        바이칼레인(baicalein)이 poly-IC와 lipoteichoic acid로 자극된 마우스 대식세포 RAW 264.7의 hydrogen peroxide 생성에 미치는 영향

        Objectives : The aim of this study was to investigate the effect of baicalein (BA) on the production of hydrogen peroxide and nitric oxide (NO) in RAW 264.7 mouse macrophages stimulated with polyinosinic-polycytidylic acid (poly-IC) and lipoteichoic acid. Methods : RAW 264.7 co-stimulated with poly-IC and lipoteichoic acid were incubated with baicalein at concentrations of 25 and 50 μM. Incubation time is 16 h, 18 h, 20 h, 22 h, and 24 h. After incubation, The production of hydrogen peroxide in RAW 264.7 was measured with dihydrorhodamine 123 assay. Chrysin was used as a comparative material. NO production was evaluated by griess assay. Results : For 16 h, 18 h, 20 h, 22 h, and 24 h incubation, BA at the concentration of 25 and 50 μM significantly inhibited the production of hydrogen peroxide in RAW 264.7 stimulated by poly-IC and lipoteichoic acid (p <0.001). In details, production of hydrogen peroxide in 'poly-IC and lipoteichoic acid'-stimulated RAW 264.7 treated for 16 h with BA at concentrations of 25 and 50 μM was 82.36% and 77.24% of the control group treated with poly-IC and lipoteichoic acid only, respectively; the production of hydrogen peroxide for 18 h was 83.15% and 77.91%, respectively;production of hydrogen peroxide for 20 h was 82.88% and 77.82%, respectively; production of hydrogen peroxide for 22 h was 83.27% and 78.17%, respectively; production of hydrogen peroxide for 24 h was 83.54% and 78.35%, respectively. Additionally, BA at the concentration of 50 and 100 μM significantly inhibited NO production in lipoteichoic acid-induced RAW 264.7 (p <0.001). Conclusions : BA might have anti-oxidative activity related to its inhibition of hydrogen peroxide production in 'poly-IC and lipoteichoic acid'-stimulated RAW 264.7 macrophages.

        배초향이 RAW 264.7의 염증인자 생성에 미치는 영향

        Objectives : The aim of this study was to investigate the effect of water extract of Agastache rugosa (AR) on productions of inflammatory mediators in lipopolysaccharide (LPS)-stimulated RAW 264.7 mouse macrophages. Methods : Cell viabilities were measured with MTT assay. The production of nitric oxide (NO) from RAW 264.7 cells was measured with Griess reagent assay. The production of cytokines in RAW 264.7 cells was measured with multiplex cytokine assay. Results : AR showed no cytotoxicity on RAW 264.7 cells. AR at concentrations of 25, 50, 100, and 200 μg/mL significantly inhibited NO production in LPS-stimulated RAW 264.7 cells. AR at concentrations of 50, 100, and 200 μg/mL significantly inhibited productions of TNF-α and IL-1β in LPS-stimulated RAW 264.7 cells; AR at concentrations of 50 and 200 μg/mL significantly inhibited productions of RANTES (CCL5) in LPS-stimulated RAW 264.7 cells; AR at concentrations of 100 μg/mL significantly inhibited productions of macrophage inflammatory protein-1β in LPS-stimulated RAW 264.7 cells; AR at concentrations of 50, 100, and 200 μg/mL significantly increased productions of IP-10 (CXCL10) in LPS-stimulated RAW 264.7 cells; AR at concentrations of 100 and 200 μg/mL significantly increased MCP-1 (CCL-2) in LPS-stimulated RAW 264.7 cells; AR at concentrations of 50 and 100 μg/mL significantly increased productions of IL-10 in LPS-stimulated RAW 264.7 cells. Conclusions : AR might have immunomodulatory effects on productions of NO, cytokines, and chemokines in LPSstimulated RAW 264.7 mouse macrophages.

