        유전지표를 활용한 사상체질 분류모델

        Objectives Genome-wide association studies(GWAS) is a useful method to identify genetic associations for various phenotypes. The purpose of this study was to develop predictive models for Sasang constitution types using genetic factors. Methods The genotypes of the 1,999 subjects was performed using Axiom Precision Medicine Research Array (PMRA) by Life Technologies. All participants were prescribed Sasang Constitution-specific herbal remedies for the treatment, and showed improvement of original symptoms as confirmed by Korean medicine doctor. The genotypes were imputed by using the IMPUTE program. Association analysis was conducted using a logistic regression model to discover Single Nucleotide Polymorphism (SNP), adjusting for age, sex, and BMI. Results & Conclusions We developed models to predict Korean medicine constitution types using identified genectic factors and sex, age, BMI using Random Forest (RF), Support Vector Machine (SVM), and Neural Network (NN). Each maximum Area Under the Curve (AUC) of Teaeum, Soeum, Soyang is 0.894, 0.868, 0.767, respectively. Each AUC of the models increased by 6~17% more than that of models except for genetic factors. By developing the predictive models, we confirmed usefulness of genetic factors related with types. It demonstrates a mechanism for more accurate prediction through genetic factors related with type.

        This study verified the degree of hair injury prevention effect of hair mist manufactured with Taraxaci Herba when it is applied as pre treatment agent at the time of permanent wave treatment with cysteine composition. In order to verify the effect of Taraxaci Herba hair mist, this study conducted experiments by classifying them into C1 in which only permanent wave was treated on virgin hair, C2 in which pre treatment with hair mist base was followed by permanent wave treatment and C3 in which pre treatment with Taraxaci Herba hair mist was followed by permanent wave treatment. As major ingredients of permanent wave composition, cysteine was used for 1st composition and sodium bromated was used for 2nd composition. As the result of measurement of hair thickness, C3``s hair was the thickest while C1``s hair was the thinnest, and as the result of measurement on the degree of hair damage with observation on minute structures of cuticles and Methylene blue dyeing method, C3``s damage was the lightest while C1``s hair damage was the most severe. Judging from the fact that hair mist base does not greatly reduce the damage when compared with Taraxaci Herba hair mist, it was confirmed that hair mist base only helps useful ingredients of Taraxaci Herba effectively penetrate into the inside of hair but does not have other independent functions. Therefore, it was verified that pre treatment with Taraxaci Herba hair mist at the time of permanent wave helps maintain healthy hair.

      • 오가피 메탄올 추출물의 기능성 화장품 소재 가능성 연구

        With the recent well-being craze and the growing preference for organic foods, various plant resources with functional effects such as antioxidant, antibacterial, anti-inflammatory, and whitening properties are being developed, in order to produce a functional, natural cosmetic material for cosmetics. The present researcher studied Acanthopanax sessiliflorus seeman as a material for functional cosmetics and came up with the following findings. The extraction yield of Acanthopanax sessiliflorus seeman methanol was found to be 6.95%. The total flavonoid content was found to be 3.393 ± 5.48 ㎎(TAE)/g. In the DPPH radical scavenging activity measurement, the scavenging activity was over 91.49% at a concentration of over 0.25%, which was on the same level as BHT, an existing antioxidant. Measurements of the cytotoxicity of the NIH/3T3 cell revealed that there was no cytotoxicity but the growth of the cell increases proportionally according to the treatment concentration. After the scavenging activity was measured for the nitrite by causing a reaction in pH of 1.2, it was found that there was similar scavenging activity to that of BHT, as with the scavenging activity of the DPPH radical. Concerning Staphylococus aureus and staphylococcus epidermidis, when the antibacterial ability of the Acanthopanax sessiliflorus seeman methanol extract was measured, it was found that the clear zone increased proportionally according to the increase in the concentration of the extract. From this, it was confirmed that antibacterial activity is dependent on the concentration. From the aforementioned research findings, the Acanthopanax sessiliflorus seeman methanol extract is deemed to have potential as a functional cosmetic material with antioxidant, anti-inflammatory and antibacterial properties.

