      • 동위효소 및 RAPD분석에 의한 한국재래종 누에계통의 계통학적 특성

        집누에는 지리적인 특징에 의해 중국종, 일본종, 유럽종, 열대종 및 한국종으로 분류된다. 한국종계통은 품종수도 적을 뿐만 아니라 관련 연구도 미미한 실정이다. 본 연구는 한국재래종으로 추측되는 품종들을 국내 ? 외로부터 수집하여 동위효소 및 체액단백지유전자와 RAPD 다형분석을 실시하여 한국재래종 계통의 품종적 유연관계와 계통특성을 밝히고자 하였다. 1. 동위효소유전자의 분석결과, 몇 개의 유전자군에서 한국재래종과 타 지역종간에 유전자형 및 유전자 빈도의 차이가 명확히 나타났다. 2. RAPD의 결과를 UPGMA법에 의해 분석한 결과, 집누에군과 멧누에군으로 크게 구분되었으며 유전적 유사계수 0.6930을 기준으로 한국재래종, 일본종 및 중국종으로 그룹화되었다. 3. 이상의 결과를 종합하면, 한국재래종 누에계통은 하나의 지역종 원종계통으로 분류될 수 있는 명확한 유전적 특성을 가지는 것은 물론 한국종의 계통특성도 뚜렷한 것으로 확인되었다. This study was conducted to elucidate phylogenetic relationships and genetic characterization of silkworms that might be recohnized as the Korean native strains. Genetic characterization in isozymes and the proteins of larval hemolymph of 17 silkworms were observed by acrylamide gel eletrophoresis, on 12 genes; Bph, Bes, Ies, Amy-hc, Ict-A, -B,-D, -E -H,Pfl, Pst, Lp. Gene frequencies in each locus were compared other geographic strains. Korean native strains were remarkably different from others considered as the genetic characterization of Korean native strains. Phylogenetic relationships in Korean native strains were analysed using RAPD-PCR markers. A total of 40 primers were used and 346 bands of amplified DNA were generated from geographic strains, Genetic similarity based on the RAPD bands was used to construct phylogenetic dendrogram based on analysis of band sharing data of amplified markers. Genetic similarity ranged from 0.595 to 0.860. In the genetic relationship based on dendrogram, they were classified into Bombyxmori group (including 16 domesticated silkworm strains) and B. mandarina group. The Bombyx mori group was separated into three sub-groups at the genetic similarity of 0.6930, including Korean, Japanese and Chinese groups. According to this result, the Korean native variety can be considered as a clearly different variety from other geographic strains. It may be concluded that the Korean native strains are also one of original geographic rariety such as Japanese, Chinese, etc.

      • 신돌연변이잠 cre(반월형란)의 유전자 연관분석과 유전형질

        새로운 난형이상 자연발생 돌연변이체인 반월형란(cre)에 대한 유전자 연관분석과 유전형질의 특성에 대하여 조사하였다. 현재까지의 유전자 연관분석에서 cre유전자는 집누에 28개 연관군중 20개의 연관군과 독립하는 것이 밝혀졌다. cre란은 수정 ? 부화 및 조란이 모두 정상적이며 번데기체액 단백질의 함량과 성분도 정상란과 차이가 없었다. 난소의 발육에 따른 난황단백질의 함량 및 성분에서도 정상과 차이가 없었다. cre란의 배면부 난각 표면구조에서 정상란과는 상이한 구조가 관찰되었으며 이 구조는 측면부의 표면구조와 유사하였다. 이상의 결과에 의해 반월형란의 난형 이상은 배면부의 난각 부위특이성과 함께 난각구축과정에서의 이상에 기인하는 것으로 추측된다. The “crescent-egg” a new spontaneous mutant was detected in a white egg strain k37. Studies were carried out the linkage analysis, to investigate phinotypic characteristics and biochemical analysis of haemolymph and ovarian protein. The mutant, cre was independent from 20 linkage groups P(2), Ze(3), L(4), oc(5), st(8), Ia(9), w-1(10), K(11), ch(13), U(14), bl(15), cts(16), bts(17), mln(18), nb(19),oh(20), Lan(21), or(22), tub(23) and Xan(27). The ferilization, hatchability and larval growth were not different from the those of normal eggs. The content and composition of yolk protein were similared to normal eggs. Scanning electron microscopy revealed the areal specific structure in dorsal region of egg-shell of cre mutant. Analysis of chorion protein by isoelectrofocusing(IEF), was resolved no difference in the composite of the chorion protein. We conclude that the egg mutant cre is expressed only in the egg-shape formation and region specific determination.

