A gene related to high pristinamycin yield in Streptomyces pristinaespiralis was selected by amplified fragment length polymorphism (AFLP) and its functions were investigated by gene disruption. First, a 561 bp polymorphic sequence was acquired by AFLP from high-yield recombinants compared with the S. pristinaespiralis ancestor ATCC25486, indicating that this approach is an effective means of screening for valuable genes responsible for antibiotic yield. Then, a 2,127 bp open reading frame of a gene designated spy1 that overlaps with the above fragment was identified and its structure and biological functions were investigated. In silico analysis of spy1 encoding a deduced 708-amino-acid-long serine/threonine protein kinase showed that it only contains a catalytic domain in the N-terminal region, which is different from some known homologs. Gene inactivation of chromosomal spy1 indicated that it plays a pleiotropic regulatory function in pristinamycin production, with a positive correlation to pristinamycin I biosynthesis and a negative correlation to pristinamycin II biosynthesis.
High Performance Current Sensing Circuit for Current-Mode DC-DC Buck Converter

A simulation study of a current-mode direct current (DC)-DC buck converter is presented in this paper. The converter, with a fully integrated power module, is implemented by using sense method metal-oxide-semiconductor field-effect transistor (MOSFET) and bipolar complementary metal-oxide-semiconductor (BiCMOS) technology. When the MOSFET is used in a current sensor, the sensed inductor current with an internal ramp signal can be used for feedback control. In addition, the BiCMOS technology is applied in the converter for an accurate current sensing and a low power consumption. The DC-DC converter is designed using the standard $0.35\;{\mu}m$ CMOS process. An off-chip LC filter is designed with an inductance of 1 mH and a capacitance of 12.5 nF. The simulation results show that the error between the sensing signal and the inductor current can be controlled to be within 3%. The characteristics of the error amplification and output ripple are much improved, as compared to converters using conventional CMOS circuits.
Physiological responses of yellow‑horn seedlings to high temperatures under drought condition

Drought and hot stresses are the primary limiting factors for the growth and seed production of yellow-horn (Xanthoceras sorbifolia Bunge), especially in Xinjiang Province of China. A better understanding of its physiological and biochemical characteristics under drought–hot stress is requisite for its efficient cultivation in the arid and semi-arid areas. In this study, the physiological and biochemical responses of 3-month-old yellow-horn seedlings were evaluated after 7 days of treatments of five temperatures (25 °C/20 °C, 30 °C/25 °C, 35 °C/25 °C, 40 °C/25 °C and 45 °C/25 °C) under two watering conditions [adequate water supply (HW): 80 ± 5% FC (field capacity); low water supply (LW): 40 ± 5% FC] by a pot experiment. The increased temperature (above to 35 °C) markedly aggravated the damage on biomass accumulation and membrane integrity of yellow-horn seedlings under the LW. The accumulation of soluble sugars and proteins exhibited a growing trend as the temperature increased from 25 to 40 °C, which declined when the temperature was higher than 40 °C except for the soluble protein accumulation under HW. In addition, captured responsive characteristics of the gas-exchange parameters and chlorophyll fluorescence have indicated that the combinations of high temperature (above to 35 °C) and LW induced significant decrease in photosynthetic activities of yellow-horn seedlings. All these results showed that the hot stress significantly aggravated the drought damage on yellow-horn plant growth, especially when the temperature increased above 35 °C under drought stress.
Role of miR-511 in the Regulation of OATP1B1 Expression by Free Fatty Acid

MicroRNAs (miRNAs) are a family of non-coding RNA that are able to adjust the expression of many proteins, including ATPbinding cassette transporter and organic cation transporter. We sought to evaluate the effect of miR-511 on the regulation of OATP1B1 expression by free fatty acids. When using free fatty acids to stimulate Chang liver cells, we found that the expression of miR-511 increased significantly while the expression of OATP1B1 decreased. We also proved that SLCO1B1 is the target gene of miR-511 with a bioinformatics analysis and using the dual luciferase reporter assay. Furthermore, the expressions of SLCO1B1 and OATP1B1 decreased if transfecting Chang liver cells with miR-511, but did not increase when transfecting the inhibitors of miR-511 into steatosis cells. Our study indicates that miR-511 may play an important role in the regulation of OATP1B1 expression by free fatty acids.
Association between Fecal Bile Acids and Colorectal Cancer: A Meta-analysis of Observational Studies

Purpose: To provide a systematic review with meta-analysis for addressing the relationship between fecal bile acids (FBAs) and colorectal cancer. Materials and Methods: Electronic databases were searched for all observational studies that examined the relationship between FBAs and colorectal cancer or adenoma, and calculated weighted mean difference (WMD) and 95% confidence interval (CI). Publication bias was assessed with funnel plot. Results: Twenty case-control or cohort studies were identified. All studies were pooled to assess the relationship between total FBAs and cancer/adenoma of the large bowel, however, no association was seen (WMD 0.61mg/g freeze-dried feces; 95% CI: -0.35-1.57). Significantly increased concentration of chenodeoxycholic acid (CDCA) was seen while pooling to assess the relationship between CDCA and cancer/adenoma of the large bowel (WMD 0.13 mg/g freeze-dried feces; 95% CI: 0.01-0.25), especially for colorectal cancer (WMD 0.28mg/g freeze-dried feces; 95% CI: 0.10-0.46). However, no significant differences in deoxycholic acid (DCA), lithocholic acid (LCA), and primary and secondary bile acids, were seen between patients with cancer and patients with matched controls regardless of fixed and random effects models. Conclusion: CDCA might play a role in the etiology of colorectal cancer.

In this work, a highly sensitive method using a colorimetric probe coupled to dispersive liquid?liquid microextraction (DLLME) was developed for the quantitative determination of dopamine (DA) in serum. The DA in serum was concentrated by DLLME to increase the detection sensitivity and reduce the matrix effects. After the DLLME process, a colorimetric probe of silver triangular nanoparticles (AgTNPs) was used to detect DA, which was based on the plasma transformation of AgTNPs caused by strong interactions with melamine (MA). The results showed that DA could inhibit the aggregation of AgTNPs induced by MA, resulting in the recovery of the surface plasmon resonance (SPR) peak of AgTNPs. Thus, the DLLME method followed by colorimetric probe detection of DA can be achieved. The parameters affecting the proposed method were optimized, under the optimal conditions, a linear calibration curve was obtained over a concentration range of 5 to 250?nM with a recovery from 94.4 to 101.3%. The detection limit was 1.6 nM (at an S/N ratio of 3). The present method was successfully applied to determine DA in human serum.

Two new complexes with 5-methyl-1-(pyridine-3-yl)-1H-1,2,3-triazole-4-carboxylic acid (Hmptc) ligand: [Cd(mptc)2(H2O)4] (1) and [Cu(mptc)4·2H2O]n (2) were prepared and their crystal structures were determined by single crystal X-ray diffraction analyses. In complex 1, the Cd(II) ions coordinates with the pyridyl nitogen atom from the Hmptc ligand, forming a mononuclear Cd(II) compound. Complex 2 exhibits a novel twodimensional (2D) polymer in which four Hmptc ligands stabilize the Cu(II) atom. And the coordination involves one nitrogen atom of the triazole, one oxygen atom of the carboxylic acid and the pyridyl nitrogen atom. In addition, FT-IR and solid-state fluorescent emission spectroscopy of two compounds have been determined.
