Immunomodulatory effects of ethanol extract of germinated ice plant (Mesembryanthemum crystallinum)

The purpose of this study was to investigate the immunomodulatory activity of ice plant (Mesembryanthemum crystallinum) extract (IPE) in vitro and in vivo. Raji (a human B cell line) and Jurkat (a human T cell line) cells were treated with various doses of IPE and cell proliferation was measured by WST assay. Results showed that IPE promoted the proliferation of both Raji and Jurkat cells in a dosedependent manner. IPE also enhanced IL-6 and TNF-α production in macrophages in the presence of lipopolysaccharide (LPS), although IPE alone did not induce cytokine production. Moreover, IPE treatment upregulated iNOS gene expression in macrophages in a time- and dose-dependent manner and led to the production of nitric oxide in macrophages in the presence of IFNγ. In vivo studies revealed that oral administration of IPE for 2 weeks increased the differentiation of CD4+, CD8+, and CD19+ cells in splenocytes. These findings suggested that IPE has immunomodulatory effects and could be developed as an immunomodulatory supplement.

Distal renal tubular acidosis is a condition characterized by an inability of the distal nephron to acidify urine, causing hyperchloremic metabolic acidosis. Distal renal tubular acidosis is classified as proton secretory defect, permeability defect and voltage defect based on its pathophysiology. In the former two, serum level of potassium decreases due to increased excretion of potassium. But in the latter (voltage defect), hyperkalemia is characteristic by impaired the generation of an optimal electrical gradient for hydrogen ion and potassium secretion. We experienced a case of Sjogren`s syndrome associated with both voltage defect distal renal tubular acidosis and nephrogenic diabetes insipidus. The patient was a 58- year-old woman who complained of general weakness, nausea and xerostomia. Laboratory analysis showed metabolic acidosis with alkaline urine and hyperkalemia. Anti-nuclear antibody and anti-ds DNA antibody were positive. She presented with polyuria, low urine osmolarity and inadequate response to DDAVP. The response to Shirmer test was decreased. Salivary scintigraphy showed decrease of uptake in the parotid and submandibular salivary glands. We believe this is the first case report in which Sjogren`s syndrome is associated with both voltage defect distal renal tubular acidosis and nephrogenic diabetes insipidus.(Korean J Med 60:485-489, 2001)

<P>The crystal structure of β-Ca<SUB>3</SUB>SiO<SUB>4</SUB>Cl<SUB>2</SUB> was determined by the <I>ab initio</I> structure determination method based on the synchrotron powder XRD data for the first time, and the luminescence properties of a Eu<SUP>2+</SUP>-doped β-Ca<SUB>3</SUB>SiO<SUB>4</SUB>Cl<SUB>2</SUB> phosphor were characterized. β-Ca<SUB>3</SUB>SiO<SUB>4</SUB>Cl<SUB>2</SUB> was found to be monoclinic (space group <I>P</I>2<SUB>1</SUB>/<I>c</I>) with the lattice parameters, <I>a</I> = 5.91234(1) Å, <I>b</I> = 10.20128(1) Å, <I>c</I> = 10.98866(1) Å, β = 90.3423(1)°. This structure can be considered as an intergrowth structure built up from alternating stacks of two layered sublattices, <SUB>∞</SUB><SUP>2</SUP>[Ca<SUB>2</SUB>SiO<SUB>4</SUB>] and <SUB>∞</SUB><SUP>2</SUP>[CaCl<SUB>2</SUB>], along the [100] direction. In this structure, the Ca atoms occupy three crystallographically distinct sites: Ca1O<SUB>4</SUB>Cl<SUB>3</SUB>, Ca2O<SUB>5</SUB>Cl<SUB>2</SUB>, and Ca3O<SUB>3</SUB>Cl<SUB>4</SUB>. The photoluminescence of the Eu<SUP>2+</SUP>-doped Ca<SUB>3</SUB>SiO<SUB>4</SUB>Cl<SUB>2</SUB> phosphor excited at 450 nm blue light shows the 150 nm wide-band emission peaked at 635 nm with about 70% quantum efficiency. The photoluminescence properties, such as centroid shifts, crystal-field splitting, and Stokes shifts, were correlated with the crystal structure through the calculation of shared-electron populations reflecting the bond covalency between Ca and O/Cl. Induction of Apoptosis by Yomogin in Human Promyelocytic Leukemic HL-60 Cells

