsin completely abolished all of the antibacterial activity against Bacillus subtilis, suggesting that the purified material is a proteinaceous antibiotic. The molecular weight of the purified material was 2571.9 Da, but no primary structural information was obtained due to N-terminal blocking. A future study should confirm the primary structure. Our results suggest that the Manila clam gill contains proteinaceous antibiotics that have a role in first-line defense. This information could be used to better understand the Manila clam innate immune system.

        Bowfin 과 Shark - neuropeptide γ 의 구조 및 수축효과

        The relatioaship between structure and contractile activity was studied on the three neuropeptide γ (mammalian-, bowfin-, shark-NPγ) that were synthesized by the solid-phase method. Circular dichroism spectra showed that mammalian-, bowfin- and sharkneuropeptide γ took an unordered structure in buffer solution and artificial liposome. The intestinal motility response was investigated with guinea-pig ileum, rat duodenum and carp intestine. In case of carp intestine, contractile activity was as follows; bowfin-NPγ>shark-NPγ>mammalian-NPγ. On the other hand, the contractile activity of mammalian-neuropeptide y was more potent than those of bowfin-, shark-neuropeptide γ in the guinea-pig ileum and rat duodenum. These results suggest that NPγ show the species-specific activity.

        The relationship between structure and biological activity was studied on the three neuropeptides (mammalian-, trout- and goldfish-neuropeptide γ) that were syntheized by the solid-phase method. Circular dichroism spectra showed that mammalian-, trout- and goldfish-neuropeptide γ adopted an unordered structure in buffer solution. In the presence of neutral and acidic liposomes, mammalian- and trout-neuropeptide γ also took a random structure. However, goldfish-neuropeptide γ took an α-helical structure in acidic liposomes. The intestinal motility response was investigated with carp intestines, guinea-pig ileums and rat duodenums. In case of carp intestine, contractile activity was as follows : goldfish-neuropeptide γ- trout-neuropeptide γ > mammalian-neuropeptide γ. On the other hand, the contractile activity of mammalian-neuropeptide γ was more potent than trout- and goldfish-neuropeptide γ in the guinea-pig ileums and rat duodenums. These results suggest that neuropeptide γs show the species-specific activity.

        부안산 참굴(Crassostrea gigas) 추출물로부터 피부 상재균에 대한 새로운 항균 펩타이드, cgCAFLP의 탐색 및 정제

        This study was performed to screen the antimicrobial activities of the extract from the Pacific oyster Crassostrea gigas against skin pathogens and to purify the relevant antibacterial peptide. The acidified extract showed potent antibacterial activities against gram-positive and gram-negative bacteria but showed no activity against Candida albicans and no significant cell toxicity. Among acne-causing pathogens, the acidified extract showed potent antibacterial activity only against Staphylococcus aureus, and its antibacterial activity was completely abolished by treatment with trypsin or chymotrypsin, and was inhibited by salt treatment. The acidified extract showed strong DNA-binding ability but did not show bacterial membrane permeabilizing ability. Based on antimicrobial activity screening and cytotoxic effects, a novel antibacterial peptide was purified from the acidified gill extract using solid-phase extraction, cation-exchange, and reversed-phase HPLC. The resulting peptide had a molecular weight of 4800.8 Da and showed partial sequence homology with the carbonic anhydrase 4 (CA4) protein in the hard-shelled mussel. Overall, we purified a novel antibacterial peptide, named cgCAFLP, which is related to carbonic anhydrase 4 (CA4) protein, against skin pathogens. Our results suggest that the Pacific oyster extract could be used as an additive to control some acne-related skin pathogens (S. aureus).

