        Propylthiouracil 및 Thyroxine의 처리(處理)가 흰쥐의 갑상선기능(甲狀腺機能)과 체성장(體成長)에 미치는 영향(影響)

        본(本) 실험(實驗)은 thyroxine 및 propylthiouracil의 투여(投與)에 의(依)한 갑상선(甲狀腺)의 기능변화(機能變化)와 아울러 체성장(體成長)에 미치는 영향(影響)을 구명(究明)코저 실시(實施)하였는 바, 25일령(日齡)의 암 흰쥐를 갑상선척출군(甲狀腺剔出群), propylthiouracil 처리군(處理群), Thyroxine 처리군(處理群) 및 정상대조군(正常對照群)의 사개군(四個群)으로 나누어 배치(配置)하고 각군(各群)을 처리후(處理後) 시간경과(時間經過)에 따라 도살(屠殺)하여 갑상선(甲狀腺)의 중량측정(重量測定) 및 조직학적(組織學的) 검색(檢索)과 아울러 체중(體重)의 변화(變化)를 조사(調査)하였다. 1. 갑상선(甲狀腺)의 중량(重量)은 모든 관찰시간(觀察時間)에서 비교군간(比較群間)에 유의성(有意性)이 인정(認定)되었는데 PTU 처리군(處理群)은 정상대조군(正常對照群)에 비(比)하여 높은 중량(重量)을 나타냈고, Thyroxine 처리군(處理群)은 낮은 값을 나타냈다. 2. 갑상선(甲狀腺)의 조직상(組織像)은 PTU 처리군(處理群)에서는 처리(處理) 1주후(週後)부터 여포상피세포(濾胞上皮細胞)가 증식(增殖) 비대(肥大)를 동반(同伴)하는 원주화현상(圓柱化現象)을 나타내기 시작하여 실험기간(實驗期間)이 경과(經過)될 수록 더욱 진행(進行)되는 조직소견(組織所見)을 보였으며, Thyroxine 처리군(處理群)에서는 여포상피세포(濾胞上皮細胞)가 도평화(屠平化)되는 퇴행성(退行性) 변화(變化)가 인지(認知)되었다. 3. 체중(體重)은 처리(處理) 2주후(週後)부터 비교군간(比較群間)에 유의성(有意性)이 인정(認定)되는 변화(變化)를 보였는데 PTU 처리군(處理群)과 갑상선척출군(甲狀腺剔出群)은 정상대조군(正常對照群)에 비(比)하여 낮은 값을 나타냈고 Thyroxine 처리군(處理群)은 높은 체중치(體重値)를 보였다. 그러나, 다중검정결과(多重檢定結果) PTU 처리군(處理群)과 갑상선척출군(甲狀腺剔出群) 상호간(相互間)에는 유의성(有意性)이 인정(認定)되지 않았다. This study was conducted to find out the effects of propylthiouracil and thyroxine on thyroid function and body growth in female rats. One hundred and forty-four female rats (Wistar-imamichi albino rats) of 25 days old were divided into 4 groups; thyroidectomy (Thx), propylthiouracil (PTU), thyroxine (Thyro.)and control (Cont.) groups. Thirty-six rats were allotted to each group, and changes of body weights were weekly checked. In addition, 6 rats in each group were sacrificed at 1, 2, 3, 5 and 6 weeks after treatments with time elapse for investigating changes of thyroid weights and tissues. The results obtained were as follows: 1. The weights of thyroid gland showed significant differences among all the compared groups at all observation times. The weights of thyroid glands in PTU group were higher than those in control group, but those in the Thyro. group were lower than those in control group. 2. In the histological changes of thyroid glands in the PTU group, the follicle epithelium showed columnar cells following hype trophy and hyperplasia from 1 week after treatment. The follicle epithelium in the Thyro. group were recognized inactive showing squamous cells. 3. The body weights showed significant differences among the compared groups from 2 weeks after treatment. The body weights decreased significantly in PTU and Thx. groups, while those in Thyro. group increased significantly in comparison with those in control group. No significant difference in body weight was noted between PTU and Thx. groups.