Yomogin is an active compound isolated from Artemisia princep, a traditional Oriental medicinal herb, which has been shown to inhibit tumor cell proliferation. In this study, we investigated the effects of yomogin on the cytotoxicity, induction of apoptosis, and putative pathways of its actions in human promyelocytic leukemia cells. Yomogin-treated HL-60 cells displayed several features of apoptosis, including DNA fragmentation, formation of DNA ladders in agarose gel electrophoresis, and externalization of annexin-V targeted phosphatidylserine residues. We observed that yomogin caused activation of caspase-8, caspase-9, and caspase-3. A general caspase inhibitor (z-VAD-fmk), caspase-8 inhibitor (z-IETD-fmk) and caspase-3 inhibitor (z-DEVD-fmk), almost completely suppressed the yomogin-induced DNA fragmentation. We further demonstrated that yomogin induced Bid cleavage, mitochondrial translocation of Bax from the cytosol, and cytochrome c release from mitochondria in a caspase-8-dependent manner. Taken together, our data indicate that yomogin is a potent inducer of apoptosis and facilitates its activity via caspase-8 activation, Bid cleavage, Bax translocation to mitochondria, and subsequent release of cytochrome c into the cytoplasm, providing a potential mechanism for the anticancer activity of yomogin.
커피전문점의 브랜드 가치수준과 인구통계학적특성에 따른 서비스품질 및 성과에 의한 시장세분화에 관한 연구:

본 연구는 커피전문점의 브랜드가치수준과 인구통계학적 특성에 따라서 지각하는 서비스품질수준과 지각된 가치, 고객만족, 재구매 의도에 대해 살펴보고 시장 세분화에 대한 시사점을 제시하고자 하였다. 본 연구는 대구지역에 커피전문점 방문경험이 있는 18세이상의 고객을 대상으로 2016년 11월 22일부터 11월 29일 까지 7일 동안 400부의 설문지를 배포하여 불성실한 설문지를 제외한 315부를 최종분석에 사용하였다. 최종분석은 SPSS 분석패키지를 사용하였다. 분석결과, 저가치브랜드는 남성고객시장, 고졸이하의 시장과 대학원졸업자의 시장, 10대 고객을 고려한 시장, 고등학교와 대학교 시장을 주 타겟으로 진입하는 것을 고려해야한다. 또한, 고가치브랜드는 여성고객시장, 2년제, 4년제대학교 졸업자들이 많이 분포되어 있는 시장, 20대를 고려한 시장, 전문직 직장인들이 많이 근무하는 시장(오피스)시장의 진입을 고려해야할 것이다. This study aimed to examine coffee shop users’ perceived value and service quality of coffee shops, satisfaction and repurchase intention according to brand value level and demographic characteristics and suggest implications for coffee shop market segmentation. To this end, 400 questionnaires were distributed during November 22-29, 2016, to consumers 18-years-old or older who have visited coffee shops in Daegu. After excluding unreliable responses, a total of 315 questionnaires were finally analyzed. Analysis results indicated that low value brands should consider entering markets targeting male customers, customers with high school or lessor educational background and those with graduate school or higher education, markets considering teenage consumers and markets with high schools and universities. Study results also indicated that high value brands should consider entering markets for female consumers, markets with graduates of two-year colleges or four-year universities, markets considering consumers in their twenties and office markets with many professional workers.

Novelty detection (ND) is an effective technique that can be used to determine whether a future observation is normal or not. In the present study we propose a novelty detection algorithm that can handle a situation where the distributions of target (normal) observations are inhomogeneous. A simulation study and a real case with the TFT-LCD process demonstrated the effectiveness and usefulness of the proposed algorithm.

<P><B>Abstract</B></P> <P> <I>Morchella esculenta</I> (Morchellaceae), one of the most widely appreciated wild edible mushrooms, has demonstrated antitumor activity both <I>in vivo</I> and <I>in vitro</I>. However, the molecular basis underlying its antitumor activity has not yet been elucidated. In this study, we examined the biological effects of the MeOH extract of <I>M. esculenta</I> fruiting bodies on four human lung adenocarcinoma cell lines. Bioactivity-guided fractionation of the MeOH extract followed by chemical investigation of its most cytotoxic hexane-soluble fraction led to the isolation of eight compounds (<B>1</B> −<B>8</B>), including three fatty acids and five sterols. This is the first report of the isolation of compounds <B>1</B> and <B>3</B>–<B>7</B> from <I>M. esculenta</I>. Among the eight isolated compounds tested, compounds <B>1</B>, <B>3,</B> and <B>5</B> exhibited the most potent cytotoxicity to human lung cancer cell lines, with IC<SUB>50</SUB> values ranging from 156.9 to 278 M; this activity was mediated by induction of apoptosis.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Morel mushroom (<I>Morchella esculenta</I>) is a wild edible mushroom. </LI> <LI> Bioactivity-guided isolation of the extract led to the isolation of eight compounds. </LI> <LI> Compounds <B>1</B> and <B>3</B>–<B>7</B> were isolated for the first time from <I>M. esculenta</I>. </LI> <LI> Compounds <B>1</B>, <B>3</B>, and <B>5</B> exhibited potent cytotoxicity to human lung cancer cells. </LI> <LI> The activity of compounds <B>1</B>, <B>3</B>, and <B>5</B> was mediated by the induction of apoptosis. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>
