        강좌 : 클라우드 컴퓨팅을 이용한 기후변화 영향평가

        Climate change could have a pronounced impact on natural and agricultural ecosystems. To assess the impact of climate change, projected climate data have been used as inputs to models. Because such studies are conducted occasionally, it would be useful to employ Cloud computing, which provides multiple instances of operating systems in a virtual environment to do processing on demand without building or maintaining physical computing resources. Furthermore, it would be advantageous to use open source geospatial applications in order to avoid the limitations of proprietary software when Cloud computing is used. As a pilot study, Amazon Web Service . Elastic Compute Cloud (EC2) was used to calculate the number of days with rain in a given month. Daily sets of climate projection data, which were about 70 gigabytes in total, were processed using virtual machines with a customized database transaction application. The application was linked against open source libraries for the climate data and database access. In this approach, it took about 32 hours to process 17 billion rows of record in order to calculate the rain day on a global scale over the next 100 years using ten clients and one server instances. Here I demonstrate that Cloud computing could provide the high level of performance for impact assessment studies of climate change that require considerable amount of data.

      • High-level Expression of Human Immune Interferon in Escherichia coli by using tac Promoter

        인간의 면역 인터페론을 코딩하는 유전자를 tac 프로모터와 rrnBT1T2 전사중지서열을 갖는 프라스미드 pKK223-3에 클론닝하여 체세포 유전자에 lacIq 제한차서열을 갖는 대장균 JM105와 체세포 유전자에 lacIq 제한자서열을 갖는 대장균 MM294와 대장균 YMC9에 각각 형질전환시켰다. 생성된 형질전환주를 immunoblotting과 생물학적 역가분석, 전기영동 그리고 전자현미경을 사용하여 확인했다. 면역 인터페론의 발현은 대장균 JM105에서 효율적으로 조절되었으며, 대장균 MM294와 대장균 YMC9에서는 IPTG의 처리없이도 생산됨을 발견하였다. 또한 면역 인터페론이 용해된 형태와 불용성 상태인 inclusion body로 생성됨을 알았다. 이때 생산성은 $9.7{\times}10^9IU/l$이며 면역 인터페론 단백질이 대장균의 전체 단백질중 약 30%에 해당함을 알았다.         수처리용 세라믹 기반 그래핀 맴브레인의 합성 및 물질이동특성

        본 연구에서는 막 기반 수처리 기술의 핵심인 막 성능 및 특성을 혁신적으로 개선시키기 위한 새로운 형태의 막으로서 그래핀과 세라믹을 결합하는 세라믹 기반 그래핀 막(Ceramic-based graphene membrane (CbGM))을 만들고, 막에서 일어나는 물질 이동 특성을 파악하였다. 수투과 및 염 제거의 역할을 하는 활성층으로서 그래핀이 사용되었으며, 간단한 Filtrationassisted assembly (FAA) 방법을 도입하여 막을 합성하였다. 합성한 막의 표면 형태 및 특성 분석을 위해 주사전자 현미경 및 접촉각을 분석하였으며, 막 성능 및 특성 파악을 위해 3가지 용질(i.e., NaCl, MgCl₂, NA₂SO₄)을 회분식 정삼투 시스템에서 용질 이동을 측정하였다. 표면 형태 특성 분석과 물질 이동 결과를 통해, 그래핀 층의 두께 보다는, 활성층을 구성하는 그래핀조각들 사이의 교합(Interlocking)이 막에 선택성 부여함에 있어서 가장 중요함을 확인하였다. 또한, 농도차를 구동력으로 하는 막 공정에 CbGM을 적용하였을 때, 수중의 음이온과 양이온의 최외각 전하비(Z -/Z+)값이 증가할수록 용질 이동이 비례적으로 촉진되었으므로, CbGM의 표면이 양으로 하전되어 있으며, 이것이 전하된 물질의 이동에 매우 큰 영향을 끼친다는 것을 확인하였다. As a novel water treatment membrane, concept of ceramic-based graphene membrane (CbGM) was suggested, and its mass transport behavior was investigated. The selectivity of CbGM was given by graphene material which is consisting of active layer, only transmitting water, but rejecting salts. Filtration-assisted assembly methods was employed as a facile method to fabricate CbGM. Surface morphology and characteristics of CbGM were analyzed by scanning electron microscopy (SEM) and contact angle. In addition, three different kinds of solutes (i.e., NaCl, MgCl₂, NA₂SO₄) were tested in batch forward osmosis system to confirm the mass transport behavior. Through surface morphology analysis and mass transport behavior, it was revealed that interlocking between graphene layers is very important, rather than thickness of laminated graphene layers, in terms of selectivity to CbGM. All the anions in each solute showed faster transport than those of cations. In addition, solutes which have high ion valence charge ratio of anion to cation (Z-/Z+) was easier to be passed through CbGM. It indirectly implied that the surface charge of CbGM appear to be positive. In addition, It showed that surface charge of CbGM has a great role on mass transport, in particular, transport of matter having charges, generally ions.

