Zinc finger-containing transcription factors are the largest single family of transcriptional regulators in mammals, which play an essential role in cell differentiation, cell proliferation, apoptosis, and neoplastic transformation. Here we have cloned a novel KRAB-related zinc finger gene, ZNF424, encoding a protein of 555aa. ZNF424 gene consisted of 4 exons and 3 introns, and mapped to chromosome 19p13.3. ZNF424 gene was ubiquitously expressed in human embryo tissues by Northern blot analysis. ZNF424 is conserved across species in evolution. Using a GFP-labeled ZNF424 protein, we demonstrate that ZNF424 localizes mostly in the nucleus. Transcriptional activity assays shows ZNF424 suppresses transcriptional activity of L8G5-luciferase. Overexpression of ZNF424 in HEK- 293 cells inhibited the transcriptional activity of NFAT and p21, which may be silenced by siRNA. The results suggest that ZNF424 protein may act as a transcriptional repressor that suppresses NFAT and p21 pathway to mediate cellular functions. [BMB reports 2010; 43(3): 212-218]

Purpose: Human papillomavirus (HPV) infection plays an important role in the development of cervical cancer, but the prevalence of HPV infection in women of Shenzhen city remains unclear. The present study was performed to describe the change of cervical HPV infection in females who participated in voluntary cervical cancer screening from 2006 to 2010 in Shenzhen city, China. Methods: A total of 4, 413 women were recruited. HPV infections were genotyped by polymerase chain reaction (PCR) and reversed dot blot hybridization in Shenzhen Maternity and Child Health Hospital. Results: The prevalence of HPV infection was 13.8%. The five most commonly found HPV types were HPV16 (3.47%), HPV58 (1.68%), HPV33 (1.38%), HPV43 (1.36%) and HPV18 (1.27%). The secular trends of major HPV type-specific were diverse. Among of them, the prevalence of HPV18 increased sharply while others increased slowly or even decreased in the period. The change of total HPV, single HPV and multiple HPV infection were similar during the five years. Conclusions: Our findings suggested that HPV infection is common with HPV16 and HPV 58 as the primary subtypes in women in Shenzhen city. The prevalence of HPV 18 infection is increasing faster than any others, which will lead it to be one of the main subtypes in this city in the future.

The generated electric porcelain waste increases with the rapid development of the electric porcelain industry. Unreasonableutilization results in the waste of resources and environmental pollution. However, at present, many coal gangues and fly ashesare wasted and underutilized in my country. In this paper, electric porcelain waste is used as the main raw material, and itssintering temperature, holding time, and heating rate are discussed to study the performance of the permeable brick preparedfrom electric porcelain waste. Results show that with the increase in sintering temperature, the porosity and waterpermeability coefficient of permeable bricks decrease, and their volume shrinkage rate and compressive strength show anincreasing trend. Furthermore, porosity decreases with the increase in holding time. The volume shrinkage rate andcompressive strength increase, and the water permeability coefficient decreases. The volume shrinkage rate of the permeablebrick decreases, and its porosity, water permeability coefficient, and compressive strength increase with the heating rate. Combined with the comprehensive consideration of the porosity, volume shrinkage rate, water permeability coefficient, andcompressive strength of permeable bricks, the best sintering process for preparing permeable bricks from electrical porcelainwaste is presented with a sintering temperature of 1225 °C, holding time of 1 h, and heating rate of 6 °C/min. The porosityof the prepared permeable brick is 79.5%, the volume shrinkage rate is 25.79%, the water permeability coefficient is 2.82×10-2cm/s, and the compressive strength is 5.79 MPa.

LBH is a transcription factor as a candidate gene for CHD associated with partial trisomy 2p syndrome. To identify potential LBH-interacting partners, a yeast two-hybrid screen using LBH as a bait was performed with a human heart cDNA library. One of the clones identified encodes αB-crystallin. Co-immunoprecipitation and GST pull-down assays showed that LBH interacts with αB-crystallin, which is further confirmed by mammalian two-hybrid assays. Co-localization analysis showed that in COS-7 cells, αB-crystallin that is cytoplasmic alone, accumulates partialy in the nucleus when co-transfected with LBH. Transient transfection assays indicated that overexpression of LBH or αB-crystallin reduced the transcriptional activities of p53 and p21, respectively, Overexpression of both αB-crystallin and LBH together resulted in a stronger repression of the transcriptional activities of p21 and p53. These results showed that the interaction of LBH and αB-crystallin may inhibit synergistically the transcriptional regulation of p53 and p21. [BMB reports 2010; 43(6): 432-437]

