초파리의 su(s)²유전적 억제현상을 규명하기 위한 6-pyruvoyl tetrahydropterin synthase의 peptide mapping 분석

초파리에서 purple 돌연변이를 회복시키는 su(s)2 유전적 억제현상을 구명하기 위하여 purple 유전자 산물인 PTP synthase를 야생형, purple, suppressed purple 돌연변이체로 부터 각각 분리하고 trypsin을 이용하여 일차원 SDS-PAGE 전기영동 상에서 단백질 절편들의 변화를 분석하였다. 그 결과 purple과 suppressed purple 돌연변이 체의 PTP synthase는 야생형의 효소와는 분명한 차이를 보여주고 있어 이 유전적 억제 현상이 mRNA 수준에서의 양적인 변화에 의한 것이 아니라 아미노산의 변화에 기인한 posttranslational 기작이 관여하고 있음을 지지하고 있다. The amount of red pteridine eye pigments (drospterins) in Drosophila is known to be reduced in the purple mutant and restored to normal levels by a suppressor mutation, su(s)2. Currently there are three different types of mechanisms proposed to explain the suppression : these are tRNA-mediation, transposable element-mediation, and a posttranslational mechanism. To elucidate the suppression mechanism the purple gene product, 6-pyruvoyl tetrahydropterin synthase (PTP synthase), was analyzed for their possible primary structure differences in wild type, purple mutant and suppressed purple mutant strains. PTP synthases resolved in SDS gel slices were subjected to tryptic peptide mapping on SDS-PAGE. Based on the peptide mapping pattern, a mechanism was suggested that involves missense suppression and a posttranslational effect on the enzyme.