      • cDNA Cloning and Expression of a Human Interferon-$\gamma$ in E. coli

        인간의 peripheralblood lymphocyte에서 분리된 poly (A)$^+mRNA$를 사용하여 ${\lambda}gt$ 10 cDNA library를 제조하였으며, 화학적으로 합성한 oligonucleotide를 이용하여, 이 cDNA library로부터 IFN-$\gamma$ 유전자를 갖는 DNA을 분리했다. 분리된 clone 가운데 하나인 pBK04는 IFN-$\gamma$의 전체 염기서열을 갖고 있었다. IFN-$\gamma$를 E. coli내에서 발현시키기 위해 lpp promotor를 함유하고 있 는 pINIIIA3 vector를 사용하여 IFN-$\gamma$를 발현시킬 수 있는 재조합 plasmid ($pKS{\gamma}3B$)를 제조하였다. $pKS{\gamma}3B$로써 형질 변환된 bacterial cell을 antiviral activity에 의해 역가 분석을 해본 결과, 높은 정도로 IFN-$\gamma$를 발현시켰으며 이 IFN-$\gamma$는 천연의 IFN-$\gamma$에 대하여 특이성이 있는 monoclonal antibody에 의해서도 인지되었다. High copy number를 갖는 expression vector를 제조 (pHK-3) 하였으며, 이 high copy number vector를 사용하므로써 IFN-$\gamma$의 생성이 증가되는 사실을 확인하였다. A ${\lambda}gt10$ cDNA library constructed with poly $A^+$ mRNA from human peripheral blood lymphocytes was screened by a synthetic oligonucleotide having IFN-$\gamma$ sequence. One of the clones (pBK04) which hybridized to the oligomer was confirmed to have the whole IFN-$\gamma$ coding sequence. The eDNA insert was used to express IFN-$\gamma$ in E. coli under the control of lpp promoter employing pINIIIA3 vector. Cells harboring the recombinant plasmid ($pKS{\gamma}3B$) expressed high level of IFN-$\gamma$ assayed by antiviral activity, The molecule was also recognized by monoclonal antibody specific to a natural IFN-$\gamma$. An expression vector with high copy number (pHK-3) was constructed and was successfully used for the improvement of the expression of IFN-$\gamma$.

        Chill Unit 축적과 휴면해제시기 공간변이 추정 프로그램 : CUMAP

        A chill unit has been used to estimate chilling requirement for dormancy release and risk of freezing damage. A system that calculates chill units was developed to obtain site-specific estimates of dormancy release date for grapes and evaluated in Baekgu myun near Kimje City, Chunbuk, Korea from September 2002 to March 2003. The system utilized daily minimum and maximum temperature maps generated from spatial interpolation with temperature correction for topography. Hourly temperature was temporally interpolated from the daily data using a sine-exponential equation (Patron and Logan, 1981). Hourly chill units were determined from sigmoid, reverse sigmoid, and negatively increasing sigmoid functions based on temperature ranges and summed for 24 h. Cumulative daily chill units obtained from measurements did not increase until 20 October 2002, which was used as a start date for accumulation to estimate the dormancy release date. As a result, a map of dormancy release date in the study area was generated, assuming 800 chill units as a threshold for the chilling requirement. The chill unit accumulation system, implemented using Microsoft Visual Basic and C++ (Microsoft, Redmond, WA, USA), runs in the Windows environment with ArcView (ESRI Inc., Redlands, CA, USA).
