In this study, epoxy/anhydride mixing ratio for the highly silica filled compounds with chromium (III) octoate catalyst was investigated at a low curing temperature (71°C for 40 hr) by evaluating the compressive strength with the weight ratio ranges from 0.3/1.0 to 1.0/1.0 of epoxy part (Part A)/anhydride part (Part B). In case of epoxy/anhydride compounds used surface unmodified silica by coupling agent, these compounds need excess anhydride unlike the weight ratio in the conventional epoxy/anhydride compounds. In curing behavior, the epoxy/anhydride compounds containing chromium (III) octoate showed high conversions at 71°C for 40 hr, even if a dipropylene glycol (DPG) was not used as a polymerization initiator. Also, DPG leads to a poor epoxy network structure. In conclusion, the appropriate weight ratio of Part A/Part B of highly silica filled epoxy/anhydride compounds with chromium (III) octoate catalyst is 0.5/1.0 and the maximum amounts of silica is 1470 phr of epoxy resin.
Testing the Efficacy of COVID-19 Vaccine and Therapeutic Candidates Using Vero 76 Cell Line

Background COVID-19 is a respiratory infectious disease that first emerged in Wuhan, China in December 2019 and has spread worldwide. It is necessary to develop preventive vaccines and therapeutics for overcoming COVID-19. For this purpose, , in vitro efficacy testing of the applicants is essential in the early stages of development. We want to share our experience of in vitro efficacy testing Methods using SARS-CoV-2 through this presentation. Methods All tests using SARS-CoV-2 were performed within the Biosafely Level 3 (BL3) facility of Masan National Tuberculosis Hospital (MNTH). And the protocols were approved by the Institutional Biosafety Committee (IBC) of MNTH prior to the study. Vaccine candidate neutralizing Serums were obtained by collecting blood from animals immunized with vaccine candidates. In 6 well plates, the Vero 76 cells were pre-culture to sub-confluence. The serum dilution and the virus was mixed in a 1:1 volume and neutralized. The serumvirus mixture was added to the cells for infection and the uninfected virus was removed. Add media containing 1% agarose, and solidify completely. After culturing for 3 days, fix with formaldehyde, remove agarose, and stain with crystal violet. The neutralizing effect was calculated by counting the plaques and comparing them with the control. Therapeutic candidates Infect SARS-CoV-2 to Vero 76 cells with an MOI 0.01. Add the test substance prepared for each concentration and Remdesivir, a positive control, and incubate. The culture medium and cell lysate are collected, the therapeutic effect is confirmed by comparing Results with the control group by Methods such as plaque assay, qPCR and WB. Results We established our optimized in vitro protocol for testing the efficacy of vaccines and therapeutic candidates against COVID-19 quantitatively. Conclusion The established Methods would be useful in confirming the efficacy of COVID-19 vaccine candidates and therapeutic candidates in the preclinical stage.

Background: Outcomes of postoperative radiotherapy(PORT) for squamous cell carcinoma of the tonsil were analyzed focusing on management and control of the neck.Methods: Medical records of 380 pathologically-confirmed tonsil cancer patients, treated with surgery and PORT from February 1993 to January 2011 at 15 institutions, were reviewed. Neck metastasis was absent(pN0) in 53(13.9%) and present(pN+) in 327(86.1%), of which 279(85.3%), 2(0.6%), and 46(14.1%) were ipsilateral, contralateral, and bilateral, respectively. Results: Neck irradiation(NI) was done in 346(91.1%) patients, while data was unavailable in 34(8.9%). After a median follow-up of 53 months, neck recurrence occurred in 28(7.4%) patients. Among the 53 pN0 patients, NI was ipsilateral-only, bilateral, and unknown in 25(47.2%), 21(39.6%), and 7(13.2%). One patient suffered from contralateral neck failure after an ipsilateral-only NI. For the 279 ipsilateral pN+ patients, NI was ipsilateral-only, bilateral, and unknown in 67(24.0%), 193(69.2%), and 19(6.8%). Neck recurrence occurred in 18(6.5%): 11(3.9%), 6(2.2%) and 1(0.4%) failed in the ipsilateral-only, contralateral-only, and bilateral neck. The 2 contralateral pN+ patients received bilateral NI and remained free of regional recurrence. NI data was available for 38(82.6%) of the 46 bilateral pN+ patients and all received bilateral NI. Three(6.5%), 2(4.3%) and 2(4.3%) patients recurred in the ipsilateral, contralateral, and bilateral neck. Conclusions: Because contralateral neck failure is rare in pN0 patients, elective NI may safely be confi ned to the ipsilateral neck-only. For pN+ patients, however, the risk of contralateral neck failure highly varies (3 to 10%). Thus clinicopathologic factors should be considered to justify the omission of contralateral NI for such heterogenous groups.