        한국재래산양정자 (韓國在來山羊精子) 의 액상 및 동결보존에 관한 연구

        This study was carried out to develop the preservation methods of fresh and deep frozen Korean native goat spermatozoa and investigate the farrowing rate of Korean native goat does inseminated with frozen-thawed semen. The results obtained were as follow. 1. Motility and NAR acrosome of fresh spermatozoa during incubation were higher at 22℃ than at 5℃ or 37℃(p $lt;0.01). The percent LAC was inversely related to the percent NAR with regard to the incubation periods of these same three treatments. 2. Motility and NAR acrosome of spermatozoa diluted to 1:4 during incubation were higher at 22℃ than at 5℃ or 37℃. Especially incubation for 0.5, 3 and 6 h in the 5℃ had a marked detrimental effect on motility and NAR acrosome of spermatozoa. 3. Motility and NAR acrosome of spermatozoa during incubation were higher for samples diluted 1:1, 1:2, or 1:4 than for samples diluted 1:6(p $lt;0.01). 4. Motility and NAR acrosome of post-thaw spermatozoa were higher at freezing rate of 12℃/min or 24℃/min than at freezing rate of 1℃/min(p $lt;0.01). 5. Motility and NAR acrosome of post-thaw spermatozoa at freezing rate of 12℃/min were higher at 9% glycerol than at 6% or 3%(p $lt;0.01). 6. After thawing of frozen spermatozoa in a 37℃ water bath for 10 sec, incubation temperatures of 12℃, 22℃ or 37℃ for 2h did not show any differences on motility and NAR acrosome. 7. Farrowing rate of Korean native goat does inseminated twice with frozen-thawed semen was 25.0%. Mean kid weight at birth was l.9kg.

        돼지의 혈청 progesterone 수준에 의한 임신진단에 의한 연구

        The study was carried out to investigate the changes of hormone levels in blood serum during the estrous cycle and the accuracy of pregnancy diagnosis by estimation of plasma progesterone at 20 to 23 days after breeding in sows. The progesterone and estradiol-17 R from blood serum samples were assayed by radioimmunoassay method. The results of this study were summarized as follows: I.The progesterone concentrations in blood serum during the estrous cycle began to decline rapidly at 2 days before estrus, and thereafter decreased to 0.7±0.18ng/ml at on the day of estrus, and thereafter reached a peak mean level of 13.1±2.15ng/ml at 15 days after estrus. 2.The estradiol-17β concentrations in blood serum during the estrous cycle showed a peak mean level of 14.6±3.24pg/ml at 18 days after estrus, and decreased from 9.8 pg/ml to 14.2 pg/ml at luteal phase. 3.During 20 to 23 days after artificial insemination, the accuracy of pregnancy diagnosis from the blood serum progesterone levels were 96.4% for non-pregnant sows($lt;2.7ng/ml), and 86.1% for pregnant sows( 3.6ng/ml). The average overall accuracy of pregnancy prediction for non-pregnant and pregnant sows was 91.3%.

