      • Brine Shrimp Bioassay를 이용한 해양생물의 세포독성검색

        As a part of chemical study on the bioactive metabolites from marine organisms, we have investigated cytotoxicity using brine shrimp bioassay for each solvent fractions of the marine algae(12 species), marine sponges(3 species), coelenterates(2 species), echinoderms(4 species), marine molluscs(17 species), and ascidians(2 species), respectively. As the results, chloroform extract of Stichopus japonicus (LC_50 : 274 ㎍/ml), ethyl acetate extract of Anthocidaris crassispina(LC_50 : 121 ㎍/ml), n-butanol extract of Undaria pinnatifida (LC_50 : 178 ㎍/ml), and water extract of Thais clavigera (LC_50 : 61 ㎍/ml), displayed the most significant cytotoxic activity against brine shrimp. Among the marine organisms tested, echinoderms and marine molluscs were thought to be the most active Phylums on screening of new bioactive compounds.

        Experience of Anti-Viral Therapy in Hepatitis B-Associated Membranous Nephropathy, Including Lamivudine-Resistant Strains

        <P><B>Background/Aims</B></P><P>Chronic hepatitis B infection is a common cause of secondary membranous nephropathy (MN) in endemic areas. Lamivudine treatment improves renal outcome in patients with hepatitis B virus-associated MN (HBV-MN), but prolonged use leads to the emergence of lamivudine-resistant variants. We describe our experience treating lamivudine-resistant and other strains of HBV-MN with new antiviral drugs.</P><P><B>Methods</B></P><P>Of the 89 patients biopsied and diagnosed with MN from 1996 to 2011, 10 positive for hepatitis B surface antigen were recruited for this study. We investigated the clinical courses, therapeutic responses, and prognoses of patients with HBV-MN.</P><P><B>Results</B></P><P>The incidence of HBV-MN among the original 89 patients was 11.2%. Of these patients, four were treated with supportive care and six with antiviral drugs. One of the four patients treated with supportive care had a spontaneous remission. Four of the six patients treated with antiviral drugs were given lamivudine, and the other two were given entecavir. Two of the four patients treated with lamivudine achieved complete remission with seroconversion (i.e., development of anti-hepatitis B e antigen antibodies), whereas the other two had lamivudine-resistant strains, which were detected at 22 and 23 months after lamivudine treatment, respectively. We added adefovir to the treatment regimen for one of these patients, and for the other patient we substituted clevudine for lamivudine. Both of these patients experienced complete remission, as did the two patients initially treated with entecavir, neither of whom showed resistance to the drug.</P><P><B>Conclusions</B></P><P>New nucleoside analogues, such as entecavir, adefovir, and clevudine, can be effective for treatment of HBV-MN, including lamivudine-resistant strains.</P>

      • Adiponectin receptor agonist AdipoRon decreased ceramide, and lipotoxicity, and ameliorated diabetic nephropathy

