        RNA sequencing reveals that Prx II gene knockout can down-regulate the allograft rejection of dermal mesenchymal stem cells

        In this study, we used RNA sequencing (RNA-seq) to analyze and compare bulk cell samples from wild-type (WT) dermal mesenchymal stem cells (DMSCs) (n = 3) and Prx II knockout DMSCs (n = 3). The purpose of the study was to elucidate the role of Prx II on allogeneic immune rejection of transplanted DMSCs. The results revealed differential expression of 472 genes (176 up-regulated and 296 down-regulated; p ≤ 0.05) between the PrxII+/+ (WT) and PrxII−/− sample groups. When highly regulated genes were categorized according to the Gene Ontology (GO) molecular function classification and the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis, the PrxII−/− samples showed a robust downward trend in allograft rejection. The study identified 43 all immunologically rejected differentially expressed genes, of which 41 showed lower expression in the PrxII−/− vs. PrxII+/+ (WT) samples. These findings suggest that Prx II gene knockout may down-regulate the allograft rejection that occurs during DMSCs transplantation and improve the survival rate of DMSCs in the host. This study provides a new perspective on the clinical treatment of stem cell transplantation.

        Anti-inflammatory effect of hispidin on LPS induced macrophage inflammation through MAPK and JAK1/STAT3 signaling pathways

        Severe inflammatory reactions caused by macrophage activation can trigger a systemic immune response. In the present study, we observed the anti-inflammatory properties of hispidin on LPS induced RAW264.7 macrophage cells. Our results showed that hispidin treatment significantly reduced the production of cellular NO, IL-6 and reactive oxygen species (ROS) while has not inhibitory effect on TNF-α productions. Excitingly, hispidin treatment retains the phagocytosis ability of macrophages which enabling them to perform the function of removing foreign invaders. Signaling studies showed, hispidin treatment dramatic suppressed the LPS induced mitogen activated protein kinases (MAPK) and JAK/STAT activations. In conclusion, our findings suggest that hispidin may be a new therapeutic target for clinical treatment of macrophages-mediated inflammatory responses.

      • The XPD Lys751Gln Polymorphism has Predictive Value in Colorectal Cancer Patients Receiving Oxaliplatin-Based Chemotherapy: a Systemic Review and Meta-analysis

        Background: The predictive value of the xeroderma pigmentosum group D (XPD) Lys751Gln polymorphism regarding clinical outcomes of patients with colorectal cancer (CRC) receiving oxaliplatin-based chemotherapy has been evaluated in numerous published studies, but the results remain inconclusive. Therefore, we performed a meta-analysis to determine the precise role of the XPD Lys751Gln polymorphism in this clinical situation and optimize individual chemotherapy. Materials and Methods: A multiple search strategy was used to identify eligible studies. Pooled odds ratios (ORs), generalized odds ratio (ORG) and their 95% confidence intervals (CIs) were used to estimate the objective response, while hazard ratios (HRs) with 95%CIs were used for progression-free survival (PFS) and overall survival (OS). Results: A total of 17 studies including 2,286 patients met the inclusion criteria. Overall, the XPD 751Gln allele was associated with a non-significant reduced objective response to oxaliplatin-based chemotherapy in all patients or in the Asian and Caucasian subgroups. However, poor PFS and OS of CRC patients treated with oxaliplatin-based regimens were significantly related to the XPD 751Gln allele in the dominant model (PFS: HR=2.10, 95%CI: 1.65-2.67; OS: HR=3.18, 95%CI: 1.57-6.47). On stratified analysis by ethnicity, these relationships were more pronounced in Asians (PFS: HR=2.49, 95%CI: 1.79-3.47; OS: HR=5.25, 95%CI: 3.46-7.94) than in Caucasians (PFS: HR=1.73, 95%CI: 1.22-2.46; OS: HR=1.78, 95%CI: 1.06-2.99). Conclusions: The XPD Lys751Gln polymorphism may have prognostic value in patients with CRC undergoing oxaliplatin-based chemotherapy.

        Nutritional Effects and Antimicrobial Activity of Kefir (Grains)

        Kefir exhibits antimicrobial activity in vitro against gram-positive and gram negative bacteria, as well as some fungi. The ability of LAB to inhibit the growth of closely related bacteria is well known. This inhibition of pathogenic and spoilage microbes may be due to the production of organic acids, hydrogen peroxide, acetaldehyde, diacetyl, carbon dioxide, or bacteriocins. Lactobacilli are the major contributors to acid production and, hence, a determining factor in the flavor development in kefir. Lactic acid, proteolytic activity, and acetaldehyde are the essential flavor compounds in kefir. Both acid and bacteriocins contribute to the antimicrobial activity of kefir and kefir grains. Kefir is rich in proteins, calcium, vitamin B<sub>12</sub>, niacin, and folic acid. Many studies have investigated the benefits of consuming kefir and have shown that it is a natural probiotic, which when consumed regularly, can help relieve intestinal disorders, promote bowel movement, reduce flatulence, and improve the overall health of the digestive system. Tibetan kefir, which is different from traditional kefir, is produced in China. It has been reported to exhibit antimicrobial activity that is nearly identical to that of traditional kefir. Kefir production is considered a rapidly growing food industry in China.

