      • 三天池의 環境과 微生物의 多樣性

        경상북도 경산시에 위치하고 있는 삼천지에 대한 환경요인 및 미생물상에 관하여 조사하였다. 삼천지 수계의 환경요인 분석결과, 그 평균값은 기온이 25.8℃, pH6.77, 경도 117.94mg/ℓ, 부유물질 4.5mg/ℓ, 용존산소량 3.3mg/ℓ, 생물학적 산소요구량 6.6mg/ℓ, 화학적 산쇼요구량 7.2㎎/ℓ, 염소이온 17.56㎎/ℓ, 황산이온 7.14㎎/ℓ로 나타났고, 중금속류는 Mn과 Zn만이 소량 검출되었으나 전반적인 오염도가 높아 수질환경 기준 IV급수에 해당하였다. 총 세균수는 3.32-8.85×?? No. inds/㎖의 분포를 나타냈고, 식물플랑크톤의 종구성은 규조류 33종, 녹조류 26종, 남조류 8종, 유글레나 13종, 와편모조류 6종 등 86종이 확인되었다. 규조류중 띠돌말과에 속하는 Fragilaria capucina, Synedra acus, S. familiaris 가 우점종이였다. Environmental factors and microflora of Samchun-impoundment in Kyongsan, Kyongpook were studied. Average data of environmental factors analysis are as followings : air temperature 25.8℃, pH 6.77, total hardness 117.94mg/ℓ, SS 4.5mg/ℓ, DO 3.3mg/ℓ, BOD 6.6mg/ℓ, COD 7.2mg/ℓ, ?? 17.56mg/ℓ, ?? 7.14mg/ℓ, Mn 0.05mg/ℓ, Zn 0.04mg/ℓ. Total bacteria in 4 site were 3.32×?? No. inds/㎖. Distribution and species of phytoplankton and zooplankton were 86 species including 33 species of bacillariophyceae, 26 of chlorophyta, 8 of cyanophyceae, 13 of euglenaceae, 6 of dinophyceae. Fragilaria capucina, Synedra acus, S. familiaris were dominant species.

      • Assessing the safety of an Ephedrae Herba aqueous extract in rats: A repeat dose toxicity study

        <P><B>Abstract</B></P> <P>Ephedrae Herba (EH) has been used in Asian traditional herbal medicine to cure bronchial asthma, cold, flu, chills, fever, headache, nasal congestion, and cough. In this study, we evaluated the subchronic toxicity of an Ephedrae Herba aqueous extract (EHAE) in male and female F344 rats. The EHAE was administered orally daily at doses of 0, 125, 250, 500, and 1000 mg/kg bw/day for 13 weeks. Toxicological assessment was performed to determine mortality, clinical signs, and changes in body weight, food consumption, ophthalmological, urinary, hematological, and serum biochemical parameters, macroscopic and microscopic evaluations, and organ weights. We found that oral administration of EHAE to F344 rats for 13 weeks resulted in histopathological changes in the kidneys and salivary glands. In the kidneys, increased incidence and severity of tubular basophilia were observed in females administered 1000 mg/kg bw/day of the extract. In the salivary glands, acinar cell hypertrophy was observed in males administered 500 mg/kg bw/day and in both sexes administered 1000 mg/kg bw/day of the extract. All test article-treated groups of males and females administered ≥250 mg/kg bw/day showed increased absolute and relative salivary gland weights. Therefore, the NOAEL (No Observed Adverse Effect Level) was determined as 125 mg/kg bw/day for both sexes of rats under the present experimental conditions.</P> <P><B>Highlights</B></P> <P> <UL> <LI> There is little data in the literature on the toxic effects of Ephedrae Herba. </LI> <LI> Three males and two females in the highest dose group (1000 mg/kg bw/day) were found dead at weeks 1 and 9. </LI> <LI> In macroscopic observation, test article-related changes were observed in the kidneys and salivary glands. </LI> <LI> The no-observed-adverse-effect level was determined as 125 mg/kg bw bw/day for both sexes. </LI> </UL> </P>

        Evaluation of Genotoxicity and 28-day Oral Dose Toxicity on Freeze-dried Powder of Tenebrio molitor Larvae (Yellow Mealworm)

