Treatment with phosphodiester CpG-ODN ameliorates atopic dermatitis by enhancing TGF-β signaling

Synthetic oligodeoxynucleotides (ODNs) containing unmethylated CpG phosphorothioate (PS CpG-ODN) are known to decrease IgE synthesis in Th2 allergy responses. Nonetheless, the therapeutic role of PS CpG-ODN is limited due to cytotoxicity. Therefore, we developed a phosphodiester (PO) form of CpG-ODN (46O) with reduced toxicity but effective against allergies. In this study, we first compared the toxicity of 46O with CpG-ODNs containing a PS backbone (1826S). We also investigated the therapeutic efficacy and mechanism of 46O injected intravenously in a mouse model of ovalbumin (OVA)- induced atopic dermatitis (AD). To elucidate the mechanism of 46O underlying the inhibition of IgE production, IgE- and TGF-β-associated molecules were evaluated in CD40/IL-4- or LPS/IL-4-stimulated B cells. Our data showed that the treatment with 46O was associated with a lower hematological toxicity compared with 1826S. In addition, injection with 46O reduced erythema, epidermal thickness, and suppressed IgE and IL-4 synthesis in mice with OVA-induced AD. Additionally, 46O induced TGF-β production in LPS/IL-4-stimulated B cells via inhibition of Smad7, which suppressed IgE synthesis via interaction between Id2 and E2A. These findings suggest that enhanced TGF-β signaling is an effective treatment for IgE-mediated allergic conditions, and 46O may be safe and effective for treating allergic diseases such as AD and asthma. A polyvinylidene fluoride (PVDF)-based film speaker is successfully fabricated with enhanced bass sound by incorporating buckled nanospring carbon nanotubes (NS-CNTs) as fillers. Various concentrations up to 1–7 wt% of uniformly dispersed buckled NS-CNTs are loaded to increase the beta (β)-phase fraction, crystallinity, and dielectric constant of the speaker, and this results in the bass part enhancement of about 19 dB full scale (dBFS) at 7 wt% filler loading of the piezoelectric film speaker.

산업현장에서는 공장 자동화의 발전에 따라 많은 로봇들이 사람을 대신하고 있다. 일반적으로 산업 현장에서 사용되는 로봇은 고정밀도의 감속기를 필요로 한다. 고정밀의 제어를 위해서는 cycloid 치형 혹은 인벌류트 치형을 가진 감속기를 사용하게 된다. 이러한 감속기는 낮은 백래쉬와 높은 감속비를 가지면서 동시에 치형의 높은 강성을 필요로 한다. 본 논문에서는 새로이 고안한 직선 치형을 가진 로봇 감속기(RSR) 치형의강성을 FEM 해석을 통해 실제로 적용이 가능한지 여부를 시뮬레이션 해보았으며, 실제로도 적용이 가능함을 알 수 있었다. In industrial site, as the technology of FA developes, many robots can take the place of human labor. Generally, robots require high precision reducers which have cycloid or involute tooth profile. These reducers provide high reduction ratios with low backlash and high accuracy but need a high stiffness of tooth. In this study, to improve the high precision, the cycloid reducers which have straight line teeth profile are used. Also the FEM analysis, which consist of straight line teeth profile, was performed to simulate the stress of teeth profile.
새로운 연노출의 효과지표로서의 혈장 δ-aminolevulinic acid 측정의 유용성