      • Olive Leaf Extract Attenuates Obesity in High-Fat Diet-Fed Mice by Modulating the Expression of Molecules Involved in Adipogenesis and Thermogenesis

        <P>The present study aimed to investigate whether olive leaf extract (OLE) prevents high-fat diet (HFD)-induced obesity in mice and to explore the underlying mechanisms. Mice were randomly divided into groups that received a chow diet (CD), HFD, or 0.15% OLE-supplemented diet (OLD) for 8 weeks. OLD-fed mice showed significantly reduced body weight gain, visceral fat-pad weights, and plasma lipid levels as compared with HFD-fed mice. OLE significantly reversed the HFD-induced upregulation of WNT10b- and galanin-mediated signaling molecules and key adipogenic genes (PPAR<I><I>γ</I></I>, C/EBP<I><I>α</I></I>, CD36, FAS, and leptin) in the epididymal adipose tissue of HFD-fed mice. Furthermore, the HFD-induced downregulation of thermogenic genes involved in uncoupled respiration (SIRT1, PGC1<I><I>α</I></I>, and UCP1) and mitochondrial biogenesis (TFAM, NRF-1, and COX2) was also significantly reversed by OLE. These results suggest that OLE exerts beneficial effects against obesity by regulating the expression of genes involved in adipogenesis and thermogenesis in the visceral adipose tissue of HFD-fed mice.</P>

        中國會計制度改革現狀及展望

        This paper gives an analysis of the status quo of and existing problems in the reform of China's accounting system as a part of the nation's globalization in accounting, and points out that China, which is at the period of changing economic pattern, still has certain difficulties in meeting the principles of international financial statement. It illustrates that these barriers will gradually ebb away as the reform and opening up progresses still further, and global economic integration accelerates. China's meeting the principles of international financial statement is inevitable. The paper suggests three aspects for future improvements. Firstly, efforts should be made to train high quality accountants fit in a global environment. Secondly, a positive external environment should be provided for the globalization of accounting, which can be achieved by the perfection of legal systems and the enhancement of a market-oriented economic system. Thirdly, the international communication and cooperation in accounting should be encouraged in order that the international accounting practices can be studied and referenced, and China's globalization in accounting can be gradually realized.

        中国上市公司股权结构对内部控制的影响研究

        股权结构是公司治理体系的产权基础,管理体系中的产权通过治理机制也会影响内部控制的效率。 本文从股权结构的角度出发,研究股权结构对内部控制有效性的影响,本文将采用案例分析法,从 食品类上市公司中选取不同股权结构的企业,研究股权结构对内部控制有效性的影响机理。在剔除 其他影响内部控制有效性的影响因素后,选取了上海梅林、三元股份、莲花味精、星湖科技、金达 威、南方黑芝麻、三全食品、汤臣倍健作为分析对象,采用内部控制有效性评价体系,从股权集中 度以及股东制衡度两个方面对内部控制有效性作出评价,也对非国有控股公司与国有控股公司的内 部控制有效性作出了对比。根据案例分析结论,本文提出保持适当的股权集中度、非国有控股公司 选择适合自身发展的战略、国有控股公司可适当地引入非国有性质的股东,国有控股公司应加强对 合法合规性、报告可靠性及经营效益的重视,以加强我国国有企业内部控制有效性建设水平。 Ownership structure is the basis of property right of the corporate governance system. The property right in the management system through the governance mechanism will also affect the efficiency of internal control. From the perspective of ownership structure, this article studies the effect of ownership structure on the effectiveness of internal control. This article uses the case analysis, selecting enterprises with different ownership structures from the listed companies in the food industry, to study the influence mechanism of ownership structure on the effectiveness of internal control. After excluding other influence factors which influence the effectiveness of the internal control, It chooses Shanghai Merlin, Sanyuan shares, Lianhua Gourmet Powder, Star Lake Bioscience, Kingdomway, Nanfang Black Sesame, Sanquan Foods, and BY-HEALTH as analysis object, to evaluate the effectiveness of internal control from two aspects: ownership concentration and check-and-balance of ownership, with the evaluation system of the effectiveness of internal control. It also compares the effectiveness of internal control between non-state-owned holding companies and state-owned holding companies. According to the conclusion of case analysis, this paper suggests that companies maintain proper ownership concentration, that non-state-owned holding companies choose strategies which suit their own development, that the state-owned holding companies can introduce appropriately unless the homologous state-owned checks and balances shareholders of checks and balances relationship, that state-owned holding companies should strengthen the importance of legal compliance, the reliability of reports, and management benefit, to improve the effectiveness level of internal control of state-owned companies in our country.
