수송 결함을 지난 리보스 결합 전구체 단백질, 복귀 전구체 단백질 및 숙성단백질을 부분 또는 순수 정제하고 이들이 기질에 결합하는 것을 조사하여 대량생산된 리보스 결합 단백질들이 native conformation을 유지하고 있음을 확인하였다. 따라서 이들 단백질을 수송 능력과 구조 사이의 관계를 알아보기 위한 in vitro 분석에 사용하였다. 이들 단백질에 리보스가 결합하는 정도의 비교, circular dichroism 의 측정 및 proteinase K에 대한 내성을 조사한 결과 steady state에서 이들 단백질 사이에 현저한 구조적 차이가 없음을 밝혔다. Three alleles of rbsB gene including export defective mutant were cloned for overproduction of proteins. Five different species of precursor and mature ribose-binding proteins were purified to homogeneity. The purified proteins were subjected to amino acid sequencing. The results confirmed the amino acid changes deduced from the DNA sequencing (Park et al., 1988a). Attempts were made to establish a correlation between export competency and conformation of the proteins by studies in vitro. In this regard, ligand binding ability, circular dichroism, and the sensitivity to the degradation by a protease were compared. However, there was no drastic conformational difference among these proteins in steady states. This results, in other aspect, implied that the proteins purified from the overproduced proteins are in native conformation and allowed further analysis for studying the mechanism of protein export in vitro.

To understand the mechanism of protein export in general and to identify component(s) of export machinery utilized by ribose-binding protein, we examined the effects of three prlA suppressors on the export of an export-defective mutant rbsB103. We also studied the requirement(s) of SecY protein and leader peptidase on the export of wild-type ribose-binding protein. The export defect of rbsB103 mutation was not suppressed by three prlA alleles tested. However, SecY protein appears to affect for the export of wild-type ribose-binding protein. Wild-type precursor ribose-binding protein was not processed by purified leader peptidase (lep product) in vitro.

We produced human recombinant interleukin-1β in E. coli to study the mechanism (s) of its pleiotropic action(s), and to develop it as an anti-cancer or anti-inflammatory drug. We constructed expression vectors using lambda P_L promoter. Expression of hrIL-1β in E. coli was induced and the production of the protein was confirmed by Western blot assay. We also confirmed the presence of biological activity of recombinant IL-1β protein by demonstrating its growth promoting effect on mouse thymocytes.
황금·황련 추출물의 항균활성이 막걸리 저장성에 미치는 영향

This research was carried out to find herbal preservatives for Makgeolli, as Makgeolli loses its commercial value due to overproduced acidic materials. When Makgeolli was kept at $25^{\circ}C$ to find the changes in acidity, total microbial cell number, yeast cell number, and bacterial species variety, a sudden increase of acidity as well as the disappearance of yeast cells occurred at day 6, and Makgeolli was changed to complete off-flavor. Acetobacter pasteurianus is the main acidifier in Makgeolli and shows a synergy effect in acid formation when cultured in combination with Lactobacillus casei. Among 12 herbs, the ethanol extract of Sutellaria baicalensis showed antimicrobial activity against A. pasteurianus, whereas the ethanol extract of Coptidis rhizoma showed antimicrobial activity against L. casei. Makgeolli added with Sutellaria baicalensis extracts demonstrated a lower acidity than that with Coptidis rhizoma extracts, which indicates that the inhibition of an acetic acid former is more important than that of a lactic acid former in Makgeolli preservation. Sutellaria baicalensis extracts prolonged the shelf life of Makgeolli by 1~2 weeks at a minimal inhibitory concentration ($0.63mg/m{\ell}$) during storage at $10^{\circ}C$.

This research was carried out in order to discover acid-forming bacteria during fermentation of Makgeolli, as Makgeolli loses its commercial value due to overproduced acidic materials. In Makgeolli kept at $25^{\circ}C$, a sudden increase of acidity as well as the disappearance of yeast cells occurred at day 6, whereas the total cell count and bacterial type remained unchanged; the result implies that a succession of bacterial types, including acid forming bacteria, occurred. Two acid-forming bacteria were isolated from acidified Makgeolli and were identified as Acetobacter pasteurianus and Lactobacillus casei. When fresh and heat-treated Makgeolli were inoculated with Acetobacter pasteurianus and/or Lactobacillus casei, the greatest amount of acid was formed in Makgeolli inoculated with Acetobacter pasteurianus and Lactobacillus casei and also in Makgeolli with Acetobacter pasteurianus alone. This result indicates that Acetobacter pasteurianus is the main acidifier; furthermore, it shows the synergy effect in acid formation with Lactobacillus casei.

N/A Objective: We studied the efficacy of the power spectral and nonspectral analysis and its diurnal variations for the early detection of the diabetic autonomic neuropathy. Method: The spectral and nonspectral analysis of 24hour-Holter monitoring were done for both diabetic neuropathy patients and controls. We also made a comparative analysis of the diurnal variations between the patient and control groups by means of hourly power spectral analysis. Result: 1) The power spectral density of the diabetic neuropathy patients was below than that of the normal controls (P<0.05) (Table 2, Fig. 1). 2) The nonspectral analysis of the diabetic neuropathy patients was below than that of the normal controls except for the mean RR intervals (P<0.05) (Table 3). 3) Every hour-power spectral analysis showed a diurnal variation of day time (06:00 to 16:00) decrease in high frequency area of the normal controls. On the contrary, there was a disappearance of the diurnal variation in patient group. Conclusion: The power spectral and nonspectral analyses after 24hour-Holter monitoring and its diurnal variation in the diabetic patients are considered as good means of the early detection of autonomic neuropathy, but further study of its diagnostic value will be needed.