        EGF와 IGF-I의 첨가배양이 돼지 미성숙 난포란의 체외성숙과 배발달에 미치는 영향

        본 연구는 돼지 난포란의 체외배양액과 배발달 배양액에 성장인자인 EGF와 IGF-Ⅰ을 각각 0, 1, 5 및 10 ng/㎖ 첨가배양함으로서 돼지 난포란의 체외성숙과 체외배발달에 미치는 영향을 구명하고자 실시하였다. 본 연구에서 얻어진 결과를 요약하면 다음과 같다. 1. 돼지 난포란을 체외성숙배양액인 NCSU-23 배양액에 EGF와 IGF-Ⅰ을 각각 0, 1, 5 및 10 ng/㎖ 첨가배양하여 성숙을 유기한 다음, 체외수정을 실시한 결과, 모든 처리구에서 핵성숙률, 정자침투율, 웅성전핵형성률, 다정자 침입률 및 평균침입정자수에서 유의적인 차이가 인정되지 않았다. 2. 체외수정을 실시한 후 , 배발달배양액인 NCSU-23에 7일간 배양한 결과, 배반포형성률은 EGF첨가군이 각각 11.2±1.5%, 15.0±0.8%, 16.8±2.8% 및 21.4±2.0%로서 10 ng/㎖의 첨가군이 유의적(P<0.05)으로 높은 결과를 나타냈으며, IGF-Ⅰ첨가군도 10.7±1.4%, 12.8±2.0%, 16.0±2.5% 및 21.9±2.4%로서 10 ng/㎖의 첨가군이 유의적으로 높은 결과를 나타냈다. 또한, 총세포수에 있어서도 EGF첨가군이 22.8±3.7개, 25.7±5.5개, 26.0±4.2개 및 35.1±4.7개, IGF-Ⅰ첨가군은 21.5±3.7개, 25.2±2.8개, 26.2±2.9개 및 33.2±3.6개로서 각각 10 ng/㎖ 첨가군이 유의적(P<0.05)으로 높은 결과를 나타냈다. 3. 돼지 난포란을 체외성숙 기본배양액인 함유된 NCSU-23 배양액에 44시간동안 체외성숙을 유기한 다음, 체외 수정용배양액인 mTBM에 정자와 같이 6시간동안 공배양함으로서 체외수정을 유기한 후, 배발달배양액인 NCSU-23에 EGF와 IGF-Ⅰ을 각각 0, 1, 5 및 10 ng/㎖ 첨가하여 7일간 배양한 결과, 배반포기 도달률은 EGF첨가군에서 각각 14.0±1.7개, 16.2±1.4개, 16.9±1.2개, 23.1±1.6개로서 10 ng/㎖ 첨가군이 유의적으로 높은결과를 나타냈고, IGF-Ⅰ첨가군도 각각 13.6±1.7개, 15.7±4.5개, 16.0±0.2개 및 25.0±0.8개로 10ng/㎖군이 유의적(P<0.05)으로 높은 결과를 나타냈으며, 총세포수에 있어서는 EGF첨가군이 각각 21.8±2.9개, 25.2±2.8개, 39.7±2.7개 및 46.2±3.6개로 10 ng/㎖첨가군이 유의적으로 높은 결과를 나타냈고, IGF-Ⅰ첨가군도 20.7±2.9개, 26.2±2.9개, 24.6±2.4개 및 46.1±3.5개로 10 ng/㎖ 첨가군이 유의적(P<0.05)으로 높은 결과를 나타냈다. 이상의 결과를 종합해 볼 때, EGF와 IGF-Ⅰ은 돼지 난포란의 체외성숙과 배발달과정에 영향을 미치며 특히, 10 ng/㎖의 농도에서 효과적인 것으로 조사되었다. The present study was carried out to examine the effect of EGF and IGF-Ⅰ in vitro maturation (IVM) of porcine oocytes and development of porcine IVM/IVF embryos. The results were summarized as follows : 1. The rates of nuclear maturation, penetrated oocytes, pronuclear formation, polyspermic oocytes and mean numbers of the penetrated sperm were not different in NCSU-23 maturation medium with 0, 1, 5 and 10 ng/㎖ EGF and IGF-Ⅰ (P>0.05). 2. The rates of blastocyst formation at day 7 after in vitro fertilization in 0, 1, 5 and 10 ng/㎖ EGF groups were 11.2±1.5%, 15.0±8.3%, 16.8±2.8% and 21.4±2.0%, also 0, 1, 5 and 10 ng/㎖ IGF-Ⅰ groups were 11.2±1.5%, 15.0±8.3%, 16.8±2.8% and 21.4±2.0%, respectively. In the total cells case, EGF groups were 22.8±3.7, 25.7±5.5, 26.0±4.2 and 35.1±4.7, also IGF-Ⅰ groups were 21.5±3.7, 25.2±2.8, 26.2±2.9 and 33.2±3.6, respectively. Both 10 ng/㎖ EGF group and 10 ng/㎖ IGF-Ⅰ group were significantly higher than those of other treatment groups (P<0.05). 3. The rates of blastocyst formation at day 7 in the NCSU23 culture medium of porcine IVF-produced embryos with 0, 1, 5, and 10 ng/㎖ EGF groups were 14.0±1.7%, 16.2±1.4%, 16.9±1.2% and 23.1±1.6%, also 0, 1, 5, 10 ng/㎖ IGF-Ⅰ groups were 13.6±1.7, 15.7±4.5, 16.0±0.2 and 25.0±0.8, respectively. And in the total cells case, EGF grups were 21.8±2.9, 25.2±2.8, 39.7±2.7 and 46.2±3.6, also IGF-Ⅰ groups were 20.7±2.9, 26.2±2.9, 24.6±2.4 and 46.1±3.5, respectively. Both 10 ng/㎖ EGF group and 10 ng/㎖ IGF-Ⅰ group were significantly higher than those of any other treatment groups (P<0.05). In conclusion, these results suggested that the addition of 10 ng/㎖ EGF and IGF-Ⅰ were effective on the blastocyst formation and total cells of blastocysts.