        <P><B>Abstract</B></P> <P><B>Background</B></P> <P>Adiponectin is known to take part in the regulation of energy metabolism. AdipoRon, an orally-active synthetic adiponectin agonist, binds to both adiponectin receptors (AdipoR)1/R2 and ameliorates diabetic complications. Among the lipid metabolites, the ceramide subspecies of sphingolipids have been linked to features of lipotoxicity, including inflammation, cell death, and insulin resistance. We investigated the role of AdipoRon in the prevention and development of type 2 diabetic nephropathy.</P> <P><B>Methods</B></P> <P>AdipoRon (30 mg/kg) was mixed into the standard chow diet and provided to <I>db/db</I> mice (<I>db</I> + AdipoRon, <I>n</I> = 8) and age-matched male <I>db/m</I> mice (<I>dm</I> + AdipoRon, <I>n</I> = 8) from 17 weeks of age for 4 weeks. Control <I>db/db</I> (<I>db</I> cont, <I>n</I> = 8) and <I>db/m</I> mice (<I>dm</I> cont, <I>n</I> = 8) were fed a normal diet of mouse chow.</P> <P><B>Results</B></P> <P>AdipoRon-fed <I>db/db</I> mice showed a decreased amount of albuminuria and lipid accumulation in the kidney with no significant changes in serum adiponectin, glucose, and body weight. Restoring expression of adiponectin receptor-1 and -2 in the renal cortex was observed in <I>db/db</I> mice with AdipoRon administration. Consistent up-regulation of phospho-Thr<SUP>172</SUP> AMP-dependent kinase (AMPK), peroxisome proliferative-activated receptor α (PPARα), phospho-Thr<SUP>473</SUP> Akt, phospho-Ser<SUP>79</SUP>Acetyl-CoA carboxylase (ACC), and phospho-Ser<SUP>1177</SUP> endothelial NO synthase (eNOS), and down-regulation of protein phosphatase 2A (PP2A), sterol regulatory element-binding protein-1c (SREBP-1c), and inducible nitric oxide synthase (iNOS) were associated within the same group. AdipoRon lowered cellular ceramide levels by activation of acid ceramidase, which normalized ceramide to sphingosine-1 phosphate (S1P) ratio. In glomerular endothelial cells (GECs) and podocytes, AdipoRon treatment markedly decreased palmitate-induced lipotoxicity, which ultimately ameliorated oxidative stress and apoptosis.</P> <P><B>Conclusions</B></P> <P>AdipoRon may prevent lipotoxicity in the kidney particularly in both GECs and podocytes through an improvement in lipid metabolism, as shown by the ratio of ceramide to sphingosines, and further contribute to prevent deterioration of renal function, independent of the systemic effects of adiponectin. The reduction in oxidative stress and apoptosis by AdipoRon provides protection against renal damage, thereby ameliorating endothelial dysfunction in type 2 diabetic nephropathy.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Superfluous ceramide via decrease in acid ceramidase activity causes lipotoxicity. </LI> <LI> Adiponectin and adiponectin receptors potentially increase ceramidase activity. </LI> <LI> AdipoRon ameliorated renal lipotoxicity by AdipoR1-AMPK and AdipoR2-PPARα pathway. </LI> <LI> AdipoRon is a promising tool for treatment of diabetic nephropathy in type 2 DM. </LI> </UL> </P> <P><B>Graphical Abstract</B></P> <P>[DISPLAY OMISSION]</P>

      • Successful transplant of a kidney with fibromuscular dysplasia having higher glomerular filtration rate than the contralateral kidney.

        <P>Fibromuscular dysplasia is the second-most commonly encountered anatomic abnormality in potential renal donors. Normotensive patients with medial fibroplasia and low-grade lesions have been used as renal donors. However, no studies have reported the optimal choice of a kidney for donation where the kidney with fibromuscular dysplasia had a larger volume and a higher glomerular filtration rate than the unaffected side. Herein, we report a case of renal transplant using a kidney with fibromuscular dysplasia that had higher glomerular filtration rate than the normal side. After transplant, hypertension and abnormal serum creatinine did not occur in either the donor or the recipient during 12 months' follow-up.</P>

        길 잃은 “여자들을 위한” 이야기: The Lost Girl

        This paper aims to analyse the way the narrator`s view of modern women`s social work negatively affects the consistency and wholeness of the narrative, in D. H. Lawrence`s The Lost Girl. Many critics have considered The Lost Girl as an unsuccessful novel, generally arguing that it lacks a consistency between the English and the Italian parts of the novel. Basically agreeing with them, this paper focuses on the problem of inconsistency in the characterization of Alvina, the heroine of this novel. It arises from the discrepancy between the Alvina as represented by the narrative itself and the Alvina conducted by the interfering narrator. Unlike Ursula Brangwen in The Rainbow and Women in Love who has a depth of insight and feeling, Alvina should have been let to live an ordinary living as a rather common girl of declining mercantile class. However, as soon as she actually takes a nursing position, the narrator interferes and prematurely dismisses the episode and furthermore, makes a sweeping criticism of the common people and their way of life. The narrator`s criticism of modern mechanical work or modern women`s preoccupation with `social` work is acceptable and valid to some extent. Nevertheless, the narrator`s pertinacious denial and neglection of her commonness or her search for common career makes this novel unconvincing, and unpopular as well.

