Methods : The subjects were 132 (67 male and 65 female), and classified to three age groups (≤39,40∼49, and 50≤). Hood lead, manganese and aluminium were analyzed by atomic absorption spectrophotometer, and blood silicon was analyzed by direct current plasma optical omission spectrometer. Results : Blood lead levels(geometric mean, S.D) were (3.49, 1.70) ㎍/dL in male auld (3.04, 1.65) ㎍/dL in female, but the difference is not significant, and there was no significant difference between age groups. Mean blood manganese level was 0.99±0.41㎍/dL, and there was no significant difference between sex or age groups. Mean blood aluminium level was 0.59±0.35㎍/dL and there was no significant difference between sex or age groups. Mean blood silicon level was 54.41±27.64㎍/dL in male and 43.34±23.51㎍/dL in female, and the level in male was significantly higher than that in female (p〈0.05). There was significant difference between age groups, and the oldest showed the highest level in male (p〈0.05), but no significant difference between age groups in female. Conclusions : Authors hope that this study would provide basic data for determininig reference values and evaluating health effects.

      • 열병합발전용 Dual Fuel Engine의 질소산화물 배출저감에 관한 연구

        This study shows the correlation between ?? emission in the exhaust gas and various operation factors of dual fuel engine for Co-generation system. General tendency was shown that the thermal efficiency was lowered by the change of operation factors. However these were not confirmed on this experiment. Increasing T4 temperature (exhaust gas temperature at turbo-charger inlet)reduces ?? emission rate. The higher T4 temperature requires lower excess air as the excess air ratio is controlled by T4 temperature on gas mode operation. Another tendency was that ?? emission rate is reduced in case of increasing boost air temperature, quantity of pilot oil bypassing flue gas through the through the exhaust gas boiler. The diameter of the fuel injection nozzle was changed smaller than design value and the injection timing was readjusted. Thus ?? enission rate could be reduced as retarding injection timing and changing hole diameter of fuel injection nozzle, however maxium engine out-put was decreased by changing fuel nozzle on the diesel mode operation.

      • Deoxypodophyllotoxin in <i>Anthriscus sylvestris</i> alleviates fat accumulation in the liver via AMP-activated protein kinase, impeding SREBP-1c signal

        <P><B>Abstract</B></P> <P>Deoxypodophyllotoxin (DPT) is a naturally occurring flavolignan in <I>Anthriscus sylvestris</I> known as cow parsley or wild chervil, and has been reported to have inhibitory effects against several pathological processes including cancer, inflammation and infection. Here, we report the effects of DPT in the fatty liver induced by high fat diet <I>in vivo</I> as well as its regulatory mechanism related with the transcription factor for lipogenic genes such as sterol regulatory element binding protein-1c (SREBP-1c) <I>in vitro</I>. C57BL/6 mice were fed high fat diet for 10 weeks and also orally administrated with DPT for additional 4 weeks. 5 and 10 mg/kg of DPT decreased lipid accumulation in the liver induced by high fat diet, as indicated by histological parameters such as Oil Red O staining and hematoxylin & eosin as well as the contents of hepatic triglyceride and cholesterol. In hepatocytes, DPT inhibited the liver X receptor α-mediated SREBP-1c induction and expression of the lipogenic genes, including fatty acid synthase, acetyl-CoA carboxylase and stearoyl-CoA desaturase-1. Moreover, DPT induced AMP-activated protein kinase (AMPK) activation, which has been known to inhibit the expression of SREBP-1c in hepatocyte. Also this compound restored the dysregulation of AMPK and SREBP-1c induced by high fat diet in mice. In conclusion, we demonstrated that DPT significantly inhibited fatty liver by adjusting lipid metabolism coordinated with AMPK activation and SREBP-1c inhibition.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Deoxypodophyllotoxin (DPT), a chemopreventive flavolignan, inhibited diet-induced fatty liver. </LI> <LI> DPT inhibited the LXR-α-mediated sterol regulatory element binding protein (SREBP)-1c. </LI> <LI> DPT blocked the expression of <I>de novo</I> lipogenic genes including FAS and ACC. </LI> <LI> DPT activated AMPK related with SREBP-1c inhibition. </LI> <LI> DPT might be a pharmaceutical candidate for hepatic steatosis. </LI> </UL> </P>

        The Effect of Composite Pig Islet-Human Endothelial Cell Grafts on the Instant Blood-Mediated Inflammatory Reaction

        <P>Instant blood-mediated inflammatory reaction (IBMIR) causes rapid islet loss in portal vein islet transplantation. Endothelial cells are known to protect against complement-mediated lysis and activation of coagulation. We tested composite pig islet-human endothelial cell grafts as a strategy to overcome IBMIR. Porcine islets were cocultured with human endothelial cells in specially modified culture medium composed of M199 and M200 for 1-9 days. A positive control group, negative control group, and the endothelial cell-coated group were examined with an in vitro tubing loop assay using human blood. The endothelial cell-coated group was subdivided and analyzed by degree of surface coverage by endothelial cells (< or = 50% vs. > 50%) or coculture time (< 5 days vs. > or = 5 days). Platelet consumption and complement and coagulation activation were assessed by platelet count, C3a, and thrombin-antithrombin complex (TAT), respectively. After 60-min incubation in human blood, the endothelial cell-coated group showed platelet consumption inhibition and low C3a and TAT assay results compared to uncoated controls. When the endothelial cell-coated group was subdivided by degree of surface coverage, the < or = 50% coated group showed less platelet consumption and less activation of complement and coagulation compared with the positive control (uncoated) group. On analysis by coculture time, only the subgroup cocultured for < 5 days showed the same protective effect. Human endothelial cell-coated pig islets, especially the partially coated and short-term cocultured pig islet-human endothelial cell composites, reduced all components of IBMIR. If the optimal endothelial cell-islet coculture method could be identified, human endothelial cell coating of pig islets would offer new strategies to improve xenogenic islet transplantation outcomes.</P>