        우고닌(Wogonin)이 poly I:C로 유발된 TM4세포 내 하이드로겐퍼록사이드 생성에 미치는 영향

        Objectives : The aim of this study is to investigate the effect of wogonin on the production of hydrogen peroxide in polyinosinic:polycytidylic acid (poly I:C)-stimulated TM4 mouse sertoli cells. Methods : TM4 were treated with poly I:C (50 ug/mL) and wogonin at concentrations of 5, 10, 25, and 50 μM for 30 min, 2 hr, 12 hr, 18 hr, and 24 hr. The production of intracellular hydrogen peroxide was measured by dihydrorhodamine 123 assay. Results : For 30 min, 2 hr, 12 hr, 18 hr, and 24 hr treatment, wogonin significantly inhibited intracellular hydrogen peroxide productions of TM4 at the concentration of 5, 10, 25, and 50 μM (p <0.05). In details, production of hydrogen peroxide in poly I:C-stimulated TM4 treated for 30 min with wogonin at concentrations of 5, 10, 25, and 50 μM was 95.67%, 92.69%, 92.05%, and 91.97% of the control group treated with poly I:C only, respectively; the production of hydrogen peroxide for 2 hr was 94.44%, 94.41%, 93%, and 92.98%, respectively; production of hydrogen peroxide for 12 hr was 96.78%, 95.32%, 94.33%, and 93.17%, respectively; production of hydrogen peroxide for 18 hr was 94.7%, 93.4%, 93.38%, and 93.35%, respectively; and production of hydrogen peroxide for 24 hr was 95.75%, 94.77%, 94.58%, and 92.8%, respectively. Conclusions : Wogonin might have anti-viral property related with its inhibition of intracellular hydrogen peroxide production in poly I:C-stimulated TM4 cells.

        A new point of view in interior trim is weight reduction. We developed a new door trim material to reduce the gravity. A Light wood-fiber material can reduce the weight( 40 % ↓) with 1.1~1.2T (down from 2.5T). But we should modify the stiffness for structure. First of all, we consider which structure is the best for quality/cost and weight. We analyze the structure for noise and stiffness. Whats more, optimize the process and reduce the increased cost.

        A new point of view in interior trim is weight reduction. We developed a new door trim material to reduce the gravity. A Light wood-fiber material can reduce the weight( 40 % ↓) with 1.1~1.2T (down from 2.5T). We mass-produced the Light wood-fiber door trim in All new K7 project with 3,000won/kg cost increasing. So, we contrive the new types of 1 step covered light-fiber door trim using that’s ventilation property and reduce the cost. First of all, we consider how to reduce the process of the covering of the wood-fiber. And describe the method to secure the quality and cost.

        바이칼레인(Baicalein)이 indomethacin으로 유발된 생쥐 Leydig세포의 일산화질소 생성에 미치는 영향

        Objectives : Baicalein (3,3′, 4′, 5, 6-pentahydroxyflavone), a type of flavonoid, is a well-known antioxidant and anti-inflammatory ingredient found in Scutellaria baicalensis root. The aim of this study is to investigate the effect of baicalein on nitric oxide (NO) production in TM3 mouse Leydig cells stimulated by indomethacin (IN). Methods : TM3 cells were treated with IN (0.5 μM) and baicalein at concentrations of 12.5, 25, 50, and 100 μM for 24 hr, 40 hr, 42 hr, 44 hr, and 64 hr. After treatments, cell viabilities were measured with the modified MTT assay. The production of nitric oxide in cells was measured by Griess reagent assay. Results : Baicalein showed no cytotoxicity on IN-stimulated TM3. NO production in IN-stimulated TM3 treated for 24 hr with baicalein at concentrations of 12.5, 25, 50, and 100 μM was 95.8%, 94.86%, 89.97%, and 81.52% of the control group treated with IN only, respectively; NO production for 40 hr was 97.34%, 97.34%, 95.15%, and 87.42%, respectively; NO production for 42 hr was 89.12%, 90.14%, 89.74%, and 90.26%, respectively; NO production for 44 hr was 83.83%, 84.94%, 85.65%, and 86.85%, respectively; NO production for 64 hr was 94.12%, 95.38%, 94.21%, and 94.12%, respectively. Specifically, baicalein at concentrations of 12.5, 25, and 50 have been shown to most efficiently inhibit NO productions in 48 hr of treatment. Conclusions : Baicalein might have anti-toxicant effect on Leydig cells related with its inhibition of NO production in Leydig cells stimulated with IN.

