拘杞子나무의 四分子期, 1核性小胞子期, 未熟花粉期의 幼葯을 Murashige and Skoog의 改良培地에 2,4-D, NAA, Kinetin을 各 各 2㎎/ℓ 씩 添加해서 만든 固體 培地에 接種하였다. 小胞子는 培養 2週 後면 肥大하기 始作하고 계속 分裂해서 自色光澤의 Callus가 葯을 헤치고 밖으로 나왔다. 葯의 發育時期에 따른 Callus形成은 1核性小子期가 四分子期나 未熟花粉期보다 良好하였다. 培養 3週後 頃이 되면 葯은 大部分 褐變하였으며 小胞子由來로 判斷된 Callus만 分離 0.1mg/ℓ 2.4-D와 0.2㎎/ℓ NAA로 各 各 減量한 培地에 斷代培養하였는데 Callus는 若干 增殖하였으나 器官分化는 全혀 일어나지 않았다. Anthers at the stage of tetrad, uninucleate microspores and unmaturated pollens of Lycium chinense were cultured on the modified Murashige and Skoog's medium supplemented with each 2㎎/1 2,4-D,NAA, Kinetin. Some of the microspores examined two weeks after culture became large in size and white lustrous callus emerged out of the anther. On the stages of anther development, uninuleate microspore was better than tetrad and unmaturated pollen stage on the callus formation. For differentiation of organ haploid calli originated from microspores and subcultured on the medium with 0.1㎎/1 2,4-D and 0.2㎎/1 NAA did not differentiate but developed calli.
우리나라 統一系 水稻品種의 分蘗力과 Callus 및 器官形成特性에 關한 硏究

The embryos of native, japonica and hybrid (Indica x Japonica) types were cultured on MS and Miller's media for 30 days 25℃ in order to investigate the relationship between the tillering ability and callus formation. The callus was first formed at embryo after about 1 week of shooting. As the plants were 2 to 4 centimeters, the callus was formed at the part, and the types of regeneration were different among varieties. The callus fresh weights originated from native, japonica and hybrid were 1.38, 1.23, and 0.43gr, respectively. The amount of callus formations was not different between culture media, however that of varietal origin varied. The callus fresh weight in vitro had significantly negative correlations with number of leaf and heading dates, but significantly positive correlation with percentage of productive tillers. The correlation coefficients among callus fresh weight number of total tillers and percentage of productive tillers were significant in early, medium and late groups. Also the coefficients for percentage of productive tillers were significant in different maturity groups classified with varietal origins.
