진주담치의 homogenate로부터 음이온교환수지와 친화력 크로마토그라피를 이용하여 렉틴성분을 분리정제하였다. 이 렉틴성분은 전기영동상에서 주변 하나의 띠로 나타났으며 분자량은 약 18,000 dalton이었고 monomer로 생각된다. 이 렉틴은 토끼 적혈구를 강하게 응집시켰으며 사람혈액형에 대하여는 특이성이 낮았고 그중에서 A형 적혈구를 잘 응집시켰다. 이 렉틴의 적혈구응집활성은 galactose 및 galactose의 유도체에 의해 저해되었으며 EDTA, $Hg^{++}$및 $Mg^{++}$ 등의 양이온에 의해서도 저해되었다. 이 렉틴은 당을 함유하지 않은 단백질로 나타났으며 열에 불안정하였다. The purification acheived was 6,628 fold from 20,000 g homogenate with a yield of 58.1%. Its isoelectric point was around pH 7.8 and its molecular weight was estimated to be about 18,000 daltons by Sephadex G-75 gel filtration and SDS-polyacrylamide gel electrophoresis. The mussel lectin was stable below $35^{\circ}C$. The lectin agglutinated rabbit erythrocytes strongly and showed the broad specificity to human blood groups. The hemagglutinating activity of the lectin was inhibited by sugars; galactose and its derivatives, and cations; EDTA, $Hg^{++}$, and $Mg^{++}$. This lectin seemed to have no carbohydrates.

한방재료 중에서 지혈, 창종, 부종의 치료에 사용되는 천연식물 70여종을 택하여 적혈구응집소의 존재 유무를 검색하였다. 그 중 괄루근(Trichosanthis Radix, TR)으로부터 렉틴을 분리정제하였다(TRL). 황산암모늄 침전법, DEAE-cellulose ion-exchange chromatography, Sephadex G-150 gel filtration 방법 등에 의해 72.95배 정제하였고 회수율은 9.96%이였다. 정제된 렉틴은 glycoprotein으로 polyacrylamide gel electrophoresis에서 단일의 띠를 보여주어 거의 순수하게 정제됨을 나타내었다. TRL은 적혈구, 임파구, 혈소판에 대해 응집현상을 나타내었고 그 외 박테리아 응집효과에서 Escherichia coli와 Pseudomonas aeruginosa를 응집하였다. 또한 TRL을 ICR mouse에 투여했을 때 백혈구와 임파구의 증가를 가져와 면역기구중의 effector로서 면역감응세포의 증식 분화를 유발시키는 것으로 추측되었다. Following injection of TRL into ICR mouse, ${\gamma}-globulin$ was increased to 83% and the number of lymphocytes was increased to 18%. These results suggest that TRL may have stimulating effects on general immune systems.

사람 Low Density Lipoprotein (LDL)에 대한 Rooibos tea의 항산화 활성

This study was investigated to antioxidative activity of crude catechin extracted from Rooibos tea against low density lipoprotein(LDL) oxidation. The crude catechin inhibited the copper-mediated oxidation of human LDL at the concentrations of 50 and 100 ㎍/㎖ in the presence of 5 μM CuSO4. The crude catechin also inhibited LDL oxidation induced by macrophage J774 cell. LDL oxidized by copper mediated and cell induced oxidation was degraded by macrophage at a much greater rate than native LDL. The electrophoretic mobility of oxidized LDL by addition of crude catechin was faster than that of native LDL. The results provided a possibility that crude catechin of Rooibos tea might protect LDL against oxidation in atheroscleotic lessions.

Since ancient times Trichosanthis Radix has been used in traditional herbdrung as an antitumor and immunologic stimulating agent. Lectin was purified to homogeneity from Trichsanthis Radix by ammonium sulfate precipitation, DEAE-Cellulose chromatography and Sephadex G-150 gel filltration. The major amino acid was tyrosine and some aspartic acid, glutamic acid and saccharides were also present. Trichosanthis Radix Lectin (TRL)strongly stimulated activity of human erythrocytes, lymphocytes, and thrombocytes. In addtion, agglutination of gram negative Escherichia coli and Pseudomonas ararugimosa was shown, but proteus, Hemophilus influenza and Staphylococcus aureus were not agglutinated. Folloaing injection of TRL into ICR mouse, granulocyte was increased by 87.5% and the number of lymphocyte was increased by 25-28%. These results suggest that TRL may have stimulating effects on the general immune systems.
