This study is designed to identify burnout factors that workers at social welfare organizations experience. That is, this study attempts to examine the influence of independent variables(workers` personal characteristics and organizational characteristics) and parameters(organizational commitment of workers) on burnout, in identifying the degree of burnout in workers in social welfare organizations. In particular, it attempts to establish a relationship between antecedent variables and resultant variables by verifying the intermediary role of organizational commitment. According to the results of verifying the differences among groups regarding personal characteristics, demographic variables that affect self-esteem significantly include marital status and organization type. This affects use organization workers more than those of accommodation organizations. Married, highly educated, and use organization workers experience more burnout than singlet, less educated, and accommodation organization workers.
Streptomyces floridae SHS-1372가 생산하는 항암항생물질 HS-1의 특성

토양에서 분리한 Streptomyces floridae SHS-1372로부터 항암 항생물질의 screening 과정중에 강한 항암활성을 나타내는 항암 항생물질 HS-1을 용매 추출, silica gel column chromatoghaphy를 통해서 분리 정제하였다. HS-1의 UV, FAB-MS, IR spectrum, ^1H-NMR과 ^(13)C-NMR, 2D-NMR등의 기기분석에 의해 actinomycin X_2로 동정되었다. Streptomyces floridae로부터 actinomycin X_2가 분리된 적이 없었다. Antitumor antibiotic HS-1 was purified from the culture broth of a streptomyces floridae SHS-1372 which had been isolated from soil, by solvent extraction, silica gel column chromatography and gel filtration. It was confirmed that HS-1 was active against gram positive bacteria and cancer cells(K562, P388, MCF-7, HT-29). Through the analysis of UV spectrum, melting point, IR spectrum, FAB-MS, ^1H-NMR, ^(13)C-NMR, 2D-NMR spectra, HS-1 could be identified as the actinomycin X_2 antibiotics containing actinocine chromophore and peptides consisted of threonine, proline, methylvaline, sarcosine, D-valine and 4-ketoproline.

In order to isolate and purify of antimicrobial substances extracted from the dried fruits of Schizandra chinensis, the antimicrobial substances from the methanol extract which exhibited a strong antimicrobial activity was purified by solvent fractionation, prep. TLC, silica gel column chromatography, prep. HPLC. The results are summarized as follows among the fractions fractionated by 4 kinds from the methanol extract, the antimicrobial activity of ethyl acetate fraction had the strongest antimicrobial activity against bacteria. And their chemical structures were determined by TLC, Mass, UV and NMR spectra analysis. The results are summarized as follows. 1. One compound having antimicrobial activity was isolated from the ethyl acetate fraction from total methanol extract. 2. Heating test on ethyl acetate fraction from Schizandra chinensis is stabilized. 3. Ethyl acetate fraction from Schizandra chinensis is antimicrobial activity on acidity. 4. The isolated compound showed antimicrobial activity having MIC range 30-500/㎍/disk against Bacillus subtilis, Micrococcus luteus and Pseudomonas aeruginosa. 5. The chemical structure of compound was identified as trimethylcitrate.
KB암세포에 효과있는 Streptomyces plicotosporus가 생산하는 항암증강물질 Rubiginone B_2에 관한 연구

최근 문제가 되고 있는 항암내성에 대해 내성을 극복하는 물질을 탐색하던 중 사람 상피암 세포주인 KB-C2에 내성을 극복하는 균주를 분리하고 그 균주가 생산하는 항생물질을 분리, 정제한 후 구조를 결정하였고 정제된 물질에 대한 항암증강활성을 조사하였다. 분리된 균주 2238-SVT4 균주는 Streptomyces plicatosporus라는 새로운 균주로 판명되었다. 또한 균체에서 ethyl acetate extraction, silicagel column chromatography, gel filtration 등을 통하여 항암증강물질을 정제한 후 UV, EI-MS, ^1H-NMR, ^13C-NMR등의 기기분석을 한 결과 본 물질은 rubiginone B_2로 결정되었다. Rubiginone B_2는 KB-C2 세포주에서 처음으로 다제내성을 극복하는 것으로 나타났다. It was confirmed that HS-2 enhanced the cytotoxicity of colchicine against multidrug-resistant tumor cells.
Actinomycin X_2를 생산하는 방선균 분리주 No.1372의 동정

토양 시료로부터 순수분리한 2500여 방선균주중에 항암 활성을 나타내는 균주 No.1372를 선별하였으며, 이 균주 No.1372를 ISP법과 화학적 방법에 의해 동정하였다. 분리주 No.1372의 포자 배열은 rectus flexible이었으며 포자 표면은 smooth로써 DAP isomer 분석에 의해 세포벽 유형은 wall chemotype I으로 결정되었으며, 형태학적 특성, 생리학적 특성등을 종합하여 볼 때 분리주 No.1372는 Streptomyces floridae로 동정되었다. 이 균주로부터 항생물질 생산의 최적 조건으로는 탄소원에 soluble starch, 질소원에 yeast extract와 (NH_4)_2SO_4 무기염에는 Fe^(2+), Mg^(2+), Ag^(2+), Zn^(2+) 순으로 나타났으며 초기 배양 pH와 온도 각각 pH 6.0, 28∼30℃ 조건이 가장 좋았다. Identification of the Actinomycetes isolate strain NO.1372, a producer of Actinomycin X_2 was performed by using ISP method. The strain, designated as No.1372, was identified as Streptomyces floridae based on its morphological, physiological and biochemical characteristics. The highest production of the antibiotics by the strain was achieved in a fermentation medium containing soluble starch, yeast extract, (NH_4)_2SO_4, K_2HPO_4, NaCl, CaCO_3, and trace element.