        The larval form of Tenebrio molitor (T. molitor) has been eaten in many countries and provides benefits as a new food source of protein for humans. However, no information exists regarding its safety for humans. The objective of the present study was to evaluate the genotoxicity and repeated dose oral toxicity of the freeze-dried powder of T. molitor larvae. The genotoxic potential was evaluated by a standard battery testing: bacterial reverse mutation test, in vitro chromosome aberration test, and in vivo micronucleus test. To assess the repeated dose toxicity, the powder was administered once daily by oral gavage to Sprague-Dawley (SD) rats at dose levels of 0, 300, 1000 and 3000 mg/kg/day for 28 days. The parameters which were applied to the study were mortality, clinical signs, body and organ weights, food consumption, ophthalmology, urinalysis, hematology, serum chemistry, gross findings and histopathologic examination. The freezedried powder of T. molitor larvae was not mutagenic or clastogenic based on results of in vitro and in vivo genotoxicity assays. Furthermore, no treatment-related changes or findings were observed in any parameters in rats after 28 days oral administration. In conclusion, the freeze-dried powder of T. molitor larvae was considered to be non-genotoxic and the NOAEL (No Observed Adverse Effect Level) was determined to be 3000 mg/kg/day in both sexes of SD rats under our experimental conditions.

        PCR-based identification of Pseudomonas fluorescens in diseased olive flounder, Paralichthys olivaceus, in Jeju Island, South Korea

        Pseudomonas is currently causing increasing mortality in farmed olive flounder in Jeju Island. It was previously reported that P. anguilliseptica is the pathogen causing the mortality. It is not known whether other sub-species are involved or not. In this study, P. fluorescens was identified from diseased olive flounder by a PCR-based diagnosis. Based on genomic sequencing and BLAST analysis, 5 out of 6 samples were closer with P. fluorescens than P. anguilliseptica. Our finding suggests that P. fluorescens may be the dominant species causing the disease in farmed olive flounder in Jeju Island, South Korea.

        Involvement of sirtuin 1 in airway inflammation and hyperresponsiveness of allergic airway disease

        <P><B>Background</B></P><P>Bronchial asthma is a chronic inflammatory disorder of the airways characterized by increased expression of multiple inflammatory genes. Acetylation of histones by histone acetyltransferases is associated with increased gene transcription, whereas hypoacetylation induced by histone deacetylases is associated with suppression of gene expression. Sirtuin 1 (SIRT1) is a member of the silent information regulator 2 family that belongs to class III histone deacetylase.</P><P><B>Objective</B></P><P>This study aimed to investigate the role of SIRT1 and the related molecular mechanisms in the pathogenesis of allergic airway disease.</P><P><B>Methods</B></P><P>By using a murine model of ovalbumin (OVA)–induced allergic airway disease and murine tracheal epithelial cells, this study investigated the involvement of SIRT1 and its signaling networks in allergic airway inflammation and hyperresponsiveness.</P><P><B>Results</B></P><P>In this study with mice after inhalation of OVA, the increased levels of SIRT1, hypoxia-inducible factor 1α (HIF-1α), and vascular endothelial growth factor protein in the lungs after OVA inhalation were decreased substantially by the administration of a SIRT1 inhibitor, sirtinol. We also showed that the administration of sirtinol reduced significantly the increased numbers of inflammatory cells of the airways; airway hyperresponsiveness; increased levels of IL-4, IL-5, and IL-13; and increased vascular permeability in the lungs after OVA inhalation. In addition, we have found that inhibition of SIRT1 reduced OVA-induced upregulation of HIF-1α in airway epithelial cells.</P><P><B>Conclusions</B></P><P>These results indicate that inhibition of SIRT1 might attenuate antigen-induced airway inflammation and hyperresponsiveness through the modulation of vascular endothelial growth factor expression mediated by HIF-1α in mice.</P>

        미용학과 헤어 전공자의 전공만족도가 직무만족도에 미치는 영향

        We aimed to investigate the effects on job satisfaction have major impacts satisfaction of Dept. of Beauty Arts Hair majors study. Targets who major in Dept. of Beauty Arts Hair were set working in Seoul and Gyeonggi-do area. Ninety-nine data which collected through questionnaire was analyzed by SPSS 20.0 and applied analytical methods were factor analysis, reliability analysis, frequency analysis, multiple regression analysis. The research results showed that the influence on job satisfaction of Dept. of Beauty Arts Hair majors' major satisfaction has significant influence. Among the job satisfaction factor, all factors affected major satisfaction except salary satisfaction factor. It can be seen that, I'm sure and satisfy on my major, regarding my job as important, enjoying task, thinking as getting recognition in workplace and satisfying on position, working environment, service environment. It was found that Dept. of Beauty Arts Hair major's major satisfaction had affect on job satisfaction. We can see the importance of choosing major that satisfaction in workplace grows as major choice satisfying. As salary and major satisfaction hasn't significant influence, it seems to be that people do not choose major having salary in mind. Therefore, based on the results of studies, if you are trying to create an fostering environment in which guaranteed future system rather than salary and in workplace, it will be able to help encourage employee's work satisfaction.
