      • Bisphenol A 아만성 노출이 랫트의 수컷 생식기계에 미치는 영향

        Bisphenol A, which is widely used in the manufacture of several types of plastics including polycarbonate and epoxy resins, elicits weak estrogenic activity in in vitro and in vivo test system. the objective of this study was to evaluate the effects of low-dose bisphenol A on male reproductive organs. Male Fischer 344 rats were given bisphenol A in the drinking water at levels of ○(control),0.1,1,10,100 ppm, respectively, for 13 weeks. Body weight changes, weights of reproductive organs(testes, epididymides, ventral prostate, dorsolateral prostate, seminal vesicles), daily sprem production, epididymal sprem density, histology of reproductive organs and testicular enzyme astivities(γ-glutamyl transpeptidase, sorbitol dehydrogenase, acid phosphatase,β-glucuronidase)were not affected bt exposure to bisphenol A at any dose examined. The results of this study suggest that exposure of F344 rats to low-dose bisphenol A might be not reduce sprem density and disrupt developnebt of the male reproductive organs.

        To investigate the lethality and dose-response relationship in liver and kidney of rats at 48 hours after cadmium exposure, single doses of cadmium chloride were given to Sprague-Dawley male rats by tail-intravenous injection with several groups of doses ranging from 2 to 8 mg CdCl_2/kg for lethality and from 0.2 to 3.2 mg CdCl_2/kg for dose-response relationship. In addition, control was treated with distilled water by same method. Hepatic and renal cadmium concentrations were analyzed with I.C.P.(Inductively Coupled Plasm) Emission Spectrometry (Jobin Yvon, Model JY 24, France). Dosages over 4 mg/kg were relevant to LD_100 and LD_50 was calculated as 3.3 mg/kg at 48 hours after cadmium chloride exposure in rat. The concentration in liver and kidney were increased according to given doses, and dose-response relationship was expressed as log Cd in liver= 0.8799+0.913×log Dose (F=1116.1, r=0.9877) and log Cd in kidney= 0.2542+0.0653×log Dose (F=884.8, r=0.9845), respectively. Also, the correlation between hepatic and renal cadmium concentration was highly significant(p<0.01), and dose-response relationship was expressed as long Cd in kidney = -0.6649+1.0259×log Cd in liver (F=411.1, r=0.9676). I consider that this data will be useful for the further studying of low-dose cadmium toxicity.

        카드뮴폭로후 혈액과 요중 카드뮴의 변화양상

        The changes of cadmium(Cd) in plasma, whole blood, erythrocyte and urine in Sprangue-Dawley male rats exposed to intravenous singleinjection of 0.8 ㎎ CdCl₂/kg of body weight were investigated. Blood was taken at 0.5 to 672 hours, and 24 hour-urine was collected by using metabolic cage for the same period. The plasma level of Cd was reached to peak at 0.5 hour after injection and reduced rapidly in 1 hour. The Cd level in blood was the highest in plasma and the lowest in erythrocyte at 0.5 hour after injection. However, in one hour postinjection, the levels of Cd were higher in order of erythrocyte, whole blood and plasma, up to 4 weeks. The changes of urinary volume and creatinine were not significant between Cd-treated and saline-treated groups. However, urinary protein was slightly increased with time in Cd-treated group. Urinary Cd level was higher in Cd treated group than control. These results suggest that the measurement of Cd in erythrocyte and urine is valuable for the biological index to estimate recent Cd exposure.

        고속액체크로마토그래피-가시선/자외선 검출기를 이용한 Di(2-ethylhexyl)phthalate 검출 방법 검증과 일부 한국 성인 남성 혈액 중 DEHP 정량