        The present experiments were carried out to develop new antioxidants from methanol extract of Polygoni mulitiflori Radix concerning total polyphenol and flavonoid content, scavenging activities on DPPH and ABTS radicals, SOD-like activities. Methanol extracts from Polygoni mulitiflori Radix showed higher level of total polyphenol content, and the flavonoid content was the highest. And showed higher level of scavenging activities on DPPH and ABTS. However, methanol extracts from Polygoni mulitiflori Radix showed SOD-like radical activity was the low.

        퍼머넌트 웨이브 시술시 포공영 헤어미스트 전처리에 따른 모발보호효과

        Recently, various studies are being conducted using natural materials and this study manufactured and applied hair mist using Taraxaci Herba extract to hair. In order to verify the effect of manufactured Taraxaci Herba hair mist, this study experimented by classifying objects into C1 which only applied permanent wave to virgin hair, C2 which preprocessed hair mist base before applying permanent wave and C3 which preprocessed Taraxaci Herba hair mist before applying permanent wave. As permanent wave solutions, first, thioglycollic acid was used as reducing agent and, second, sodium bromate was used as oxidizing agent. As the result of measurement on wave efficiency, the efficiency of C3 proved the highest while that of C1turned out to be lowest. As the result of measurement of tensile strength and degree of hair damage using Methylene blue dying method and of observation on cuticle and structure of minute hair, degree of hair damage of C1 proved to be the most severe. Given that hair mist base did not reduce damage to a remarkable extent when compared with Taraxaci Herba hair mist, antioxydant function of Taraxaci Herba extract is presumed to have reduced the damage of the hair. Therefore, it was confirmed that preprocessing of Taraxaci Herba hair mist at the time of permanent hair treatment can provide elastic curl while maintaining healthy state of hair.

        적하수오 추출물 크림의 SD-rat 두피손상 완화 및 안전성 검사

        Oxidized hairdyes which are frequently used in dyeing cause abnormal scalp and contact dermatitis. And they even do hair loss in severe cases. Pleuropterus multflorus extract cream was used to relieve damaged scalp due to frequent use of oxidized hairdyes. First, the result which carried out the cytotoxicity test (mtt-assay) to check the safety of Pleuropterus multflorus extract has found that there is no cytotoxicity on concentration of less than 100 ?g/mL. The result which observed the effect by applying polygonum multiflorum extract cream which was made based on them three times per week to the SD-rat back part that caused scalp damage as the oxidized hairdye is as follow: We applied Pleuropterus multflorus cream to SD-rat whose scalp was damaged by oxidized hairdye. In contrary to control group, erythema and edema appeared in first 2~4 weeks which completely alleviated in week 6~8. We also conducted a patch test on 20 people to investigate allergic reaction of Pleuropterus multflorus cream. As a result, P.I.I was 0.072, which is equivalent to non-irritation reaction, thus proving rare existence of skin irritation. Therefore, the study confirmed safety of Pleuropterus multflorus cream and alleviation effect on scalp damage through basic level test.

        염색 시술 시 포공영 샴푸 처리에 따른 모발 손상도 및 색상도 변화 연구

        This study aimed to produce a shampoo with using extract of Taraxacum Herba and to develop hair care products. For this, changes in the degree of hair damage and hair color were observed by washing dyed hair with an oxidized hair dye to verify the efficacy of the Taraxacum Herba shampoo. For accurate verification, a comparative analysis was conducted using various shampoos on the market. After dyeing test hair with the oxidized hair dye, they were washed with each shampoo. The shampoos manufactured by ‘A’ were divided into regular shampoo, color care shampoo, conditioning shampoo and Taraxacum Herba shampoo and comparatively analyzed. In terms of tensile strength, the hair washed with the Taraxacum Herba shampoo was the greatest, followed by the conditioning shampoo, color care shampoo and regular shampoo. According to the analysis of hair components, in terms of sulfur contents, hair washed with the Taraxacum Herba shampoo was the highest, followed by the conditioning shampoo, color care shampoo and regular shampoo. In terms of hair damage measured using the MB dye method, hair washed with the Taraxacum Herba shampoo was the weakest, followed by the conditioning shampoo, color care shampoo and regular shampoo. In terms of the degree of hair color in which L* values were comparatively analyzed, hair washed with the regular shampoo was the brightest, followed by the conditioning shampoo. The color care shampoo and Taraxacum Herba shampoo were less bright, relatively. The said results found that when the Taraxacum Herba shampoo was used after hair dyeing, hair damage and loss of hair color were minimized. Therefore, it was shown that the Taraxacum Herba shampoo has a positive effect on dyeing properties as well.

