        근대개혁기의 역사서술과 변법론

        The field of modern Korean history had to overcome premodern perspectives and systems of historiography as well as the colonial historiography from the Japanese colonal period. It attempted to discard chronologies and other earlier methods of historiography for narratives that were oriented more towards human relationships where the nation and people were the subject of history. Those peopie the base of modern historical studies were ‘reformed Confucianists’ (kaesin yuhak cha) who were active during the time of the Great Han Empire. Their modern reform theories could be called pyonbop ron(變法論) or legal reform theories. They attempted to modernize by adopting modern political theories that they then applied to a foundation based on traditional culture informed by Confucianism. In this respect,although they were inclined towards complete Westernization, their ideas of modernization differed from other enlightenment theories Their compromise between new learning and old learning, which formed the basis of their reform ideas, also attached great importance to the writing of history. At the time, this system of modern historiography was presented as ‘new history’(sinsa).They pointed out the deficiencies of historical methods as they compiled numerous histories written by Japanese. The Japanese perspective on Korean history permeated these works. Advocates of pyonbop theories who believed in the need for a nationalist approach to historical writing were harshly critical of this aspect and instead turned their focus to the historical studies of sirhak (‘practical learning’) scholars. Critical of Sino-centric approaches to history, they stressed ‘national history’ (kuksa) and carried on a tradition of evidential analysis using primary documents. The kyemong enlightenment movement that unfolded after 1905 was a movement that targeted serf-strengthening and the reclamation of national rights. Although education and industrial development was deemed necessary for self-strengthening, they also emphasized mental and spiritual elements, such as concepts of a fatherland (choguk chongsin), national spirit (kukhon), and national characteristics (kuksu). They attempted to develop national spirit and patriotism through the writing of national history, a kind of historiography that was spearheaded by none other than pyonbop theorists such as Pak Un-sik(朴殷植, 1859-1925), Sin Chae-ho(申采浩, 1880-1936) and others. Criticizing the historical writing that held China in such high esteem, they promoted the importance of national history, with particular emphasis its heroes and historical writing about heroes who triumphed over foreign invasion. By 1910, the limitations to self-strengthening and productivity theories were increasingly apparent, causing a rift within the enlightenment movement itself. Among those, Pak Un-sik and Sin Chae-ho arrived at their own distinct interpretations of the true nature of Social Darwinist theories and the despotic nature of imperialism, and came to view the new citizen (sin kungmin) and the people (kungmin) as the subjects of history. While also pointing out the mistakes of Confucianism, upon which they had been dependant, they instead emphasized Tangun and traditional religions that put forth a new historical paradigm centered on Tangun, Koguryo and Parhae. Actively pursuing such historical research in the 1910s, they presented nationalism in terms of preservation of national characteristics while they groped for new alternatives to the national movement. Into the 1920s, as theories of national spirit and national characteristics were fused together with new concepts, a modern nationalist historiography emerged. Stressing humanism and equality among citizens, they viewed the masses as a solution to the national problem. Even though Pak Un-siks The Bloody History of the Korean Independence Movement(『韓國獨立運動之血史)), written in 1920, continued to advocate national spirit, after World War I and the Russian Revolution, his historical writings focused increasingly on worldwide changes and the issue of nation and the people. Sin Chae-ho also presented a historical theory based on the struggle between the self(我) and the non-self(非我), claiming that indigenous morals was a foreign concept and arguing that the national problem had to be understood from the perspective of the masses. This ultimately formed the foundation of modern Korean historiography.

        Development of a simple and sensitive HPLCMS/MS method for determination of diazepam in human plasma and its application to a bioequivalence study

        We developed a simple, sensitive, and effective ultra-performance liquid chromatography/tandemmass spectrometry (HPLC-MS/MS) method with an electrospray ionization (ESI) interface in multiplereaction monitoring (MRM) and positive ion modes to determine diazepam concentrations inhuman plasma using voriconazole as an internal standard (IS). Diazepam and IS were detected attransition 285.2→193.1 and 350.2→127.1, respectively. After liquid-liquid extraction (LLE) using 1.2ml of ethyl acetate:n-hexane (80:20, v/v), diazepam and IS were eluted on a Phenomenex CadenzaCD-C18 column (150 × 3.0 mm, 3 μm) with an isocratic mobile phase (10 mM ammonium acetatein water:methanol [5:95, v/v]) at a flow rate of 0.4 mL/min. The peak retention time was 2.32 minfor diazepam and 2.01 min for IS, respectively. The lower limit of quantitation (LLOQ) was 0.5 ng/mL (S/N > 10) using 50 μL of plasma, and no interferences were observed in chromatograms. Ouranalytical method was fully validated and successfully applied to a bioequivalence study of two formulationsof diazepam in healthy Korean volunteers.