      • 곤충의 식물병원성 진균에 대한 항균활성

        본 연구는 식물병원성곰팡이에 대한 곤충 혈액 및 유충 체의 항곰팡이 활성을 조사하였다. 집누에, 장수풍뎅이, 흰점박이 꽃무지 모두 곰팡이 저해활성을 나타냈으며, 집 누에의 계통간 비교에서는 1087계통의 활성이 가장 높았다. 또한, 혈액 보다는 유충체를 이용한 경우가 더 강한 항곰팡이 활성을 나타냈다. 곤충의 항곰팡이 활성은 곰팡이균주에 따라 다르며, 본 연구에 사용된 3종은 A pannax, C. gloeosporioides 및 P. oryzae 균에 항곰팡이 활성을 가진다. The hemolmyph and whole body of insect, Bombyx mori, Allomyrina dichotoma and Neotocia brevitarsi, conspicuously inhibited the mycelial growth of several plant pathogenic fungi. The hemolymph of 1087 strain among the 16 strains of B. mori has inhibition activities against the 3 species of fungi, Alternaria panax, Collctotrichum gloeosporioides, and Pyricularia oryzae. The whole body of B. mori was more effective than the hemolymph as a inhibitor on fungi growth. The antifungal activity of B. mori was variable to the fungi species. In addition, A. dichotoma and N. brevitarsi showed antifungal activities against the same fungi as did B. mori. These data showed that the insect has potent antifungal activity. Whereas, the level of activities were differ according to the fungal species. This finding underlines that the possibility of the insect can be use of the agent as a inhibitor against the plant pathogenic fungi.

      • 누에 형질전환에 적합한 실용품종 누에알의 제조

        Silkworm transgenesis scientists have done some genetic modification work on multivoltine silkworms, but that type of silkworms is less commercial feasible. They are easy to manipulate, because they breed all year round. But the commercial silkworm variety must undergo hydrochloric acid treatment at a high temperature to be artificially hatched. Hydrochloric acid penetrates through the holes in the silkworm eggs, fatally damaging their reproduction. So it had been thought that altering the properties of the commercial silkworm variety would be very difficult. So we have tried to make from diapause to non-diapause eggs using diapauses varieties, ``Backokjam`` and ``Jam 124``. At present, our group has establishing the conditions for non-diapause eggs. Oviposited eggs after 40 ~ 60 hours were incubated for 24 hours at 15 ~ 20oC with dark condition. Non-diapause eggs were completely induced. The hatching rate, molting rate and pupation rate of non-diapause ``Jam 124`` and ``Backokjam`` eggs showed no differences compared to diapause eggs. When transgenic silkworm using the non-diapause eggs, the hatching rate showed that non-diapause eggs induced from diapause were 40 ~ 70%, diapause eggs treated with artificial incubation were 10 ~ 30%, and polyvoltine strains, HM eggs were 30 ~ 50%. Therefore, we suggest that modification techniques of the commercial silkworm eggs adequate for silkworm transgenesis can be used to develop transgenic silkworms more easily.

      • 種繭의 保護溫度가 蠶蛾의 産卵性에 미치는 影響

        The study is conducted to clarify the difference of oviposition ability of the silkworm mother moths sampled from race, Bombyx mori. which had been preserved in various temperatures in pupal stages. The results obtained are summarized as fellows; The number of mature eggs and the ration of total eggs laid in low temperature preservation in the preceeding term of the pupal stage are higher than those in high temperature preservation. Among the treatments the high and low temperature in the late term of pupal stage revealed the worst results. Ratio of normally oviposited eggs is exceedingly low if kept at low temperature in previous term and it is higher in the Chinese race than in the Japanese. However preservation temperatures affected not only oviposition ability, but states of egg laying. Ratio of unfertilized eggs is higher at low and high temperature during the pupal period and low temperature is more effective than high temperature in previous term. Correlation between the number of mature eggs in oviduct and normally oviposited eggs is high significance in every races and every treatments.

      • 腎藏形卵에 있어서 卵殼構造의 解析

        The structure ki mutant chorion is described by the ultrastructural level with the scanning electron microscope. The micropyle on the anterior, posterior, leteral and dorsal side region surface patterns do not so much differ from ki mutant and normal strains. The surface structure of ventral side of ki mutant, which is the presumptive embryonic region in oocytes, were netted part corresponding to the boundary of follicle cells that is similar to lateral side structure of egg·shell. The egg-shell thickness of ventral side on the ki mutant is mostly thicker. However, the structure of transverse section of normal and ki mutant egg-shell are composed of the outer, middle and inner layers. The ki not effects the formation of the lamellar layer in the egg-shell. It is observed that the characteristic structure in ki mutant egg chorion may be attributed to a areal specificity of epithelium in ventral side of egg-shell formation, and it affects the process of the accumulation of secreted chorionic material and the egg-shell architecture.

        This studf was carried out to investigate the elucidation of gene expression in embryo formation of the Kidney mutant, especially yolk proteins in unfertilized eggs and template activity of m·RNA extracted from them. The results were summarized as follows: There was no recognized qualitative difference in yolk proteins of unferilized eggs between the Kidney mutant and normal. There was not any difference, between Kidney mutant and normal in the molecular species of m-RNA derived from maternal origin with the template activity in vitro.