        Histological Comparison Study with Primo Node and Immature Liver Tissue on Liver Surface in Rat

        목적 : 프리모 모드와 미성숙 간조직에 대한 조직학적 특성을 비교하고자 하였다. 방법 : 흰쥐에서 간절제한 후, 간장기 표면에서 관찰되는 프리모 모드와 그와 유사한 미성숙 간조직을 H&E, Oil red O, Masson trichrome and van Gieson 염색방법을 통해서 비교 연구하였다. 결과 : 상기 실험 연구를 진행한 결과, 다음과 같은 결론을 얻었다. 1. 조직의 일반적인 특징을 관찰할 수 있는 H&E 염색결과, 프리모 노드에서는 많은 수의 조그마한 동(sinus)이 관찰되었고 적혈구나 영양을 공급하는 혈관의 분포는 관찰되지 않았다. 이와 반대로 미성숙 간조직에서는 동이 관찰되지 않았으며 혈관의 분포와 적혈구가 관찰되었다. 2. 양 조직의 지방성분의 유무를 관찰하기 위한 Oil red O 염색결과, 프리모 노드에서는 지방성분이 관찰되지 않았으나 미성숙 간조직에서는 관찰되었다. 3. 양 조직의 콜라겐성분의 유무를 관찰하기 위한 Masson trichrome and van Gieson 염색결과, 프리모 노드에서는 약간의 콜라겐 성분이 관찰되었으나 elastic 성분은 관찰되지 않았으며, 미성숙 간조직에서는 콜라겐 성분과 elastic 성분이 관찰되지 않았다. 결론 : 상기 결과는 프리모 노드와 간조직과는 조직학적 성질이 다른 것으로 사료되며, 특히 콜라겐 성분의 적은 결과는 프리모 노드가 불규칙한 형태를 이루고 있는 이유에 대한 실마리를 제공하였다. 이러한 결과는 프리모 노드의 특성에 대한 기본 자료로 활용될 수 있을 것이다.

        Permanent wave after the procedure due to the addition of rosehip extract hair dyed hair during treatment of physical and chemical changes measured by varying the concentration of rosehip extract added to permanent wave 1 solution to wave treatment and efficiency, thickness, moisture content, tensile strength and elongation, hair cuticle observed, the rate of amino acid composition, was a result of analyzing the colors, dyed hair damage and softening rosehip extract added at the same time, to increase the rate of formation of wave. thioglycolate and cysteine Compared wave formation rate thioglycolate is slightly higher but the hair damage was somewhat lower cysteine. Rosehip extract concentration of more appropriate by setting a more effective hair treatment products and methods to study the basis of these results, it can be regarded as an important task.

        천화분 약침액의 A549 폐암 세포주에서 apoptosis 유발효과

        Objectives: In order to confirm the anti-cancer effect of Trichosanthes kirilowii pharmacopuncture fluid, this study was proceeded. Methods: A549 lung cancer cells were cultured to be treated by Trichosanthes kirilowii pharmacopuncture fluid as dose dependent manner for 72 hours. And then the cell viability, nucleus fragmentaion, p21 and p53 protein expression, Bcl-2 and Bax protein expression, procaspase-3 PARP protein expression. Results: 1. Trichosanthes kirilowii pharmacopuncture fluid decrease A549 cell viability as dose dependent manner. 2. Trichosanthes kirilowii pharmacopuncture fluid induced the nucleus fragmentation in A549 lung cancer cells as dose dependent manner. 3. Trichosanthes kirilowii pharmacopuncture fluid increase the p21 and p53 protein expression. 4. Trichosanthes kirilowii pharmacopuncture fluid decrease the Bcl-2 protein expression but cannot affect the Bax protein expression. 5. Trichosanthes kirilowii pharmacopuncture fluid increase the activation of caspase-3 and PARP protein. Conclusions: As the above results, it was conclused the Trichosanthes kirilowii pharmacopuncture fluid had the anti-cancer effects to induce apoptosis.
