        진피 플라보노이드 생물전환 균주 분리 및 췌장 베타세포에 대한 산화적 손상 억제 효과

        In this study, the optimum conditions of fermentation were determined by isolating the microorganisms with the ability to bioconvert the Citrus peel flavonoid, and the effect of the fermented Citrus peel extract which was bioconverted on the oxidative damage of HIT-T15 cell was investigated. The Aureobasidium pullulans Y-12 was isolated and identified with the strains having bioconversion activity. The fermentation conditions for bioconversion activity were confirmed to be optimal when culturing for three days at 25℃, 150 rpm in a culture medium containing 5% Citrus peel power and 0.8% casitone. As a result of bioconversion, 32.8 mg/g and 21.5 mg/g of naringenin and hesperetin, which were aglycone flavones, were produced respectively. Also in the flavonoid content, it was confirmed that FCP produced 154.8 mg/g while CP produced 33.7 mg/g, thus producing more by approximately 4.6 times. As a result of treating FCP and CP after inducing the oxidative damage for HIT-T15 cell by treating the deoxy-D-ribose with IC50 (38 mM) concentration, the surviving rate was recovered to 90% for FCP treatments in the 0.01 mg/mL concentration and for CP treatments in the 0.025 mg/mL concentration. Also in the insulin secretion rate, FCP treatments increased by 206% and CP treatments by 132% when treated in the 0.1 mg/mL concentration. As the bioconverted FCP can inhibit the oxidative damage of HIT-T15 cell in the low concentration, it was considered its usability as the functional material for prevention of the type 2 diabetes.

        고지방식이를 섭취한 흰쥐에서 진피 에탄올추출물이 혈청지질 및 체지방에 미치는 영향

        In this work, the major flavonoid content of citrus peel ethanol extract and the changes in the serum lipid and body fat of high-fat-diet-fed rats were examined. The subjects were divided into four groups: the normal, control, O1 (0.1% citrus peel ethanol extract), and O5 (0.5% citrus peel ethanol extract) groups. The experimental groups (O1 and O5) were fed a high-fat diet mixed with 0.1 and 0.5% citrus peel ethanol extract, respectively, for 10 weeks. The results of the analysis showed that the main ingredients of the flavonoid in the citrus peel ethanol extract (60 brix) were 38.21±1.37 mg/g hesperidin and 15.02±0.48 mg/g narirutin. With regard to the changes in the serum total-cholesterol level, compared with the control group`s 79.1±4.6 mg/dL, the serum total-cholesterol levels of the O1 and O5 groups were significantly lowered to 64.6±5.4 and 59.5±4.7 mg/dL, respectively. As for the changes in the LDL-cholesterol level, compared with the control group`s 18.4±3.7 mg/dL, the LDL-cholesterol levels of the O1 and O5 groups were significantly lowered to 13.0±3.6 and 13.0±1.9 mg/dL, respectively. For the changes in the serum triglyceride level, compared with the control group`s 74.5±13.2 mg/dL, the serum triglyceride level of the O5 group was significantly lowered to 48.6±11.2 mg/dL. Finally, with regard to the changes in body fat, compared with the control group, the O5 group showed a significant decrease. In conclusion, it was observed that citrus peel ethanol extract was effective in lowering the total-cholesterol, LDL-cholesterol, and triglyceride levels of high-fat-diet-fed rats.

        진피 에탄올 추출물이 Alloxan에 의해 유도된 HIT-T15 세포의 산화적 손상에 미치는 영향

        본 연구는 진피 에탄올 추출물의 alloxan에 의해 유도된 HIT-T15 세포의 산화적 손상으로부터 세포 생존율 및 인슐린분비 조절능에 대해 조사하였다. 진피 에탄올 추출물의 총 폴리페놀 함량과 총 플라보노이드 함량을 측정한 결과, 각각 57.00±2.91 ㎎/g, 8.11±2.83 ㎎/g으로 나타났으며 alloxan 처리 농도가 증가됨에 따라 HIT-T15 세포의 생존율은 감소하였으며, alloxan 11.58 mM에서 약 50%의 세포독성이 일어남을 확인하였다. 동결건조 된 진피 에탄올 추출물(CP-Et) 0.125~0.75 ㎎/mL 농도에서 무처리구의 세포생존율은 100.90±1.51~97.56±0.73%로 나타나 HIT-T15 세포에 대한 CP-Et의 처리 농도는 0.125~0.75 ㎎/mL로 결정하였다. 0.125, 0.25, 0.5 ㎎/mL 농도의 CP-Et를 처리한 결과, alloxan에 의해 유도된 산화적 세포손상으로부터 세포생존율이 각각 80.52±3.29, 74.17±6.75, 67.53±5.8%로 나타나 유의성 있게 보호되어짐을 확인하였다. CP-Et를 처리한 HIT-T15 세포에서는 산화적 손상에 대한 세포 생존율과는 다르게 0.125 ㎎/mL 농도에서 116.93±2.11 ㎍/㎎로 인슐린 분비가 대조구와 비교 시 유의적으로 증가되었음을 확인 할 수 있었다. 따라서 진피 에탄올 추출물에 의한 항산화적 방어로 2형 당뇨의 β-cell 양의 감소와 인슐린 저항성에 대한 개선이 가능함을 제시하며, 그 작용 기작에 대해서는 차후 더 많은 연구가 필요하다 하겠다. This study was carried out to investigate the effect of ethanol extract from citrus peels (CP-Et) against the alloxan-induced oxidative damage on HIT-T15, Hamster pancreatic β-cell. Total polyphenol and flavonoid contents in CP-Et were 57.00±2.91 ㎎/g and 8.11±2.83 ㎎/g, respectively. Cell toxicity on HIT-T15 by CP-Et (0.125~0.75 ㎎/mL) was not observed. CP-Et (0.125 ㎎/mL) increased cell proliferation rate of HIT-T15, which was treated alloxan (IC50=11.58 mM) (cell viability=80.52±3.29% of normal cell, p<0.05). In comparison with insulin secretion of oxidative damaged HIT-T15, 1.5 fold (116.93±2.11 μg/㎎ protein) was increased by treatment CP-Et treatment (0.125 ㎎/mL) in HIT-T15 (p<0.05). These results showed that CP-Et contribute to repairing cells and improvement of insulin expression on oxidative stress pancreatic β-cell, and also suggested application of CP-Et as a functional food material for type 2 diabetes.
