This study tested the relative and combined efficacy of ShPx2 and ShPAL transgenes by comparing Nicotiana tabacum hybrids with enhanced levels of $_L$-phenylalanine ammonia lyase (PAL) activity and cationic peroxidase (Prx) activity with transgenic parental lines that overexpress either transgene. The PAL/Prx hybrids expressed both transgenes driven by the 35S CaMV promoter, and leaf PAL and Prx enzyme activities were similar to those of the relevant transgenic parent and seven- to tenfold higher than nontransgenic controls. Lignin levels in the PAL/Prx hybrids were higher than the PAL parent and nontransgenic controls, but not significantly higher than the Prx parent. All transgenic plants showed increased resistance to the necrotrophs Phytophthora parasitica pv. nicotianae and Cercospora nicotianae compared to nontransgenic controls, with a preponderance of smaller lesion categories produced in Prx-expressing lines. However, the PAL/Prx hybrids showed no significant increase in resistance to either pathogen relative to the Prx parental line. These data indicate that, in tobacco, the PAL and Prx transgenes do not act additively in disease resistance. Stacking with Prx did not prevent a visible growth inhibition from PAL overexpression. Practical use of ShPAL will likely require more sophisticated developmental control, and we conclude that ShPx2 is a preferred candidate for development as a resistance transgene.

포스트 모던 사회에서 관광은 떠날 때의 기대와 희망, 돌아올 때의 기쁨과 평화를 담보해주는 자기 후생적 활동으로 인식되고 있다. 따라서 현대인들은 실존의 확인을 위해 탈 도시화, 탈 일상화를 추구하고 있다. 우리나라는 1989년 1월 1일 해외여행 자유화 조치 이후 아웃바운드 국제관광객수는 지속적인 증가 추세를 보여왔다. 최근에는 체재형 관광 활동을 선호하면서 한국인들도 점차 도서 관광지역을 찾는 경향성이 두드러지고 있다. 특히 괌은 태평양 지역 중 우리나라와의 지리적 접근성이 매우 뛰어난 지역으로 일본인 다음으로 제2의 관광시장으로 부상하였다. 따라서 이 지역에서의 한국인 관광객들의 관광관련 특성을 밝히는 것은 상당한 의미가 있다. 이러한 측면에서 본 연구는 괌 지역의 한국인 관광객들의 관광활동 특성을 밝히는 것이다. 본 연구를 위한 출구조사는 괌 아가나(Agana) 국제공항에서 2000년 1월 24일부터 28일 까지 5일간 실시되었다. 그 결과 대부분의 한국인 관광객들은 신혼여행 목적을 패키지 관광형태로 괌을 선택하여, 관광 관련 정보는 인터넷을 통해 얻는 것으로 확인되었다. 괌의 관광환경은 전반적으로 높게 평가되었고 친절한 서비스, 가이드의 정직성, 괌의 안전성 등이 관광객들의 출발전 기대보다 높게 나타났다. 현지에서의 관광활동은 해양 스포츠 활동이 주를 이루며, 대표적인 방문지역으로는 연인절벽(Two Lovers Point)과 아가나(Agana) 지역으로 확인되었다. 또한 관광가이드의 투어서비스와 여행사의 패키지 상품 관련사항에서는 GITs의 경우 투어관련 불만이 가장 높았고 한국인 관광 가이드에 대해서는 긍정적인 평가가 나타났다.

Haraldiophyllum hawaiiense sp. nov. is described as a new mesophotic alga and a new genus record for the Hawaiian Islands. Six specimens were collected at a depth range of 81-93 m from Papahānaumokuākea Marine National Monument, and their morphology investigated, as well as molecular phylogenetic analyses of the plastidial ribulose-1,5-bisphosphate carboxylase-oxygenase large-subunit (rbcL) gene and a concatenated alignment of rbcL and nuclear large-subunit rRNA gene (LSU) sequences. Phylogenetic analyses supported H. hawaiiense sp. nov. as a distinct lineage within the genus Haraldiophyllum, and sister to a large clade containing the type species, H. bonnemaisonii, as well as H. crispatum and an undescribed European specimen. The six Hawaiian specimens were shown to be identical, but unique among other species of the genus as well as the recently segregated genus Neoharaldiophyllum, which comprises half of the species previously included in Haraldiophyllum. The vegetative morphology of H. hawaiiense sp. nov. resembles Neoharaldiophyllum udoense (formerly H. udoensis); however, no female or post-fertilization structures were found in the Hawaiian specimens to allow a more comprehensive comparison. The molecular phylogenies demonstrate that Haraldiophyllum is paraphyletic, suggesting either that the Myriogrammeae tribe includes undescribed genera, including Haraldiophyllum sensu stricto, or that Neoharaldiophyllum species should be transferred into the genus Haraldiophyllum. However, based on vegetative morphology and molecular analyses, and pending resolution of this taxonomic issue, the Hawaiian specimens are placed within the genus Haraldiophyllum. This new record for the Hawaiian Islands highlights the novel biodiversity from mesophotic depths, reaffirming the need for further investigation into the biodiversity of Mesophotic Coral Ecosystems.
Surgical Intervention to Rescue Hirschsprung Disease in a Rat Model

Background/Aims Rats with a spontaneous null mutation in endothelin receptor type B or Ednrb (sl/sl; spotting lethal) lack enteric neurons in the distal bowel and usually die within the first week after birth. This early postnatal lethality limits their use for examining the potential of cell therapy to treat Hirschsprung disease, and for studies of the influence of EDNRB on the mature CNS and vascular systems. Methods We have developed a surgical intervention to prolong the life of the spotting lethal sl/sl rat, in which we perform a colostomy on postnatal (P) day 4-6 rats to avoid the fatal obstruction caused by the lack of colonic enteric neurons. Results The stomas remained patent and functional and the rats matured normally following surgery. Weight gains were comparable between control and Hirschsprung phenotype (sl/sl) rats, which were followed until 4 weeks after surgery (5 weeks old). We confirmed the absence of enteric neurons in the distal colon of rats whose lives were saved by the surgical intervention. Conclusions This study provides a novel approach for studying EDNRB signalling in multiple organ systems in mature rats, including an animal model to study the efficacy of cell therapy to treat Hirschsprung disease. <P>Adoptive transfer of peripheral blood–derived, melanoma-reactive CD8<SUP>+</SUP> cytotoxic T lymphocytes (CTLs) alone is generally insufficient to eliminate bulky tumors. Similarly, monotherapy with anti-CTLA4 infrequently yields sustained remissions in patients with metastatic melanoma. We postulated that a bolus of enhanced IL-21–primed polyclonal antigen-specific CTL combined with CTLA4 blockade might boost antitumor efficacy. In this first-in-human case study, the combination successfully led to a durable complete remission (CR) in a patient whose disease was refractory to both monoclonal CTL and anti-CTLA4. Long-term persistence and sustained anti-tumor activity of transferred CTL, as well as responses to nontargeted antigens, confirmed mutually beneficial effects of the combined treatment. In this first-in-human study, Chapuis et al. demonstrate that the combination of adoptive cellular therapy with CTLA4 blockade induces long-term remission in a melanoma patient resistant to both modalities administered serially and individually.</P>