        염증성 피부질환 개선 기능 소재로서 둥근전복(Haliotis discus) 유래 항균펩타이드 Ab4-7의 항염증 효과

        서해안에 서식하는 해양생물인 전복은 항산화 및 항염증 효과가 있으며, 항생제 개발 및 화장품 원료 등 다양한 산업에 활용될 수 있는 잠재력을 가진 자원이다. 본 연구에서 우리는 서해안의 다양한 해양 생물중에서 전복을 선택하였으며, 전복에서 분리한 항균펩타이드(AMP)를 이용하여 항균펩타이드 유도체인 Ab4-7를 규명하고 생리활성을 연구하였다. Ab4-7의 항염증 효능을 확인하기 위해 염증이 유도된 RAW 264.7에 Ab4-7을 처리한 결과, 염증성 사이토카인, TLR4, TNF-α, IL-1β, COX-2, iNOS 등을 억제하고 항산화와 관련된 유전인자인 HO-1의 mRNA 발현을 증가시켰다. Ab4-7의 강력한 항산화 및 항염증 효과를 바탕으로, 염증이 유도된 RAW 264.7 세포에서 Ab4-7이 extrocellular matrix metalloproteinase (ECM) 분해에 관여하여 다양한 염증성 피부질환을 유도하는 유전자인 matrix metalloproteinase (MMPs)를 조절하는 효과를 RT-PCR을 통해 확인하였다. Ab4-7은 강력한 항염증 및 항산화 효과를 가지고 있으며, 항염증 및 항산화 효과를 통한 MMPs 관련 유전인자 조절을 통해 다양한 염증성 피부질환 치료제의 기능성 재료로 활용될 수 있다고 사료된다. Abalone, a marine organism inhabiting the west coast of Korea, has antioxidant and anti-inflammatory effects, and is a resource with potential to be used in various industries such as antibiotic development and cosmetic raw materials. In this study, we chose abalone among various marine lives on the west coast. Antibacterial peptide (AMP) was separated from abalone and its derivative Ab4-7 was identified and its physiological activity was studied. The treatment of Ab4-7 in inflammatory RAW 264.7 to check the anti-inflammatory efficacy nhibited inflammatory cytokines, TLR4, TNF-α, IL-1β, COX-2, and iNOS and increased mRNA manifestation of HO-1, genes related to antioxidants. Based on the strong antioxidant and anti-inflammatory effects of Ab4-7, the effects of Ab4-7 in the inflammatory RAW 264.7 cells were identified through RT-PCR, which regulates the gene Matrix Metalloproteinase (MMPs) that induces a variety of inflammatory skin diseases by engaging in the decomposition of the extrocellular matrix metalloproteinase (ECM). Taken together, it is concluded that Ab4-7 has a powerful anti-inflammatory and antioxidant effect and can be used as a functional material for various inflammatory skin disease treatments by controlling the genes associated with matrix metalloproteinase (MMPs).

        Synthesis and Biological Activities of Myomodulin E and its Analogs

        군소인 Aplysia kurodai의 중추신경절로부터 발견된 myomodulin A (MMA, PMSMLRLamide)와 myomodulin E (MME, GLQMLRLamide)는 Mytilus edulis의 anterior byssus retractor muscle (ABRM)을 활성측정시스템으로 사용하여 정제되었다. . 정제된 MMA와 MME는 연체동물에서 발견된 myomodulin 계열의 펩타이드와 동일한 일차구조를 지닌다. MME의 구조와 활성간의 상관관계를 알아보기 위해서 MME, 유도체 및 다른 신경성 펩타이드들을 합성하였다. MME의 유도체인 Des[Gly¹]-MME, Des[Gly¹,Leu²]-MME 및 Des[Gly¹, Leu²,Gln³]-MME의 일차구조는 각각 LQMLRLamide, QMLRLamide 및 MLRLamide이다. 합성 물질들을 사용하여 ABRM에 대한 phasic -contraction을 측정하였다. MME는 1×10<SUP>-9</SUP> M 또는 더 높은 농도에서 ABRM의 phasic contraction을 저해하였다. 또한 MME는 1×10<SUP>-8</SUP> M에서 catch-tension에 대해 이완활성을 나타내었다. 합성 펩타이드들을 사용하여 Africa giant snail, Achatina fulica의 소낭과 penial retractor muscle에 대해서도 활성을 측정하였다. MME와 유도체들은 소낭에 대해서는 수축반응을 보였지만, penial retractor muscle에 대해서는 이완 활성을 나타내었다. 이러한 결과들은 MME와 그 유도체들은 연체동물의 다양한 조직에 대해 조절 효과를 가지고 있다는 것을 의미한다. 본 연구는 생체 내에서 발생하는 신경 및 circuit의 변화를 조절하는 작용 연구에 대한 기본적인 자료가 될 것이다. Previous work has characterized myomodulin A (MMA, PMSMLRLamide) and myomodulin E (MME, GLQMLRLamide) purified from the central nervous systems of the sea hare, Aplysia Kurodai, using the anterior byssus retractor muscle (ABRM) of the mussel, Mytilus edulis. The amino acid sequences of MMA and MME were the same as those of the myomodulin family peptide found in other mollusks. In this study, we synthesized MME, its derivatives, and other neuropeptides to investigate the relationship between the structure and biological activity of MME. The primary structures of MME’s derivatives, Des[Gly¹]-MME, Des[Gly¹,Leu²]-MME, and Des[Gly¹,Leu²,Gln³]-MME, were LQMLRLamide, QMLRLamide, and MLRLamide, respectively. MMA and synthetic peptides were tested on ABRM in M. edulis as well as muscle preparations in Achatina fulica. MME displayed an inhibitory effect on phasic contraction of the ABRM at 1×10<SUP>-9</SUP> M or higher. MME also had a relaxing effect on the catch-tension of AMRM at 1×10<SUP>-8</SUP> M. Both MMA and its analogs stimulated a contractile response on the crop and relaxed the catch-relaxing response on the penial retractor muscle of A. fulica. These results suggest that MME and its analogs have modulatory effects on various muscles of mollusks. This study has also laid the groundwork for future neural and circuit modulation studies during animal behavioral changes.