        저온 민감성 바실러스 서브틸리스 돌연변이 균주에서 glycine betaine의 저온 내성에 미치는 영향에 대한 연구

        높은 염분 농도에서 glycine betaine은 Bacillus subtilis 안으로 유입되어 세포 생장이 지속될 수 있게 한다. 뿐만 아니라 최근 연구 결과에 따르면 저온에서도 glycine betaine이 세포 생장을 지속시키는 것으로 알려져 있다. 저온에서 Bacillus subtilis의 생장을 저해시키는 세포 대사 활동으로는 세포막 운송과 단백질 합성을 들 수 있다. 세포막 구조와 관련하여 저온에서 세포막 운송에 영향을 주는 유전자들로는 bkdR과 des가 있고, 단백질 합성 과정에서 RNA helicase 유전자인 ydbR과 yqfR들은 저온 민감성을 보인다. 따라서 Bacillus subtilis 저온 민감성 유전자 결손 세포들에 대한 glycine betaine의 효과를 조사하여 저온에서의 glycine betaine 생리적 기능에 대해 알아보고자 하였다. 이 결과 glycine betaine의 존재 유무에 따라 야생형 Bacillus subtilis와 ydbR과 yqfR 결손 균주의 저온생장에 큰 차이를 보였다($T_d$차이 190~686 min). 반면에 bkdR이나 des 결손균주의 경우에는 glycine betaine 존재 유무에 따라 차이를 보이지 않았다. Glycine betaine의 전구체인 choline으로 대치하여도 저온에서의 생장은 같은 결과를 보였다. Glycine betaine의 영향이 세포막 구조와 관련이 있는 유전자 bkdR과 des 결손균주에 미치는 영향이 적은 것을 알아보기 위해 세포막에 영향을 주는 세제의 효과를 조사하였다. Triton X-100과 N-lauryl sarcosine 세제에 의해 bkdR 결손 균주가 야생형에 비해 더 영향 받는 것을 확인하였고 이는 bkdR 결손이 저온에서 막 구조를 변형하여 glycine betaine의 투과에 영향을 미치는 것으로 보인다. At high salt concentration, glycine betaine is transported into Bacillus subtilis and growing rate of the cell is not suppressed. Also according to recent studies, cell growth is maintained normal growth rate at low temperature. Low temperature results in a stress response of Bacillus subtilis that is characterized by strong repression of major metabolic activities such as translation machinery and membrane transport. In this regards, genes showing cold sensitive phenotype are cold-induced DEAD box RNA helicases (ydbR, yqfR) and fatty acid desaturases (bkdR, des). Therefore to understand the effect of glycine betaine on cold growth of Bacillus subtilis, we investigated the effect of glycine betaine on growth rate of these deletion mutants showing cold sensitive phenotype. Glycine betaine strongly stimulated growth of wild type Bacillus subtilis JH642 and deletion mutants of ydbR and yqfR at $20^{\circ}C$ (190~686 min $T_d$ difference). On the other hands, glycine betaine does not show growth promoting effects on deletion mutants of bkdR, and des at cold conditions. Same cold protectant growth results were shown with the precursor choline instead of glycine betaine. We investigated the effects of detergents on the cell membrane in bkdR and des deficient strains associated with cell membrane. It was identified that bkdR deficient strain shows retarded growth with detergent such as Triton X-100 or N-lauryl sarcosine compared with wild type cell. Thus, it is possible that deletion mutation of bkdR modifies membrane structure and effects on transport of glycine betaine.

        LDAP기반의 산업기술 유출방지에 관한 연구

        본 연구에서는 LDAP(Lightweight Directory Access Protocol)디렉터리 서비스를 기반으로 강화된 인증 및 보안체계와 사용자실명화 체계를 구축하여 인증된 사용자 및 전산자산만이 네트워크 자원 및 전산자원을 사용할 수 있도록 하고 보안프로그램의 강제 설치화로 보안프로그램 무력화에 대응하고 기존의 산업기술 유출방지 방법과의 연동을 통하여 사용자 기반의 보안정책 적용과 더불어 산업기술 유출자를 정확하게 추적할 수 있는 방법에 대해 연구하였다. 본 연구을 통해 기존이 산업기술 유출방지 관리체계를 강화하고 산업기술 유출방지 방법의 취약점을 보완하여 통합된 보안 인프라체계를 구현함으로써 강화된 산업기술 유출방법을 구현할 수 있다. This study researched into the method that allows only the certified user and computational engineer to possibly use network resources and computing resources by implementing the system of the intensified certification and security based on LDAP(Lightweight Directory Access Protocol) directory service, that copes with incapacitation in security program due to making the security program forcibly installed, and that can correctly track down the industrial-technology exporter along with applying the user-based security policy through inter-working with the existing method for the protection of industrial technology. Through this study, the intensified method for the protection of industrial technology can be embodied by implementing the integrated infra system through strengthening the existing system of managing the protection of industrial technology, and through supplementing vulnerability to the method for the protection of industrial technology.

        Black-box Modeling for Aircraft Maneuver Control with Bayesian Optimization

        This paper proposes a new method of designing a data-driven controller for aircraft maneuver. Assuming that we do not have knowledge of the controller and the controlled aircraft, we propose a controller design with explorations of the control inputs and their responses from the aircraft. Specifically, we utilize Bayesian optimization (BO) with Gaussian process (GP) regression for black-box modeling of the aircraft responses from the explored controls, which are selected as samples to experiment with BO. We tested the proposed controller with a rigid six degrees of freedom(6DoF) nonlinear aircraft model by varying the kernel structures of the GPregressions. Our proposed method shows shorter flight times and smaller deviations navigating fixed waypoints compared to the tuned Proportional Integral Derivatives (PID) controller. The proposed controller can be an alternative to PID control, particularly when both controller structure and controlled plant model information are unknown.