        목적: 이 연구는 HPLC-UV를 이용한 생체 시료 중 저농도의 DEHP 분석법을 CDER 기준에 따라 검증하고 검증된 분석법으로 일부 한국의 혈장 중의 DEHP를 분석하여 인체 노출 정도를 뒷받침할 수 있는 기초 자료를 제시하고자 한다. 방법: 혈액 내 DEHP 정량을 위한 HPLC-UV 정량 분석법을 CDER의 검증 기준인 검량선 및 직선성과 정밀성, 정확성에 대해 시행하였다. 검증된 분석법으로 울산과 춘천 일부 지역의 근로자 30세 이상 60세 이하의 건강한 남성 255명으로부터 채취한 혈액 내 DEHP를 정량하였다. 결과: 혈액 중 DEHP 정량을 위해 사용된 HPLC-UV는 표준액 0∼150 μg/l 에서 직선성을 나타냈다(r²=0.999). 검량한계는 5.22 μg/l , 정량한계는 15.81 μg/l 이었다. 정밀성과 정확도는 2.5 μg/l DEHP 농도에서 첫째 날을 제외하고는 둘째 날과 셋째 날은 CDER 기준에 적합하였다. 50 μg/l 과 150 μg/l DEHP 농도는 모두 CDER 기준에 적합하였다. 대상자의 98 %가 10 μg/l 이하이었고, 85%는 5 μg/l 이하이었고 왼쪽으로 치우친 분포를 나타냈으며, 최고 농도는 18.9 μg/l 이었다. 대상자의 혈액의 DEHP 농도의 기하평균과 기하표준편차는 0.4±1.5 μg/l 이었다. 결론: 본 연구에 사용된 HPLC-UV를 이용한 혈액 중 DEHP 분석법은 CDER 기준에 따라 검증한 결과 정밀도, 정확도, 검출한계 및 정량한계 등 모든 항목에서 기준에 적합하였다. 일부 한국 성인 남자의 혈액 중 DEHP 농도는 0∼18.9 μg/l 이었으며 왼쪽으로 치우친 분포를 나타냈다. Objectives: This study was conducted to validate a simple, rapid and sensitive reverse-phase high-performance liquid chromatographic method with UV detector (HPLC-UV) and present the plasma level of di(2-ethylhexyl)phthalate (DEHP) in some Korean male workers. Methods: HPLC-UV for quantification of plasma DEHP was validated by the following guideline from the Center for Drug Evaluation and Research (CDER) - calibration/standard curve, precision, accuracy and recovery. Plasma DEHP from 255 healthy Korean male workers aged from 30 to 60 years was analyzed by validated HPLC-UV method. Results: The calibration curve over the range 0∼150 μg/l for the plasma DEHP standard solution showed linearity(r²=0.999). The limit of detection (LOD) and limit of quantification (LOQ) of plasma DEHP were 5.22 μg/l and 15.81 μg/l , respectively. The accuracy and precision for 2.5 μg/l of DEHP were acceptable in CDER guideline on the second and third day but not first day, and those for 50 μg/l and 150 μg/l of DEHP were acceptable on all three days(Ed-confirm this addition). The distribution of plasma DEHP level was skewed to the left and ranged from 0 to 18.9 μg/l. The plasma DEHP level was lower than 10 μg/l for 98 % of subjects and lower than 5 μg/l for 85 %. The geometric mean and standard deviation of plasma DEHP were 0.4±1.5 μg/l. Conclusions: The HPLC-UV method for quantification of plasma DEHP was acceptable by CDER guideline. The plasma DEHP of 255 Korean male workers ranged from 0 to 18.9 μg/l and the distribution was skewed to the left.

        Effects of ESG Management of Retail Companies on Consumer Values and Attitudes

        Purpose: This study aims to investigate the effect of retailer’s ESG performance on consumption value. The research analyzed the effect of consumption value that can be obtained through ESG management of retail companies on consumer attitude and repurchase intention. Research design, data, and methodology: A research model and hypothesis were established, and statistical analysis was performed on a total of 278 samples of all age groups. To secure the reliability and validity of the data, confirmatory factor analysis and discriminant validity analysis were performed, and the causal relationship of latent variables was verified through path analysis. Results: As a result of the analysis, it was found that among the characteristics of consumption value according to ESG management, social value, economic value, accessible value, and eco-friendly value had an effect on social contribution attitude. Also, the economic value was found to have an effect on the self-actualization attitude, and both the social contribution attitude and the self-actualization attitude have an effect on the purchase intention. Conclusions: This study empirically analyzed the value of ESG management in the retail industry that consumers are paying attention to recently. This will serve as a basis for revising and supplementing the retail companies’ strategy in the near future.

        Combination Therapy with a PI3K/mTOR Dual Inhibitor and Chloroquine Enhances Synergistic Apoptotic Cell Death in Epstein-Barr Virus-Infected Gastric Cancer Cells