To evaluate the usefulness of the measurement of plasma δ - aminolevulinic acid (ALAP) as a new effect indicator of lead exposure, the association of ALAP with blood lead, DMSA chelatable lead and bone lead with a special emphasis of genetic interaction of ALAD polymorphism was studied with 419 lead exposed workers and 85 non-lead exposed workers. Lead workers were recruited from storage battery industries, secondary smelting industries and other lead using industries and non-lead workers selected from electronic refrigerator manufacturing industry. Blood lead, DMSA chelatable lead and bone lead were chosen for parameters of lead exposure. Plasma and urinary ALA were selected for the effect parameter of lead intoxication. Information for age, smoking, drinking habit and body mass index were also obtained. Job duration for lead workers was also collected. The overall prevalence of the variant allele, ALAD type 1-2 or 2-2(ALAD2) in lead exposed workers was 10.0% (42 out of 419 lead workers) which was not differed from control workers (9.4% : 8 out of 85). The means blood lead and DMSA chelatable lead of lead workers with ALAD 1(ALAD type 1-1) were lower than those of lead workers with ALAD2, but the differences were not statistically significant. On the other hand, the mean of tibia lead of lead workers with ALADI was higher than that of lead workers with ALAD2, but the difference was not statistically significant. DMSA chelatable lead showed highest correlation with log transformed plasma ALA(LALAP)(r=0.682) and blood lead and tibia lead had r=0.650, r=0,402 correlation coefficient with LALAP respectively On the other hand. log transformed urinary ALA(LALAU) had lower correlation coefficient with all three parameters of lead exposure than LALAP. The equation of robustic simple linear regression of blood lead on LALAP was LALAP=2.0439+0.0165PbB in ALADI lead workers and LALAP=2.031+0.0121PbB in ALAD2 lead workers. ALAD genotype in the multiple regression analysis of blood lead with LALAP after adjusting for possible confounders(sex, ago, BMI, smoking and drinking status and job duration) showed significant main effect on LALAP resulting lower LALAP in ALAD2 lead workers than ALADI lead workers. On the other hand, effect modification of ALAD genotype was observed in the multiple regression analysis of tibia lead with LALAP after adjusting for possible confounders, but there was no significant effect of ALAD genotype in the multiple regression analysis of DMSA chelatable lead with LALAP after covariate adjustment. With above results, plasma ALA was found to be a useful effect parameter of lead exposure compared with urinary ALA. It was also observed that ALAD polymorphism made significant effect on the association of blood and tibia lead with ALAP. Thc lower plasma ALA in ALAD2 lead workers suggested supportive effect of ALAD2 in lead exposure.
톨루엔 폭로근로자의 요중 마뇨산과 자각증상에 관한 연구

To study the relationship between the urinary hippuric acid and subjective symptoms in toluene exposed workers, urinary hippuric acid was measured and subjective symptoms questionnarire was surveyed in 93 toluene exposed male workers and 96 non-exposed office male workers. The results are as follows: 1. Mean concentration of urinary hippuric acid, which is significantly higher in exposed group than in non-exposed group, was 1.3 g/1, 1.2(GM, GSD respectively) in exposed group and 0.6 g/1, 1.2(GM, GSD respectively) in non-exposed group. 2. Mean concentration of toluene in the air in the workplace, which is significantly higher in the department of coater than in the department of mexing, was 23.4 ppm, 6.2(GM, GSD respectively) in the department of mixing and 59.8 ppm, 3.4(GM, GSD respectively) in the department of coater. 3. Complained rates of subjective symptoms were significantly higher in exposed group than in non-exposed group in all items. 4. In exposed group, complained rate of CNS related symptoms was higher than that of irritation ralated symptoms in the first part but there was no significant difference and complained rate of irritation related symptoms in the second part was significantly higher than that of CNS related symptoms. 5. In exposed group, complained rates of subjective symptoms were not compatible with dose-response relationship by the concentration of urinary hippuric acid, duration of work, department of work, and age group retrospectively.

Recently, piezoelectric actuator has been applied in the various fields of industry such as life science, automotive, IT and telecom. The PZT actuator is used by the reverse piezoelectric effect, which generated a mechanical displacement in the electric field. Because the displacement of this actuator is 1/1000 times of PZT element thickness or height with high voltage, this actuator has to combine with a displacement amplified mechanism. In this paper, the PZT actuator has 10 times amplified displacement using hinge-lever magnification mechanism. The free load displacement of multilayer PZT is 56㎛ at input voltage of 100Vdc, and the proposed mechanism has maximum amplified displacement of 684㎛ through the experiment. The frequency response of PZT actuator having hinge-lever mechanism is measured as resonance frequency. As a result, the resonance frequency of PZT actuator is 209.7㎐ in simulation and 204㎐ in experiment.