In this study, we report the identification and characterization of a novel C2H2 zinc finger protein, ZNF552, from a human embryonic heart cDNA library. ZNF552 is composed of three exons and two introns and maps to chromosome 19q13.43. The cDNA of ZNF552 is 2.3 kb, encoding 407 amino acids with an amino-terminal KRAB domain and seven carboxyl-terminal C2H2 zinc finger motifs in the nucleus and cytoplasm. Northern blotting analysis indicated that a 2.3 kb transcript specific for ZNF552 was expressed in liver, lung, spleen, testis and kidney, especially with a higher level in the lung and testis in human adult tissues. Reporter gene assays showed that ZNF552 was a transcriptional repressor, and overexpression of ZNF552 in the COS-7 cells inhibited the transcriptional activities of AP-1 and SRE, which could be relieved through RNAi analysis. Deletion studies showed that the KRAB domain of ZNF552 may be involved in this inhibition. [BMB reports 2010; 43(3): 193-198]

Yun Xue,Yue-Lin Wang,Yu Li,Wen-Da Xu,Fu-Qiu Ma,Yang-Hai Zheng,Qing-Guo Zhang,Zhi Zhang,Mi-lin Zhang,Yong-De Yan 한국공업화학회 2023 Journal of Industrial and Engineering Chemistry Vol.126 No.-
Cation exchange resins (CERs) are widely applied to purify waste liquids generated during the operationof nuclear reactors. The radioactive nuclides 60Co and 58Co are important corrosion activation products inreactor cooling water. In this study, the simulated Co doped CERs were oxidized with ternary carbonate. According to the thermogravimetric analysis (TG), the decomposition of Co doped CERs includes threeprocesses: 1. Elimination of the osmotic water; 2. Pyrolysis of sulfonic acid group; 3. Destruction of styrene–divinylbenzene copolymer. The X-Ray Diffraction (XRD) patterns indicate that sulfur mainly exists inthe form of sulfate in waste salt. The Co2+ undergoes the path of CoS2 ? Co3O4 with the increase of temperatureand the transition point is 650 C. Combined with Fourier Transform Infrared Spectrometer (FTIR)spectra and X-ray Photoelectron Spectroscopy (XPS) analysis, sulfonic acid groups begin to decomposeat 350 C. During the molten salt oxidation process, most of the sulfur in sulfonic acid groups is entrappedby carbonate as the form of sulfate, and a little of which remains as sulfone group, sulfoxide group andsulfur bridge in residue. When the resins are oxidized at 800 C, the retention rate of Co2+ is 97.3%, indicatingthat the molten salt oxidation can effectively remain Co2+ and convert it into a more stablesubstance.

This study is on how ohmic area etching affects the buffer breakdown voltage of AlGaN/GaN HEMT. The surface morphology of the ohmic metal can be improved by whole etching on the ohmic area. The buffer breakdown voltages of the samples with whole etching on the ohmic area were improved by the suppression of the metal spikes formed under the ohmic contact regions during high-temperature annealing. The samples with selective etching on the ohmic area were investigated for comparison. In addition, the buffer leakage currents were measured on the different radii of the wafer, and the uniformity of the buffer leakage currents on the wafer were investigated by PL mapping measurement.

Zinc finger (ZNF) proteins play a critical role in cell growth, proliferation, apoptosis, and intracellular signal transduction. In this paper, we cloned and characterized a novel human KRAB-related zinc finger gene, ZNF425, which encodes a protein of 752 amino acids. ZNF425 is strongly expressed in the three month old human embryos and then is almost undetectable in six month old embryos and in adult tissues. An EGFP-ZNF425 fusion protein can be found in both the nucleus and the cytoplasm. ZNF425 appears to act as a transcription repressor. Over-expression of ZNF425 inhibits the transcriptional activities of SRE, AP-1, and SRF. Deletion analysis indicates that the C2H2 domain is the main region responsible for the repression. Our results suggest that the ZNF425 gene is a new transcriptional inhibitor that functions in the MAPK signaling pathway. [BMB reports 2011; 44(1): 58-63]

Compared with the conventional steel structure, the high-strength steel structures are at more risk of brittle fracture, especiallyin cold regions. In the present study, a series of tests (such as uniaxial tensile test, Charpy impact test and three-point bendingtest) were carried out at low temperature to investigate the mechanical properties and toughness of Q460C steel and its buttwelded joint, fracture micro-mechanisms were analyzed as well. The ductility indices and the toughness indices all decreasewith temperature decreases, the heat affected zone (HAZ) in welded joint is more critical to fracture than the base material. The fracture toughness of high-strength steel Q460C is relatively lower than the other three conventional steels (i.e. Q235, Q345and Q390). In this study, rich experimental data were collected so as to provide reference for the fracture resistant design ofhigh-strength steel structures in cold regions.
