Lysosomal enzyme activity in the retinal pigment epithelium (RPE) of rabbit eyes was measured using acid phosphatase(ACPase) as an enzyme marker. Distributions of ACPase in RPE were measured by standard enzymological techniques. The REP and choroidaltissues were first homogenated and then centrifuged. Sediments were RPE which wasconfirmed electron microscopically. In the subcellular fractions of RPE, the specific activity of the ACPase was most Prominent in the Iysosomal fraction (80.15± 26.82?g p-nitrophe-nol/mg protein by Fukuzawa method). The nuclear (31.60± 19.38), mitochondrial (27.87± 18.23) and soluble fraction (53.63± 21.70) also showed the specific of the enzyme.The preparation of microsomal fraction showed a trace of the specific acticity of the engyme.
새삼(Cuscuta japonica Choisy)및 실새삼(C. australis R.Be) 추출물의 여드름 유발균 Propionibacterium acnes 증식 억제 효과

According to the traditional chinese herbal Cuscuta japonica Choisy and C. australis R.Be (original plant of Semen Cuscutae in Korea) have been used to cure pimple. In order to prove their bacteriostatic effects against Propionibacterium acnes, we performed the micro broth dilution method. The minimal inhibitory concentration (MIC) of C. japonica juice was 27.3 mg/ml, but C. australis juice showed no effect. The MIC of water extracted C. japonica seed and Wontosa and Bupjetosa were 960 μg/ml. The MIC of both water and ethanol extracts of C. australis seed was 240 μg/ml. But ethanol extracts of C. japonica seeds, Wontosa and Bupjetosa had no bacteriostatic activity. We also performed the disk diffusion test, C. japonica juice had prominent effect. Water extracts of C. japonica seed, Wontosa and Bupjetosa showed bacterio-static effect in 10 mg/ml, and C. australis seed in 10 mg/ml. The ethanol extract of C. japonica seed, Wontosa, Bupjetosa, and of C. australis seed showed bacteriostatic effect in 20 mg/ml.

In continuation of studies on the umbelliferous fruits, inner morpholoy of Coelopieurum fuits was examined and its results are discussed in comparison with those of angelica Archangelica, Ostericum, Dystaenia, Ligusticum of Tilingla and Cnidium. The differences between Coelopleurum and the others are as fallows. arrangement of vascular bumdlis is the same as that of angelica and rehexigenetic intercelluar space at mesocarp in mature fruits is simillar to that of Archangelica, and Ostericum.

Cathepsin D was purified from rabbit eyes homogeneously. The homogeneity of the preparation was determined by SDS-polyacrylamide gel electrophoresis and densitometer trace. The isolation scheme involved an homogenization, acid treatment, ammonium sulfate fraction, and sephadex G-200 chromatography. The end product represent about a 61. 3-fold purification, and a 26.1% recovery. The apparent molecular weight was estimated to be 63,000 by electrophoresis in polyacrylamid gel containing sodium dodecyl sulfate according to the method of Weber and Osborn.

In continuation of studies on the umbelliferous fruits inner morphology of kanwhal fruits was ixammined This plant was known the origin of korean "Qianghuo" and its scientific name was Angelica koreana Maxim-in general. The arrangement of vascular bundles was the same as Angelica group (Angelica, Ostericum,, Dystaenia), and among these. the other characters was the same as Ostericum.

Acid phosphatase [Orthophosphoric monoester phosphohydrolase(acid optimum), EC 3.1.3.2, ACPase] was purified from the retinal pigment epithelium of rabbit eyes near homogeneity. The purification procedure included lysosomal fractionation, acid treatment, ammonium sulfate fractionation, gel filtration chromatography on sephadex G-100 and ion exchange chromatography on DEAE-cellulose. ACPase was purified about 47 fold and it is mollecular weight was estimated to be approximately 126 kDa, and consists of two identical subunits(54 kDa) by Sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The optimum temperature of ACPase activity determined with p-nitirophenyl phosphate was 60℃. ACPase was inhibited by L-(+)-tartarate, fluoride, cupric ion, zinc ion and mercuric ion, whereas activated by magnesium ion, reducing agents(cystein, 2-mercaptoethanol or ascorbic acid) and EDTA. Although the enzyme appears to be the most activity toward P-nitrophenyl phosphate, marked activity was observed with Glucose-1-phosphate. ATP, ADP and AMP were not hydrolyzed by ACPase.
새삼(Cuscuta japonica Choisy)및 실새삼(C. australis R.Be) 추출물의 Mushroom Tyrosinase 활성 억제 효과

The purpose of this study was to evaluate mushroom tyrosinase inhibitory activity of Cuscuta japonica Choisy and C. australis R.Be. The experimental materials were expressed juice from their stems and flowers, both water and ethanol extracts, their seeds, and two kinds of commercially available cosmetic packing Wontosa and Bupjetosa (made from seeds of C. japonica.). The 50% inhibitory concentration (IC50) of C. japonica juice was 5.4 mg/ml. However, C. australis juice showed negligible mushroom tyrosinase inhibitory activity. The IC50 of water extracted C. japonica seed was 54.0 μg/ml, water extracted product of Wontosa 50.0 μg/ml and Bupjetosa 40 μg/ml. The IC50 of ethanol extracted C. japonica seed was 10 μg/ ml, Wontosa 10 μg/ml and Bupjetosa 20 μg/ml.