        The phosphoinositide 3-kinase/protein kinase B/mammalian target of rapamycin (PI3K/AKT/mTOR) signaling pathway is a promising target for gastric cancer (GC) treatment; however the efficacy of PI3K/mTOR dual inhibitors in GC has not yet been maximized. Additionally, the effect of autophagy regulation by PI3K/mTOR dual inhibitors has not been clearly elucidated in GC treatment. We aimed to show that our newly developed PI3K/mTOR dual inhibitor, CMG002, when combined with an autophagy inhibitor, chloroquine (CQ), potently induces effective cancer cell death in Epstein-Barr virus (EBV)-associated gastric cancer (EBVaGC) cells, where both the PI3K/AKT/mTOR and autophagy pathways play important roles in disease pathogenesis. EBV- and mock-infected AGS and NUGC3 GC cell lines were treated with CMG002 +/- CQ. PI3K/AKT/mTOR signaling pathway mediators, cellular apoptosis and autophagy markers were confirmed by Western blot assay. Cell viability was assessed using the Cell Counting Kit-8 (CCK-8) assay. CMG002 effectively blocked the PI3K/AKT/mTOR pathway by markedly decreasing phosphorylation of AKT and its downstream mediator S6. CMG002 induced G0/G1 cell cycle arrest and enhanced apoptotic cell death in AGS and NUGC3 cells, particularly EBV-infected cells compared with mock-infected cells, as confirmed by flow cytometric analyses and TUNEL (terminal deoxynucleotidyl transferase dUTP nick end labeling) assays. The combination of CMG002 plus CQ synergistically increased apoptotic cell death in EBV-infected GC cell lines when compared with CMG002 alone (P < 0.05). Our results suggest that the new PI3K/mTOR dual inhibitor, CMG002, when used in combination with the autophagy inhibitor, CQ, provides enhanced therapeutic efficacy against EBVaGC.

      • A Case of Dapsone Hypersensitivity Syndrome

        Dapsone (4,4'-diamino-diphenyl sulfone), a potent anti-inflammatory and antiparasitic compound, is still used as a first line drug for leprosy. In Europe and the United states of America, however, it finds application mainly in the treatment of bullous dematoses and dermatitis herpetiformis. The most frequent side effects are dose-related methemoglobinemia and hemolytic anemia, and rarely, it can cause an idiosyncratic reaction, called dapsone hypersensitivity syndrome (DHS). The authors report a case of dapsone hypersensitivity syndrome occurring in a 21-year-old female diagnosed by the presentation of systemic erythematous papuloplques, hepatosplenomegaly, acute hepatitis, lymphadenopathy, and mild pericardial effusion with 3 weeks intake of dapsone for the treatement of acne that showed rapid improvement of symptoms after cessation of the drug and application of systemic steroids.

        Effects of Sophoraflavanone G, a Prenylated Flavonoid from Sophora Flavescens, on Cyclooxygenase-2 and In Vivo Inflammatory Response

        Previously, several prenylated flavonoids having a C-8 lavandulyl moiety were found to inhibit cyclooxygenase-1 (COX-1) as well as 5-lipoxygenase (5-LOX), and sophoraflavanone G was the most potent inhibitor against these eicosanoid generating enzymes among 19 prenylated flavonoids tested. In this investigation, effects of sophoraflavanone G on COX-2 induction from RAW 264.7 cells and in vivo inflammatory response were studied. Sophoraflavanone G inhibited prostaglandin $E_2{\;}(PGE_2)$ production from lipopolysaccharide (LPS)-treated RAW cells by COX-2 down-regulation at 1-50 uM. Other prenylated flavonoids including kuraridin and sanggenon D also down-regulated COX-2 induction at 10-25 uM, while kurarinone and echinoisoflavanone did not. In addition, sophoraflavanone G showed in vivo anti-inflammatory activity against mouse croton oil-induced ear edema and rat carrageenan paw edema via oral (2-250 mg/kg) or topical administration (10-250 ug/ear). Although the potencies of inhibition were far less than that of a reference drug, prednisolone, this compound showed higher anti-inflammatory activity when applied topically, suggesting a potential use for several eicosanoidrelated skin inflammation such as atopic dermatitis.

        Long Photoperiod Affects Gonadal Development in Olive Flounder Paralichthys olivaceus

        To effects of sex maturation in olive flounder by regulating long photoperiod, gonadal development and GTH mRNA expression in the pituitary were investigated. Photoperiod was treated natural photoperiod and long photoperiod (15L:9D) conditions from September 2011 to March 2012. The results showed that natural photoperiodic group showed a higher gonadosomatic index (GSI) than long photoperiodic group during the spawning season (March 2012). The histological analysis of ovarian tissue showed that natural photoperiod group of ovaries contained vitellogenic oocytes, but long photoperiod group of ovaries mainly contained perinucleolus staged oocyte and oil-drop staged oocytes. The FSH mRNA of olive flounder, under natural photoperiod group, showed a significantly higher expression but no significant difference under long photoperiod group. The $LH{\beta}$ mRNA showed a significantly higher expression only under natural photoperiod group. These results may suggest that long photoperiodic information regulates secretion of pituitary FSH and LH and maintain early growing stage of gonadal development in this species.