        진주담치(Mytilus edulis) 추출물의 항균활성 및 단백질 분해효소에 대한 안정성 탐색

        This study was performed to screen the antimicrobial activities and proteolytic enzyme stability of the acidified extract of the Blue mussel Mytilus edulis. The acidified extract showed potent antimicrobial activities against Grampositive bacteria, Bacillus subtilis, and Gram-negative bacteria, Escherichia coli D31, but had no activity against Candida albicans. Treatment of extract with trypsin completely abolished all or significant antibacterial activity against the tested bacteria, but slightly decreased antimicrobial activity against B. subtilis, and treatment of extract with chymotrypsin retained almost antibacterial activity against the tested bacteria except for E. coli D31. To confirm the additional enzyme stability of the extract, antimicrobial activity of the extract was tested after treated with several enzymes. Enzymes treated extract showed potent antimicrobial activity against B. subtilis and its activity was also retained for 5 h after trypsin treatments. Non-proteinaceous materials in the acidified extract also showed strong DNA-binding ability but did not show bacterial membrane permeabilizing ability. All our results indicate that mussel extract might contain the proteinaceous or non-proteinaceous antibacterial materials target not bacterial membrane but intracellular components. These results could be used to develop mussel extract as an additive for the improvement of stability or antimicrobial activity of antibiotics against specific bacteria.

        군소 ( Aplysia kurodai ) 의 중추신경계로부터 myomodulin A 와 E 의 정제

        군소의 중추신경계로부터 myomodulin A (MMA)과 myomodlin E (MME)의 정제에 대해 보고하고자 한다. 500마리 군소의 중추신경계를 산조건으로 추출한 후 4종류의 HPLC column system에 적용하여 두가지 신경성 펩타이드를 정제하였다. HPLC 분리과정에서 얻어진 모든 분획의 활성을 조사하기 위하여 bioassay system으로 진주담치 (Mytilis edulis)의 ABRM을 이용하였다. 최종 정제된 신경성 펩타이드는 Edman 분해법을 이용한 아미노산 서열 분석기, 질량분석기와 화학합성을 통해 완전한 일차구조를 밝힐 수 있었다. 정제한 MMA와 MME의 일차구조는 각각 Pro-Met-Ser-Met-Leu-Arg-Leu-NH₂(847.41 Da)과 Gly-Leu-Gln-Met-Leu-Arg-Leu-NH₂, (830.50 Da) 이었다. 또한 이들 합성 펩타이드는 진주담치의 ABRM에 대해 phasic contration의 조절활성을 나타내었다. This paper reports the purification of myomodulin A (MMA) and myomodulin E (MME) from the sea hare. The central nervous systems of 500 sea hare were extracted in an acidified solvent, after which four HPLC column systems were used to obtain pure peptides. The phasic contraction bioassay using a Mytilus edulis anterior byssus retractor muscle (ABRM) was applied to monitor all collected fractions. The pure peptides were submitted to Edman degradation based automated microsequencing. Mass spectrometry and chemical synthesis confirmed the sequence. The primary structures of MMA and MME were Pro-Met-Ser-Met-Leu-Arg-Leu-NH₂(847.41 Da) and Gly-Leu-Gln-Met-Leu-Arg-Leu-NH₂(830.50 Da), respectively. Synthetic peptides showed a modulating activity of phasic contraction in the ABRM of Mytilus edulis.
