많은 관심이 집중되고 있는 환경오염의 대처방안의 일환으로, plastic 및 wrap 제조시 가소제로 가장 널리 사용되고 폐수 중에 대량 유출되어 인체에 유독한 난분해성 고분자 유기합성 물질로서 문제가 되고 있는 phthalate ester의 일종인 dibutyl phthalate(DBP)를 분해자화할 수 있는 미생물을 분리하고자 했다. 저자들은 대구 근교의 토양 및 sludge에서 DBP 분해 관련 효소인 protocatechuate dioxygenase을 생성하는 새로운 균주를 분리하여 동정하였다. 그 결과 통성혐기성균의 일종인 Klebsiella oxytoca와 형태적, 생화학적 특성이 거의 일치하는 새로운 균주임을 알게되어 Klebsiella spp. KBO-222로 동정했다. 본 효소를 생성하기 위한 최적조건으로서 배지조성은 C원으로서 PCA 0.1%, N원으로서 yeast extract와 ammonium sulfate를 각각 0.1%되게 함께 넣었다. 배양 최초 pH가 7이 되게 하여 30℃에서 40-48시간 진탕배양했을 때가 가장 효소활성이 높았다. 또한 본 균이 생산하는 protocatechuate dioxygenase는 유도 효소이고, 또한 세포내 효소로서 적어도 cell membrane과 관계 있거나, periplasmic과 관계하는 듯 했다. This strain produced the enzyme in question under the shaking cultivation at 30℃ for the 48 hrs in the medium containing 0.1% protocatechuate as the sole carbon source, 0.1% ammonium sulfate and 0.1% yeast extract as the nitrogen source and mineral salt mixture of magnesium sulfate, sodium chloride, calcium chloride, ferric chloride, manganese sulfate, zinc sulfate and cupric sulfate. This enzyme was intracellularly localized and probably linked to cell membrane, and induced by protocatechuate.

A strain degrading phthalate ester was isolated from a sludge of Taegu area and identified as a strain of Klebsiella. The optimum culture conditions for the protocatechuate dioxygenase production were oleo investigated. This strain produced the enzyme in question under the shaking cultivation at 30℃ for the 48 hrs in the medium containing 0.1% protocatechuate as the sole carbon source, 0.1% ammonium sulfate and 0.1% yeast extract as the nitrogen source and mineral salt mixture of magnesium sulfate, sodium chloride, calcium chloride, ferric chloride, manganese sulfate, zinc sulfate and cupric sulfate.
