배독환발효방(排毒丸醱酵方)의 면역조절작용을 통한 항아토피 효능

Herbal medicine has a high body absorption rate when it ferments. Biological and clinical research on the fermented herb gradually increases because it has effective materials for the treatment of a disease and it is a little bitter. In this study, we investigated the effect of fermented Baedokhwanbalhyobang (BDHBH) on attenuation of the development of atopic dermatitis in NC/Nga mice by evaluating the cytokine level in serum, the mRNA expression of cytokine and histological alteration of the skin, and the skin severity. We have come to the following conclusion. BDHBH led to a significant decrease in the skin severity score (63.1%) as compared to the control group. CD4+/CD45+, CD4+, B220+/CD23+, and CD11b+/Gr-1+cells of peripheral mononuclear cells(PBMCs) in the BDHBH-treated group were decreased to 6.7%, 31.1%, 22.4%, 36.6%, respectively. CD3+and CD11b+/Gr-1+immune cells in dorsal skin of the BDHBH-treated group were decreased to 52.9% and 28.0%. The levels of IL-5 and IL-13 in serum of the BDHBH-treated group were inhibited to 18.8% and 5.1%. The mRNA expressions of IL-5 and IL-13 in dorsal skin were also decreased to 30.6% and 27.8% after the treatment of BDHBH. BDHBH inhibited the proliferation and differentiation of eosinophils. In histological examination, BDHBH decreased the thickness of epidermis and dermis, and infilatration of mast cells as compared to the control group. These results indicate that BDHBH inhibits the pathogenic development of atopic dermatitis in NC/Nga mice. These results may indicate that BDHBH attenuates the development of atopic dermatitis-like lesions by lowering immune cells and inflammatory cytokine levels, and that it is valuable in drug development for the treatment of atopic dermatitis. Further experiments on the components of BDHBH will be needed to better understand the effect of a fermented herb as compared to a herb.

Volatile odor substance originating from drying and making dry peel of tangerine from the fruit skin were collected through modern equipment. The anti-microbial activity of the substance against various skin-residing bacteria including acne, dandruff, athelete`s foot, and gingivitis inducing microorganisms were tested. Anti-microbial activity was observed in purified oil, where 87 to 92% was D-limonene. Against P. acnes, 103cfu/㎖of P. acnes were suppressed at 0.1% Unshiu oil, and the MIC was measured to be 0.3%. Against P. ovale, a dandruff inducing bacteria, 104cfu/㎖were suppressed at 0.1% Unshiu oil, and the MIC was measured to be 0.1%. Against T. rubrum and T. Mentagrophytes, both of which are athelete`s foot inducing microorganisms, 83% of T. Mentagrophytes and 99.9% of T. rubrum were suppressed at 0.1% Unshiu oil, and the MIC were 0.3% and 0.05% respectively. Against S. aureus, a skin infection inducing bacteria, 103cfu/㎖of the bacteria were suppressed at 0.1% Unshiu Oil. Against B. subtilis, a non-pathogenic sporulating bacteria, 104cfu/㎖of the bacteria were suppressed at 0.1% Unshiu Oil. Against C. albicans, found in mucous membranes, 104cfu/㎖of the bacteria were suppressed at 0.1% Unshiu Oil. Against Aspergillus niger, an otomycosis inducing microorganism, 99.9% were suppressed at 0.1% Unshiu Oil. The results above indicate that low concentration of purified oil extracted from tangerine had strong antimicrobial activities against bacteria and fungi residing on the skin, and that it may be developed into skin disorder treating products in the future.
청기소독탕(淸肌消毒湯)이 아토피피부염 염증 관련 인자에 미치는 영향

In vitro tests were performed using CST to investigate its role on oxidative damages and inflammatory cytokines. 90% or higher cell viability was observed in CST treated groups from 25 to 200 ㎍/㎖using Raw 264.7 cells. CST showed dose-dependent DPPH scavenging activity, with 91.3% and 92.2% scavenging activities at 400 and 800 ㎍/㎖concentrations, respectively. CST showed dose-dependent suppression activity of ROS production, especially at 200 ㎍/㎖of 41.3%. CST decreased NO production activity, with significant decrease of 16.2% and 33.5% at 100 and 200 ㎍/㎖concentrations, respectively. IL-1β, IL-6, MCP-1 production rate were significantly decreased by 30.0%, 27.2%, 22.1% when Raw 264.7 cells were treated with LPS and with CST of 200 ㎍/㎖. Also, TNF-α production rate was decreased by 28.6%. The results above indicated therapeutic effect of CST on the AD through anti-oxidative and immune modulatory effect. Various blending of drug substances with CST should be clinically tested.
아토피피부염 동물 병태 모델에서 청화탕(淸華湯)의 면역조절작용에 관한 연구

Objectives : The aim of this study was to investigate the effect of Chunghwatang (CHT) for atopic dermatitis. Methods : CHT, a verified anti-oxidant and anti-inflammatory effect, was treated in atopic dermatitis animal model to investigate cytokine levels and immunoglobulin production. Results : Clinical skin index was 47.1%, suggesting significant efficacy of CHT in atopic dermatitis treatment. Serum IL-4, IL-6, IL-13, TNF-α and histamine productions were significantly decreased to 52.3%, 61.8%, 68.0%, 37.4%, and 46.6%, respectively. The production of IL-5 was decreased to 33.3%. The increase of Immunoglobulin IgG1 production along with IgE through the interaction of IgE and IL-4 induced by IL-4 and IL-13 were measured as 20.7% and 23.4%, respectively. Conclusions : The results above, along with the in vitro test results, strongly supports the CHT samples as effective immunomodulator in AD treatments, suggesting its use in clinical practice and basic information for EBM.