        로렌스의 유령 이야기: 「국경선」과 「유쾌한 유령들」을 중심으로

        Focusing on "The Border-Line" and "Glad Ghosts", this paper aims to examine and reevaluate D. H. Lawrence`s ghost stories that have attracted relatively little critical attention. Lawrence`s ghost stories were written in the late phase of his literary career when the images of death were frequent in his short stories. Two overlapping backgrounds are behind this: the First World War was synonymous with death, and the devastated post world-war European countries were all engulfed by the tide of death; and around the same time, suffering from tuberculosis, Lawrence had to face the shadow of death hung upon his own life. Lawrence`s ghost stories were also inspired by the English ghost story tradition which achieved great popularity between 1850 and 1930. However, Lawrence radically deconstructed the fundamental hypothesis of this genre where the ambiguity of the ghost, that is, whether it really exists or it`s just a symptomatic phenomena of an unsettled and disturbed psyche, plays an pivotal role in arousing tension and suspense. Most of Lawrentian ghosts return without giving fear or suspense and with the purpose of fulfilled relationship between them and the living people who expect and even waitfor their return. Both "The Border-Line" and "Glad Ghosts" partly fall a little short of the principles of successful short story such as condensed atmosphere, economy in narrative, dramatic climax, and final reversal. However, "The Border-Line" presents a new concept of ghost, and "Glad Ghosts" goes beyond the spirit-body dichotomy and continues to search for more complex layers in the meaning of the ghost. Creating a subtle "varied tone" in the narrative, and interweaving the ghost theme with other realistic or exotic motifs like class and swapping, "Glad Ghosts" is highly probably an attractive Lawrentian ghost story.

        「표 주세요」의 "거친 로맨스"에 담긴 의미

        As often in Lawrence`s fiction, the man and woman relationship explored in "Tickets, Please," one of the war-time short stories can be more richly understood in the context of his view on the crisis of human relationship in capitalist modern age. Even if the settings obviously reflect the first world war`s impacts and the characters` love relationship really takes a form of battle in "Tickets, Please," the frame of battle between man and woman may unduly restrict the implications of this story. Rather "Tickets, Please" can be read as an exploration of a modern man and woman relationship with both dangerous recklessness and liberative vitality that the Midlands tram car also stands for. The revenge of the girl conductors against their elusive lover, John Thomas, is put into a dilemma because their "wild romance" aboard the tram car relies on their "dash and recklessness" and his very elusiveness itself as well. In some sense, the romance between Annie, one of the girl conductors and the heroin of this story and John Thomas has a liberative power that they are supposed to explore and substantiate. But John Thomas and Annie just typically perform the conventional romantic love which the narrator parodies in the fairground scene by the insertion of many "of course`s" and "after all`s." Likewise, Annie`s will to revenge for his betrayal as well as her assurance of his love is an old and ineffectual response to his liberative and reckless love. As the girls` Bacchante-like attack subtlely intrigued with physical desire for John Thomas approaches the highest peak, and when John Thomas finally chooses Annie with full malice and violently confirms his male power, Annie finds herself in a kind of cul-de-sac. "Something" that "was broken in her" is the contradictory logic of the "certain wild romance" she and the conductor girls have involved in. Presenting an analogy between the violent liberating rush of the tram car and the adventurous romance of the conductor girls, "Tickets, Please" explores a new relationship between men and women who live and love as "shipmates aboard this careering vessel of a tram-car" in modern age. Now that the liberative and reckless power of their romance also corresponds to the ambivalence of modernity, the significance of this peculiar romance represented in "Tickets, Please" cannot be overemphasized.

        『마사 퀘스트』의 곤경 : 1차 대전의 상처와 “분리 원칙”에 사로잡힌 현대적 공동체의 위기

        This paper studies Martha Quest focusing on the heroine’s desperate struggle for a new community. Martha is a sensitive and precocious English girl growing in a southern African country. With a painful sense of estrangement and frustration, she finds herself and the world only through books. To begin with, this paper defenses the heroine’s strange passivity and inertia that have frequently been criticized as a flaw in characterization since the publication of this novel, and explains how deeply it is influenced by her father’s “fatal lethargy”. He left England with a trauma from the participation in World War I and came to Africa, but he was still fettered by the old European ideologies. In a sense, Martha takes over her father’s failure in the reconciliation between self and society, as well as “the morbid strain”. But by virtue of her imaginative and intelligent power, Martha catches an impressive vision of “a noble city” where people live harmoniously with no discrimination. It may be regarded as an idealistic innocent vision of a teenage girl. Nevertheless, by intuitively reflecting that there can exist no community without a mechanism of exclusion, Martha’s vision has something more than just a sheer ideal. Of course, the accomplishment of her vision is another question not only because she lives in a split community where “the principle of separateness” dominates, but because she cannot easily overcome her own inveterate inertia. Thus Martha Quest presents the passionate vision of an ideal community and leaves it rarely accomplished or even attempted; however, this unique balance between vision and anti-vision is more convincing and persuasive in the whole context of this novel.