Background: Tuberculosis is spreading from person to person through the air. When a person with infectious tuberculosis disease coughs or sneezes, tiny particles containing Mycobacterium tuberculosis are expelled into the air and remain suspended in the air for several hours. Therefore, it is important to control the transmission of M. tuberculosis through air. This study was conducted to determine the inactivation efficacy of the plasma generator against the M. tuberculosis. Methods: The attenuated M. tuberculosis H37Ra inoculated on the solid medium were placed in hospital wards and BL3 laboratory, and a plasma generator (Model TB-500, Shinyoung Airtech, Seongnam-si, Korea) was operated by time interval. The growth of M. tuberculosis was determined by plasma exposure time. Results: When M. tuberculosis was exposed to non-thermal plasma for 3, 6, 9, and 24 hours, the inhibition effects were 45-75%, 69-86%, 93-100%, and 100%, respectively. Thus, it was found that the inhibition effect of plasma generators against M. tuberculosis were proportional to the plasma exposure time. Conclusion: The non-thermal atmospheric pressure plasma generator may be applied as a promising technique for the prevention of M. tuberculosis infection in healthcare and other public facilities, if it is operated more than 9 hours. *This article has been accepted for publication on Korean J Healthc Assoc Infect Control Prev.
β-ketoacyl Carrier Protein Synthase Inhibitors Analogs as Potential Antituberculotics

Tuberculosis is a significant global health threat, with one-third of the world’s population infected with its causative agent Mycobacterium tuberculosis. Fatty acid biosynthesis is one of the relatively targets in antibacterial drug discovery and most bacterial fatty acid biosynthesis enzymes are essential for viability make this a very attractive antimicrobial drug target. The enzyme β-Ketoacyl-acyl carrier protein synthase (ACPS), contributing in mycolic acids biosynthesis, has been established as promising target of novel antimycobacterial drugs. The ACPS inhibitor analogs have been investigated against Mycobacterium tuberculosis grown either in broth medium or inside macrophages. Our compounds displayed a diversity of action by acting either on extracellular Mycobacterium tuberculosis bacterial growth only, or both intracellularly on infected macrophages as well as extracellularly on bacterial growth with very low toxicity towards host macrophages. Among the eighteen potential ACPS inhibitor analogs identified, YM-12 exhibited the best extracellular antitubercular activity (MIC50 4 ug/ml). In a macrophage model of infection, YM-12 dramatically decreased the number of intracellular Mycobacterium tuberculosis present at 5 days after infection at concentraction of 1.25 ug/ml and 2.5 ug/ml. These results might provide insights into the development of antituberculotics against Mycobacterium. Hence, the mechanism of action of the antituberculous drug on ACPS inhibitors has further investigated.
Temporal changes of Korean cicada in relation to meteorological factors

In every summer, cicadas emerge and become numerically and ecologically dominant in Korean penninsula. Especially, cicada emergence is affected by the environmental factors. In order to evaluate the effect of environmental factors in cicada species, we analyzed the temporal changes in cicada exuviae based on meteorological and non-meteorological factors such as artificial light intensity and habitat characters on urban park area surrounded by residential houses. Combined multivariate analyses with a cluster analysis and a principal component analysis (PCA) were conducted. Samples were classified into 3 different cluster based on differences of meteorological factors such as temperature and humidity. Moreover, Random Forest model (RF) showed a high predictability of daily mean temperature on species peak abundance. These results provide a strong evidence that meteorological factors have significant effects on cicada emergences. Regarding the non-meteorological factors, we found no relationship in cicada emergence.

The computational science and engineering field, which combines basic science and computing technology, has emerged as a third scientific methodology, following theories and experiments. This study aimed to identify factors and relationships that affect the continued use of the computational science and engineering (CSE) platform for its successful operation, utilization, and diffusion. To that end, the quality factors of the platform were derived by combining the information system success model and the technology acceptance model. These factors affected user satisfaction and intention to reuse through users’ perceived usefulness and perceived ease of use of the platform. An empirical analysis was conducted through a questionnaire survey of 373 users of the EDISON platform, a representative CSE platform in Korea. The results revealed that all quality factors have a positive influence on perceived usefulness and perceived ease of use. Specifically, information quality has a significant influence on perceived ease of use, and system quality has a significant influence on perceived usefulness. Perceived ease of use has a greater impact on user satisfaction than perceived usefulness, and satisfaction affects intention to reuse. The results can contribute to the development of CSE platforms and the development strategy to expand the number of users.

<P>Low in vivo transduction efficiency and safety concerns have been hurdles for effective hematopoietic stem cell (HSC) gene therapy. Here, we investigate whether the safety and efficiency of retroviral gene transfer into HSCs can be improved by using human allogeneic umbilical cord blood (UCB)-derived supplements instead of fetal bovine serum (FBS). When CD34(+) cells were cultured ex vivo in UCB-derived serum (CBS) or plasma (CBP), comparable or higher maintenance of HSCs was observed than in FBS and serum-free substitution medium (SFM) as assessed by the frequency of positive engraftment and the level of engraftment in NOD/SCID mice after transplantation of cultured cells. CBS and CBP also exhibited higher level stabilization of retroviral particles than SFM during in vitro culture of retrovirus pseudotyped with gibbon ape leukemia virus or vesicular stomatitis virus glycoprotein. Retroviral gene transfer into CD34(+) cells performed with CBS or CBP resulted in increased gene transfer into CD34(+) cells and increased transduction of reconstituted bone marrow cells compared to transfers with SFM or FBS. The increased transduction of bone marrow cells was associated with a larger number of transduced progenitors in the recipient mice. Significant oligoclonality in the transduced progenitors, as determined by ligation-mediated polymerase chain reaction, suggested efficient retroviral targeting of multiple HSCs in the CBS- or CBP-supplemented media. Combined, our results show that allogeneic UCB-derived serum or plasma is a safe and easily accessible serum supplement that can support efficient retroviral gene transfer into HSCs for the clinical-grade manipulation of HSCs.</P>