In order to investigate the possibility of Kamisamultang(KMSMT) as therapeutic for the treatment of atopic dermatitis(AD), cytotoxicity, anti-oxidant activity, modulatory and suppression activities of KMSMT were tested. 90% or higher cell viability was observed in all tested groups from 25 to 200 ㎍/㎖using Raw 264.7 cells. KMSMT showed dose-dependent DPPH scavenging activity, with more than 80% scavenging activities at 400 and 800 ㎍/㎖concentrations. KMSMT showed dose-dependent suppression activity of reactive oxygen species(ROS) production, especially at 200 ㎍/㎖of 42.6%. KMSMT decreased nitric oxide(NO) production activity dose dependently, expecially at 200 ㎍/㎖of 30.9%. IL-1β, IL-6, MCP-1, TNF-α production rate were decreased by 45.7%, 15.5%, 8.9%, 16.5% respectively when Raw 264.7 cells were treated with LPS and with KMSMT of 200 ㎍/㎖. However, only IL-1β and MCP-1 showed significant changes. The results above strongly suggest the modulatory and suppressive effect of KMSMT. The results above indicate that KMSMT significantly reduces the effect of oxidative and inflammatory cytokines. The use of KMSMT in atopic dermatitis can be widely suggested.
외치방인 청기패독산(淸肌敗毒散)의 아토피피부염 동물 병태 모델에서의 면역조절작용에 관한 연구

In order to study the effect of CPS in the treatment of atopic dermatitis (AD), its role on various immune related cytokines were tested. Levels of liver and kidney function markers such as ALT, AST, BUN in NC/Nga mice were all normal indicating no toxicity in CPS treated group. Significant recovery from AD could be observed in CPS treated group through naked eye observation. Dermatitis index was significantly decreased after 11, 12, and 13 weeks of treatment. CD4+, CD8+, CD3+ /CD69+ immune cell ratio in DLN were decreased significantly by 37.2%, 49%, 20.8% in CPS treated group. CD4+ and CD11b+ /Gr-1+ immune cell ratio in dorsal skin were decreased significantly by 50.8% and 59.2% in CPS treated group. Expression of IL-4, IL-5, IL-6, IL-13 and TNF-α in spleen were decreased by 78.8%, 97.8%, 64.7%, 73.6%, and 68.4%, respectively in CPS treated group. The results above strongly indicated the significant immune modulatory effect of CPS and thus clinical application of CPS on AD treatment.
TNCB로 유발한 아토피피부염 생쥐 모델에서 천일염가미방(天日鹽加味方)과 청기해독산(淸肌解毒散)의 병용 효과

Objectives: Atopic dermatitis is a chronic inflammatory disease characterized by typically distributed eczematous skin lesion with pruritus, lichenification and dry skin. In this study, we performed to assess the therapeutic effects of co-treatment of Chenilyeomgamibang (CGB) and Chenggihaedok-san (CHS, C&C) on the TNCB(trinitrochlorobenzene)-induced atopic dermatitis in NC/Nga mice, characterized by the onset of atopic dermatitis along with an increase the number of inflammatory cells and dysregulation of Th2 cytokines. Methods: Defined amount of CGB was sprayed on mice skin and CHS was simultaneously orally administrated to TNCB treated NC/Nga mice for 5 weeks. The immune cell types were caracterized by flow cytometry using each specific antibody. The amount of Th2 cytokines in serum and splenocytes culture supernatant were measured by ELISA. Results: Administration of C&C significantly reduced clinical dermatitis severity including pruritus, edema, eczematous and erythema. Histological findings indicated that the thickening of epidermis/ dermis and dermal infiltration of inflammatory cells were dramatically reduced. Flow cytometry analysis showed that infiltrated immune cell numbers of CCR3+, B220+/IgE+, Gr-1+/CD11b+, and CD117+ were significantly reduced in C&C-treated dorsal skin lesion. Furthermore, T cell composition rate in PBMC was also dramatically decreased by the treatment. C&C greatly down-regulated production of Th2 cytokines including IL-4, IL-5, IL-13 in the serum. The down-regulatory effects of C&C on these Th2 cytokines production were also detected in CD3/ CD28 activated splenocytes. Conclusions: These results indicated that C&C is a plausible therapeutic agent for treatment of atopic dermatitis through regulating the Th2 skewed immune system.