      • Inhibition of Autophagy Promotes Salinomycin-Induced Apoptosis via Reactive Oxygen Species-Mediated PI3K/AKT/mTOR and ERK/p38 MAPK-Dependent Signaling in Human Prostate Cancer Cells

        <P>Recently, the interplay between autophagy and apoptosis has become an important factor in chemotherapy for cancer treatment. Inhibition of autophagy may be an effective strategy to improve the treatment of chemo-resistant cancer by consistent exposure to chemotherapeutic drugs. However, no reports have clearly elucidated the underlying mechanisms. Therefore, in this study, we assessed whether salinomycin, a promising anticancer drug, induces apoptosis and elucidated potential antitumor mechanisms in chemo-resistant prostate cancer cells. Cell viability assay, Western blot, annexin V/propidium iodide assay, acridine orange (AO) staining, caspase-3 activity assay, reactive oxygen species (ROS) production, and mitochondrial membrane potential were assayed. Our data showed that salinomycin alters the sensitivity of prostate cancer cells to autophagy. Pretreatment with 3-methyladenine (3-MA), an autophagy inhibitor, enhanced the salinomycin-induced apoptosis. Notably, salinomycin decreased phosphorylated of AKT and phosphorylated mammalian target of rapamycin (mTOR) in prostate cancer cells. Pretreatment with LY294002, an autophagy and PI3K inhibitor, enhanced the salinomycin-induced apoptosis by decreasing the AKT and mTOR activities and suppressing autophagy. However, pretreatment with PD98059 and SB203580, an extracellular signal-regulated kinases (ERK), and p38 inhibitors, suppressed the salinomycin-induced autophagy by reversing the upregulation of ERK and p38. In addition, pretreatment with <I>N</I>-acetyl-<SMALL>L</SMALL>-cysteine (NAC), an antioxidant, inhibited salinomycin-induced autophagy by suppressing ROS production. Our results suggested that salinomycin induces apoptosis, which was related to ROS-mediated autophagy through regulation of the PI3K/AKT/mTOR and ERK/p38 MAPK signaling pathways.</P>

        Improved accuracy in periodontal pocket depth measurement using optical coherence tomography

        Purpose: The purpose of this study was to examine whether periodontal pocket could be satisfactorily visualized by optical coherence tomography (OCT) and to suggest quantitative methods for measuring periodontal pocket depth. Methods: We acquired OCT images of periodontal pockets in a porcine model and determined the actual axial resolution for measuring the exact periodontal pocket depth using a calibration method. Quantitative measurements of periodontal pockets were performed by real axial resolution and compared with the results from manual periodontal probing. Results: The average periodontal pocket depth measured by OCT was $3.10{\pm}0.15mm$, $4.11{\pm}0.17mm$, $5.09{\pm}0.17mm$, and $6.05{\pm}0.21mm$ for each periodontal pocket model, respectively. These values were similar to those obtained by manual periodontal probing. Conclusions: OCT was able to visualize periodontal pockets and show attachment loss. By calculating the calibration factor to determine the accurate axial resolution, quantitative standards for measuring periodontal pocket depth can be established regardless of the position of periodontal pocket in the OCT image.

      • HOG와 SVM을 사용한 지하철 실내 픽토그램 인식

        지하철 실내의 표지판은 다양한 정보들을 포함하고 있다. 그 중에서 시설물의 형상을 단순화하여 나타낸 픽토그램 사인은 국제적으로 표준화 되어 있어 검출이 용이하다고 볼 수 있다. 일반적으로 객체를 인식하는 방법은 후보영역을 설정하는 검출단계와 후보영역을 인식하는 단계로 나뉘어 진다. 본 논문에서는 후보영역 검출단계에서 단일 값을 가지는 이진화로 픽토그램 영역이 분리가 잘 안되는 문제점을 해결하기 위하여 적응적 이진화를 사용하였고 인식을 위한 특징추출로 HOG서술자를 사용하고 학습과 인식에는 SVM을 사용하였다. 실험 결과를 통하여 HOG서술자로 픽토그램 사인을 인식하는 것이 적합한 것인지 확인한다.

      • 에지 이미지를 사용한 컷 검출의 구현

        최근에는 텍스트정보 보다 동영상정보를 다루는 일이 많아졌고 그에 따라 동영상 데이터의 분할, 색인, 검색 등을 위해 장면 전환 검출이 필요하게 되었다. 장면 전환 검출 기술은 비디오 데이터의 장면 변화가 발생하는 경계를 검출하는 기술이다. 본 논문에서는 에지 이미지를 이용한 장면전환 검출과 이를 위한 임계값 설정, 그리고 결과에서 중복된 이미지와 오 검출 된 이미지를 줄여줄 수 있는 구현에 대하여 실험결과를 바탕으로 설명한다.