        A new point of view in interior trim is decreasing thickness for weight reduction. We developed a new door trim with 2.0T (down from 2.5T). A whisker is added in PP to secure the stiffness. A whisker is a type of needle crystal having a high stiffness, but it causes PP to become anisotropic. This causes mismatching with counterparts and deformation. To solve this problem, we address the topic of whisker arrangement using mold flow analysis. Depending on the arrangement of whiskers, anisotropic deformation and bending of the door trim can occur. In addition, the mold shape having a large slope or step can cause deformation and bending as well. This paper classifies the characteristics of the whiskers based on mold shape and mold flow. We find that when the whiskers are arranged in an irregular pattern, the material has less anisotropic characteristics.

        Objectives : The aim of this study was to investigate the effect of baicalein (BA) on the production of hydrogen peroxide in peptidoglycan-stimulated RAW 264.7 mouse macrophages. Methods : Peptidoglycan-stimulated RAW 264.7 were incubated with baicalein at concentrations of 50 and 100 μM. Incubation time is 30 min, 2 h, 12 h, and 18 h. After incubation, The production of hydrogen peroxide in RAW 264.7 was measured with dihydrorhodamine 123 assay. Berberine and gallic acid were used as the comparative materials. Results : BA at the concentration of 50 and 100 μM did not show cytotoxicity on RAW 264.7 for 24 h incubation. For 30 min, 2 h, 12 h, and 18 h incubation, BA at the concentration of 50 and 100 μM significantly inhibited the production of hydrogen peroxide in RAW 264.7 stimulated by peptidoglycan (p < 0.05). In details, production of hydrogen peroxide in peptidoglycan-stimulated RAW 264.7 treated for 30 min with BA at concentrations of 50 and 100 μM was 93.91% and 93.52% of the control group treated with peptidoglycan only, respectively; the production of hydrogen peroxide for 2 h was 93.8% and 92.71%, respectively; production of hydrogen peroxide for 12 h was 94.86% and 95.93%, respectively; production of hydrogen peroxide for 18 h was 95.37% and 96.48%, respectively. Conclusions : BA might have anti-oxidative activity related to its inhibition of hydrogen peroxide production in peptidoglycan-stimulated RAW 264.7 macrophages.

        건설 산업은 혼합 폐기물 발생이 많고, 잦은 설계변경으로 인해 전략적인 폐기물 관리가 어려운 것이 현실이다. 이러한 현실 때문에 현장에서는 단순히 발생된 폐기물을 분류하여 보관 및 처리하는 수동적인 사후대처식 폐기물 관리방식을 고수하고 있다. 그러나 사후대처식 폐기물 관리방식은 체계적인 건설폐기물 관리가 어려워 현장의 폐기물 절감 노력을 저해할 뿐만 아니라 폐기물 중간 처리 업체에 의존하는 원인이다. 본 연구에서는 사후대처식 건설폐기물 관리 방식의 한계를 극복하고 보다 체계적으로 건설폐기물 관리를 수행하기 위해 폐기물의 성상, 발생량, 발생 시점을 예측하는 건설폐기물 관리시스템과 건설폐기물의 현장재활용을 유도하는 폐기물 공유 체계를 제안하고, 대상건축물에 적용하여 시스템의 유효성을 검증하고자 한다. The problems of environmental pollutions and resources depletion have been growing issues in global construction recently. Efforts to reduce $CO_2$ emission have been also made in all sectors of construction industry these days. As one of the biggest industries that consume a huge amount of resources and generate complex construction wastes, the construction industry has significant impacts on environment issues. However, systematic approach to manage wastes has been rarely made, and most construction wastes from construction sites are being land-filled or incinerated. In this study, a system is proposed to predict the amount of wastes in visual formats, and to control the process of wastes management. The system's main functions include : (1) to estimate the amount of wastes to be generated in project schedule, (2) to categorize the types of wastes, (3) to determine the timing of taking out wastes from sites, and (4) to share information regarding wastes for recycling. A huge amount of wastes are generated in construction process, but most of the wastes have been discharged in forms of mixed wastes, which make them hardly reused. The system not only provide information on wastes to be generated, but also prevent mixing various wastes by classifying them by types and schedules. This features of the system, along with functions to share wastes information with other agencies outside the site, are expected to enhance the level of wastes recycling to a great extent. By saving construction materials through wastes recycling, the system also contributes in reducing $CO_2$ emission.
